MICROCIRCULATION OF THE LIVER AND ITS RELATION TO LIVER DISORDERS

Microcirculition in the liver and gastrointestional mucosa is closely associated with the cellular function, and hence, its disturbance relates intimately with the pathogenesis and healing of the liver diseases and the mucosal lesions. Understanding of organ microcirculation is also important for elucidating the mechanism of progression of tumors and action of drugs. Hepatic blood flow, regulated by systemic neuronal and humoral factors, is modified by prostaglandins, platelet-activating factor, and other endogeneous chemical mediators in the liver and GI tract. Endothelium-derived factor, endothelin, that produces vasoconstriction is found to have a role that regulates microcirculation in the liver. These regulators of hepatic and GI-mucosal microcirculation are discussed in terms of data abtained by in vivo microscopy and organ reflectance spectrophotometry in the liver in vivo and hemoglobin-free perfused liver.

This paper describes an investigation of driver’s behavior assessment while driving on downgrades with different gradients and effect of road safety countermeasures using a coloured pavement or a light-emitting delineator. The driving simulator used in the experiments consists of 300-degree fields of view screen, a hexapod motion platform, a sound system with spatial placed eight speakers, and so on. The target was two kinds of downgrades after curved and more gradual downgrade section: one is the existent downgrade with 8% gradient and with one traffic lane in each direction, and another has 7% gradient with two lanes that will be constructed for improvement of the existing downgrade. The existent downgrade has a temporary road structure and we investigated the effective countermeasures of the coloured pavement or a light-emitting delineator to reduce driving speed on the temporary downgrade. The simulator precisely represented these road structures using CAD data of the real and planned roads. Two downgrade sections were connected each other in one experimental route in order to compare the driving between on different downgrades and the driving with and without the countermeasures. The results of participated 31 drivers suggest that there are almost no differences in road safety between the existent and improved downgrades in terms of objective and subjective assessments. The evaluation results of the effect of the countermeasures imply the necessity of the interface design that affords drivers easy recognition of onset location of the steep downgrade.

The design and current status of EarthCARE/CPR are described in this report. Basic design of CPR will be confirmed in this year, and engineering model development and testing will be done this year and the next year. In parallel with the development activity of CPR, algorithm development activity is in progress. The data from CPR is expected to contribute to reveal the detailed information of clouds and to the studies on global warming. It is also expected the continuation of cloud observation from CloudSat data that was launched in 2006[4].
16S rRNA Genes PCR-RFLP Analysis for Rapid Identification of Oral Anaerobic Gram-Positive Bacilli

Oral anaerobic gram-positive bacilli are frequently isolated from oral infectious sites including carious, endodontic and periodontal lesions. Identification of these bacteria by conventional methods are labor-intensive and time-consuming because of poor growth and non-reactivity in most of the biochemical tests. In this study, in order to develop the rapid identification method for these bacteria, restriction fragment length polymorphism analysis of PCR-amplified 16S ribosomal RNA genes〔16S rRNA genes PCR-RFLP〕was applied to anaerobic gram-positive bacilli such as Eubacterium, Mogibacterium and Pseudoramibacter, and the obtained findings were compared with the results of biochemical identification methods. 16S rRNA gene sequences were amplified from isolated genomic DNA samples by the PCR with universal primers. The PCR products were then purified and characterized by single digestion with restriction endonuclease HpaII, and this allowed discrimination among the respective reference strains〔type strains of ATCC〕. In addition, 55 strains were assigned to one of the reference species on the basis of their restriction profiles by digestion with HpaII, and these results were corresponded to those of biochemical identification methods. Furthermore, the 16S rRNA gene sequences were accessed from the GenBank and the RDP data base, and the restriction profiles expected are in accordance with the obtained RFLP patterns in this study. therefore, 16S rRNA genes PCR-RFLP is a rapid and reliable identification method for oral gram-positive bacilli including Eubacterium, Mogibacterium and Pseudoramibacter.

Restriction fragment length polymorphism (RFLP) analysis of polymerase chain reaction〔PCR〕-amplified 16S ribosomal RNA genes〔16S rRNA genes PCR-RFLP〕using MnlI has been shown to be a useful tool in defferentiating oral Actinomyces species. In this study, the method was applied to Actinomyces isolates from human dental plaque〔n=77〕, and the findings were compared with those obtained by biochemical identification methods. As a result, 46 strains out of 55 A. naeslundii genospecies 1 strains were mis-identified as A. israelii〔n=35〕 and A. meyeri〔n=11〕; and 2 strains out of 12 A. naeslundii genospecies 2 strains were mis-identified as A. israelii by biochemical identification methods. Therefore, one should pay attention to the mis-identification of a. naeslundii species by conventional biochemical tests.
Unrestrained Sensors Using Piezoelectric Elements for Bed-Leaving Prediction

This paper presents a sensor system that predicts behavior patterns that occur when a patient leaves a bed. We originally developed plate-shaped sensors using piezoelectric elements. Existing sensors such as clip sensors and mat sensors require that patients be restrained. The features of our sensors are that they require no power supply or patient restraint for privacy problems. Moreover, we developed machine-learning algorithms to predict behavior patterns without setting thresholds. We evaluated our system for three subjects at an experimental environment constructed in reference to a clinical site. The mean recognition accuracy was 78.6% for seven behavior patterns. Especially, the recognition accuracies of lateral sitting and terminal sitting were each 94.4%. We consider that these capabilities are useful for bed-leaving prediction in practical use.
