        Heterologous expression of cyanobacterial PCS confers augmented arsenic and cadmium stress tolerance and higher artemisinin in Artemisia annua hairy roots

        The present study provides the frst report of heterologous expression of phytochelatin synthase from Anabaena PCC 7120 (anaPCS) into the hairy roots of Artemisia annua. Transformed hairy roots of A. annua expressing anaPCS gene showed better tolerance to heavy metals, viz., arsenic (As) and cadmium (Cd) owing to 143 and 191% more As- and Cd-accumulation, respectively, as compared to normal roots with a bioconcentration factor (BCF) of 9.7 and 21.1 for As and Cd, respectively. Under As and Cd stresses, transformed hairy roots possessed signifcantly higher amounts of phytochelatins and thiols probably due to the presence of both AaPCS (Artemisia annua PCS) and anaPCS. In addition, artemisinin synthesis was also induced in transformed hairy roots under heavy metals stresses. In-silico analysis revealed the presence of conserved motifs in both AaPCS and anaPCS sequences as well as structural modelling of PCS functional domain was conducted. Interaction of AaPCS and anaPCS proteins with CdCl2 and sodium arsenate gene ontology analysis gave insights to anaPCS functioning in transformed hairy roots of A. annua. The study provides transformed hairy roots of A. annua as an efcient tool for efective phytoremediation with added advantages of artemisinin extraction from hairy roots used for phytoremediation.         Genotypic influence of a-deletions on the phenotype of Indian sickle cell anemia patients

        Background :Some reports have shown that co-inheritance of a-thalassemia and sickle cell disease improves hematological parameters and results in a relatively mild clinical picture for patients; however, the exact molecular basis and clinical significance of the interaction between a-thalassemia and sickle cell disease in India has not yet been described. There is little agreement on the clinical effects of a-thalassemia on the phenotype of sickle cell disease. Methods :Complete blood count and red cell indices were measured by an automated cell analyzer. Quantitative assessment of hemoglobin variants HbF, HbA, HbA2, and HbS was performed by high performance liquid chromatography (HPLC). DNA extraction was performed using the phenol-chloroform method, and molecular study for common a-deletions was done by gap-PCR. Results :Out of 60 sickle cell anemia patients, the a-thalassemia genotype was found in 18 patients. Three patients had the triplicated a-genotype (Anti a-3.7 kb), and the remaining patients did not have a-deletions. This study indicates that patients with co-existing a-thalassemia and sickle cell disease had a mild phenotype, significantly improved hematological parameters, and fewer blood transfusions than the patients with sickle cell anemia without co-existing a-deletions. Conclusion :Co-existence of a-thalassemia and sickle cell anemia has significant effects on the phenotype of Indian sickle cell patients.

        Computer-Aided Genetic Algorithm Based Multi-Objective Optimization of Laser Trepan Drilling

        The laser trepan drilling (LTD) has proven to produce better quality holes in advanced materials as compared with laser percussion drilling (LPD). But due to thermal nature of LTD process, it is rarely possible to completely remove the undesirable effects such as recast layer, heat affected zone and micro cracks. In order to improve the hole quality, these effects are required to be minimized. This research paper presents a computer-aided genetic algorithm-based multi-objective optimization (CGAMO) methodology for simultaneous optimization of multiple quality characteristics. The optimization results of the software CGAMO has been tested and validated by the published literature. Further, CGAMO has been used to simultaneously optimize the recast layer thickness (RLT) at entrance and exit in LTD of nickel based superalloy sheet. The predicted results show minimization of 99.82% and 85.06% in RLT at entrance and exit, respectively. The effect of significant process parameters on RLT has also been discussed.

        Influence of thermally induced, dehydroxylated nanoclay on polymer nanocomposites

        This work reports a novel approach towards a chemical-free treatment of nanoclay through extensive thermalexposure. Dehydroxylation at high temperature was utilized to enhance the influence of nanoclay on the propertiesof polymer. The effect of this treatment of nanoclay, on the polymer properties, with reference to Polypropylene (PP)has been investigated. The FTIR spectra revealed the successful removal of water from the intergallery spacing of thenanoclay. The maintained structural configuration of the clay was confirmed using WAXD pattern. The uniform dispersionand exfoliation of thermally treated clay layers inside the polymer matrix was confirmed through enhancedmechanical properties. Improved crystallization properties, thermal stability and flame retardant characteristic werealso noticed in the nanocomposites reinforced with thermally dehydroxylated clay. This study revealed that the dehydroxylationapproach of modification of nanoclay may provide much enhanced properties of polymer, without involvementof any chemical for modification.

      • DETERMINANTS OF WILLINGNESS TO PAY FOR TRADITIONAL BUNDLES

        The present work aims to extend Meyer, Shankar, & Berry (2017)’s work on antecedents of consumer willingness to pay (WTP). This research paper investigates the role of antecedents such as autonomy, quality variability, complementarity, overall quality and bundle quality, for the traditional bundles (i.e., bundles consisting of either goods or services). The paper uses the choice-based conjoint methodology to measure consumer’s WTP for the traditional bundles in an experimental vignette set-up. The study uses four different types of traditional bundles as consumer stimuli to analyze the factors mentioned above. The results indicate that autonomy, overall bundle quality, and quality variability were significant in influencing the consumer’s WTP. Consumer WTP has been widely used as a factor to derive the optimal prices of the bundles in the literature. For a retailer selling a traditional bundle, it is imperative to have an understanding of consumer WTP and also make an attempt to increase it by offering high customer value. By knowing the factors determining the antecedents of consumer WTP, one can reap higher benefits monetarily as well as in the form of increased customer satisfaction. The paper also challenges the view that it is difficult to influence the consumer’s WTP. It advances the understanding of the factors which influence the consumer WTP for the widely used traditional bundles.

      • MARKETING RESEARCH FOR MANAGERS: REVISITED

        The present paper is an addition to the relevance debate in marketing literature. Research is said to be ‘relevant,’ if it leads to change, alteration or validation of how managers think, talk or act. However, the majority of the literature on relevance debate talks about the decline in relevance, yet no study has scientifically measured it. The present study uses content analysis to analyze the trends in different types of relevance across three different eras of marketing eras (i.e., commoditization, generalization and post-debate era). We also conducted a second study to check the relative importance of different types of relevance to the managers using Analytic Hierarchy Process (AHP). The results reveal a U-shaped curve of the relevance across these different eras. They also reveal the influence of the relevance debate on the research conducted by academicians. Out of the eight types of managerial relevance identified, “Forecasts” was ranked the highest, followed by “Rhetoric devices” and “Uncovering causal relationships.” Finally, the study also presents a view for an academician to shape his/her research concerning the current needs of the industry.

        MECHANICAL OXIDATION OF MULTI-WALLED CARBON NANOTUBES AS A REPLACEMENT OF ACID OXIDATION METHOD

        In the present study, a candid approach using mechanical oxidation, for modifcation of multiwalled carbon nanotubes (CNTs) has been utilized, and its advantages over conventional acid oxidation methods have been established. Higher concentration of acidic groups was found in case of mechanically oxidized CNTs (i.e., McCNT) as compared to acid oxidized CNT (i.e., ACNT). Raman spectrum exhibited greater degree of transformation of sp2 carbon into sp3 carbon. X-ray diffraction (XRD) analysis reveale maintained crystalline organization in McCNT. The major finding of the study was that, the mechanical oxidation method can be used easily to replace the conventional acid treatment method. This is because in the mechanical oxidation crystalline organization of CNTs can be maintained, relatively higher number of acid groups can be incorporated, no loss to the mass of CNTs is allowed etc.

        Study of UV aging on the performance characteristics of vegetable oil and palm oil derived isocyanate based polyurethane

        The impact of UV aging on the characteristic properties of synthesized polyurethane (PUs) from castor oil and transesterified castor oil with palm oil based isocyanate and 1,4 butanediol was investigated at different exposure time of 0, 250, 500, 750 and 1,000 h, respectively. The aging properties of the PU films were analyzed through differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), Scanning electron microscopy (SEM) and Tensile tests. The PU films derived from castor oil showed noticeable changes in chemical structure after 250 h of UV exposure owing to chain scission of the urethane group. A considerable decrease in the thermal and tensile properties of the developed PU films was observed due to the plasticization phenomenon. SEM micrographs revealed morphological changes through the appearance of cracks and blisters composed of primary amines on the surface of exposed PU films. However, PUs derived from transesterified castor oil with a higher hydroxyl value exhibited improved UV resistance characteristics due to higher crosslinking and degrade only after 750 h of exposure. Thus, the synthesized green PUs based on transesterified castor oil with palm oil based isocyanate compositions can be candidate materials for developing UV resistant coating material.

        LEADERSHIP IN STREET-LEVEL BUREAUCRACY: AN EXPLORATORY STUDY OF SUPERVISORWORKER INTERACTIONS IN EMERGENCY MEDICAL SERVICES

        Street-level bureaucrats operate in a world relatively free of supervision, exercising discretionary abilities often without the presence of formal authority figures or managers. Although wide latitude in decision making is a norm of frontline work, leaders may occasionally be present to supervise service provision. This exploratory research employs narrative inquiry to examine the interactions of street-level personnel and formal authority figures during service provision in emergency medical services. Results indicate that occasions for compliance and disregard for managerial directives are manifest. Compliant behavior was evident when patient clinical needs were relatively clear and the effects of the directives were reasonably consistent with the paramedic s preconceived notions of appropriate action. Deviation from managerial directives was apparent when patient s positive outcomes were dependent on ignoring orders. Contributions to theory and practice, as well as avenues for future research are discussed.

        Generalized anxiety and sleep quality among health care professionals during the COVID-19 pandemic: a cross-sectional study from a tertiary healthcare institution in Eastern India

        Objectives: With the emergence of the coronavirus disease 2019 (COVID-19) pandemic, healthcare professionals (HCPs) have experienced high levels of stress and anxiety because of the high risk of infection for themselves and their families. This has led to acute sleep problems for HCP. This study was designed to assess the anxiety and sleep quality of HCPs during the COVID-19 pandemic. Methods: This cross-sectional study analyzed 370 HCPs employed at All India Institute of Medical Sciences Patna over 3 months, using the standard Generalized Anxiety Disorder 7-item scale (GAD-7) for suspected GAD and the Pittsburgh Sleep Quality Index for sleep quality. Results were tabulated and multivariable binomial logistic regression analysis was done to determine the predictors of poor sleep. Significance was attributed to p < 0.05. Results: Of the 370 HCPs screened, 52 (14.1%; 95% confidence interval [CI], 10.8%–18.1%) were found to have GAD and 195 (52.7%; 95% CI, 47.5%–57.9%) were found to be poor sleepers. The presence of any addictive habit (adjusted odds ratio [AOR], 1.833; 95% CI, 1.12–2.8), unprotected contact with COVID-19 cases (AOR, 1.902; 95% CI, 1.1–3.3), and the presence of GAD (AOR, 5.57; 95% CI, 2.5–12.4) were found to be predictors of poor sleep quality among HCPs. Conclusion: A significant proportion of HCPs were found to have suspected GAD and were poor sleepers. This highlights the need for measures to confront this problem.
