        Alpha-Amylase Immobilization on Epoxy Containing Thiol-Ene Photocurable Materials

        Thiol-ene polymerization is a versatile tool for several applications. Here we report the preparation of epoxide groups containing thiol-ene photocurable polymeric support and the covalent immobilization of α-amylase onto these polymeric materials. The morphology of the polymeric support was characterized by scanning electron microscopy (SEM), and energy dispersive spectroscopy (EDS) coupled with SEM was used to explore the chemical composition. The polymeric support and the immobilization of the enzyme were characterized by FTIR analysis. SEM-EDS and FTIR results showed that the enzyme was successfully covalently attached to the polymeric support. The immobilization efficiency and enzyme activity of α-amylase were examined at various pH (5.0-8.0) and temperature (30-80oC) values. The storage stability and reusability of immobilized α-amylase were investigated. The immobilization yield was 276 ± 1.6 mg per gram of polymeric support. Enzyme assays demonstrated that the immobilized enzyme exhibited better thermostability than the free one. The storage stability and reusability were improved by the immobilization on this enzyme support. Free enzyme lost its activity completely within 15 days. On the other hand, the immobilized enzyme retained 86.7% of its activity after 30 days. These results confirm that α-amylase was successfully immobilized and gained a more stable character compared with the free one.

        Experimental Cyclic Behavior of Precast Hybrid Beam-Column Connections with Welded Components

        Post-earthquake observations revealed that seismic performance of beam-column connections in precast concrete structures affect the overall response extensively. Seismic design of precast reinforced concrete structures requires improved beam-column connections to transfer reversed load effects between structural elements. In Turkey, hybrid beam-column connections with welded components have been applied extensively in precast concrete industry for decades. Beam bottom longitudinal rebars are welded to beam end plates while top longitudinal rebars are placed to designated gaps in joint panels before casting of topping concrete in this type of connections. The paper presents the major findings of an experimental test programme including one monolithic and five precast hybrid half scale specimens representing interior beam-column connections of a moment frame of high ductility level. The required welding area between beam bottom longitudinal rebars and beam-end plates were calculated based on welding coefficients considered as a test parameter. It is observed that the maximum strain developed in the beam bottom flexural reinforcement plays an important role in the overall behavior of the connections. Two additional specimens which include unbonded lengths on the longitudinal rebars to reduce that strain demands were also tested. Strength, stiffness and energy dissipation characteristics of test specimens were investigated with respect to test variables. Seismic performances of test specimens were evaluated by obtaining damage indices.

        Experimental work on seismic behavior of various types of masonry infilled RC frames

        Reinforced concrete frame structures with masonry infill walls constitute the significant portion of the building stock in Turkey. Therefore it is very important to understand the behavior of masonry infill frame structures under earthquake loads. This study presents an experimental work performed on reinforced concrete (RC) frames with different types of masonry infills, namely standard and locked bricks. Earthquake effects are induced on the RC frames by quasi-static tests. Results obtained from different frames are compared with each other through various stiffness, strength, and energy related parameters. It is shown that locked bricks may prove useful in decreasing the problems related to horizontal and vertical irregularities defined in building codes. Moreover tests show that locked brick infills maintain their integrity up to very high drift levels, showing that they may have a potential in reducing injuries and fatalities related to falling hazards during severe ground shakings.
