The slaughter process for cattle will inevitably transfer some bacteria onto the carcasses. The goal of food safety programs is to minimize and effectively remove this contamination. This study was attempted by the Verif EYETM machine-vision technology that might be useful for reducing microbial indicator counts and could reduce the contamination chance of E coli O157:H7 and Salmonella spp on beef carcasses. For the evaluation of the effectiveness of the Verif EYE TM technology, 80 samples were examined by the inspection device over 15 days. On an examination of FDS- positive samples compared to negative controls from the same carcasses, aerobic plate counts were bigger than the negative control samples(5.26 vs 4.60log). Enterobacteriaceae counts were greater on the positive samples than the corresponding negative control samples(2.07 vs 1.17log). There was a consistent correlation between samples detected by the Verif EYETM system with detectable counts. For example, 100% of positive samples had detectable APC and 91.2% of positive samples had detectable TCC. Therefore, if areas detected as positive for contamination by the Verif EYETM system were removed from the carcasses, significant sources of microbial contamination will be reduced for objective compliance with HACCP. This results suggest that the use of Verif EYETM machine-vision technology might be useful for reducing microbial indicator counts(APC, TCC) and could help reduce the risk of presence of E coli O157:H7 and Salmonella spp on Beef carcasses.

<P>Embryonic stem cells (ESCs) have attracted great interest in the fields of tissue engineering, regenerative medicine, and organogenesis for their pluripotency and ability to self-renew. ESC aggregation, which produces an embryoid body (EB), has been widely utilized as a trigger of in vitro directed differentiation. In this paper, we propose a novel method for constructing large numbers of deep concave wells in PDMS microfluidic chips using the meniscus induced by the surface tension of a liquid PDMS prepolymer, and applied this chip for the mass production of uniform sized EBs. To investigate if the microenvironment in the deep concave well is suitable for ES cells, the oxygen diffusion to the deep concave well was analyzed by CFD simulation. Murine EBs were successfully formed in the deep concave wells without loss of cells and laborious careful intervention to refresh culture media. The size of the EBs was uniform, and retrieving of EBs was done just by flipping over the chip. All the processes including EB formation and harvest are easy and safe to cells, and their viability after completion of all processes was over 95%. The basic properties of the EBs were generated and their capacity to differentiate into 3 germ layers was investigated by analyzing the gene expression profile. The harvested EBs were found to differentiate into cardiac cells and neurons, and neurofilaments formed branches of elongated extensions more than 1.0 mm in length.</P>

<P><I>Pseudomonas aeruginosa</I>, a Gram-negative bacterium of clinical significance, produces elastase as a predominant exoprotease. Here, we screened a library of chemical compounds currently used for human medication and identified diethylene triamine penta-acetic acid (DTPA, pentetic acid) as an agent that suppresses the production of elastase. Elastase activity found in the prototype <I>P. aeruginosa</I> strain PAO1 was significantly decreased when grown with a concentration as low as 20 μM DTPA. Supplementation with Zn2+ or Mn2+ ions restored the suppressive effect of DTPA, suggesting that the DTPA-mediated decrease in elastase activity is associated with ion-chelating activity. In DTPA-treated PAO1 cells, transcription of the elastase-encoding <I>lasB</I> gene and levels of the <I>Pseudomonas</I> quinolone signal (PQS), a molecule that mediates <I>P. aeruginosa</I> quorum sensing (QS), were significantly downregulated, reflecting the potential involvement of the PQS QS system in DTPA-mediated elastase suppression. Biofilm formation was also decreased by DTPA treatment. When A549 alveolar type II-like adenocarcinoma cells were infected with PAO1 cells in the presence of DTPA, A549 cell viability was substantially increased. Furthermore, the intranasal delivery of DTPA to PAO1-infected mice alleviated the pathogenic effects of PAO1 cells in the animals. Together, our results revealed a novel function for a known molecule that may help treat <I>P. aeruginosa</I> airway infection.</P>

We performed a survey for flavivirus infection and distribution of Aedes albopictus that known as Zika and Dengue virus vector using black–light trap and BG-sentinel trap around urban area in Korea. Mosquitoes were collected in 27 cities during March to November (twice a month) year 2016. Total numbers of mosquitoes collected 102,102 including 19 species 8 genera during collecting period. Total 21,467 Ae. albopictus was collected that 20,961(24.3%) by BG-sentinel trap and 506 (3.2%) by Black-light trap in urban area. Trap index(trap/night) of Ae. albopictus was showed highest in Hamyang (TI:992.3) and lowest in Taebaek (TI:0.3) there was only collected by Black-light trap. A total of 894 pools from all collecting Ae. albopictus were performed a Flavivirus detection. Flavivirus was not detected during study period. This study may provide basic information for surveillance of imported diseases (include Zika virus) and vectors in Korea.

<P>Precise control of media delivery to cells in microfluidic systems in a simple and efficient manner is a challenge for a number of cell-based applications. Conventional syringe pumps can deliver culture media into microfluidic devices at precisely controlled flow rates, but they are bulky and require a power source. On the other hand, passive microflow-generating systems cannot maintain continuous, controllable and long-term delivery of media. We have developed an on-chip microflow control technology that combines flow rate control and passive, long-term delivery of media to microwell tissue culture chambers. Here, a passive flow is initiated using the siphon effect and a yarn flow resistor is used to regulate the flow rate in the microchannel. Using the yarn flow resistor, the medium flow rate into the microfluidic cell culture system is made adjustable to a few hundred microliters per hour. To evaluate the effects of controlled flow on microfluidic cell culture properties (feasibility test), we measured the cell alignment and cytoskeletal arrangement of endothelial cells cultured in a microwell array inside the microfluidic channel.</P>

<P>Curcumin [1,7-bis(4-hydroxy-3-methoxyphenyl)-1,6-heptadiene-3,5-dione] induces heme oxygenase-1 (HO-1) expression via activation of the nuclear factor-erythroid-2-related factor 2 (Nrf2), whereas tetrahydrocurcumin [1,7-bis(4-hydroxy-3-methoxyphenyl)-3,5-heptanedione], one of curcumin <I>in vivo</I> metabolites, has no effect on HO-1 expression and Nrf2 activation. The aim of this study was to investigate whether dimethoxycurcumin [1,7-bis(4,3-dimethoxyphenyl)-1,6-heptadiene-3,5-dione], a synthetic curcumin analogue with higher metabolic stability over curcumin, could induce HO-1 expression to the same extent as curcumin in RAW264.7 macrophages. Dimethoxycurcumin and curcumin, but not tetrahydrocurcumin, induced HO-1 expression and Nrf2 nuclear translocation, suggesting that the unsaturated nature of the diarylheptanoid chain of the compounds are crucial for HO-1 expression and Nrf2 activation. Blockage of Nrf2 synthesis by small interfering RNA abolished HO-1 expression by dimethoxycurcumin, indicating that dimethoxycurcumin may induce HO-1 expression via Nrf2 activation. In comparison, dimethoxycurcumin and curcumin had about the same effect on HO-1 expression, suggesting that dimethoxycurcumin retains the HO-1-inducing activity of its parent compound curcumin in RAW264.7 macrophages.</P>

Sigumjang prepared from fermented barley bran is a traditional fermented food found only in the Gyeongsang-do area of South Korea. There have been no studies reported to date despite the potential bioactivities of sigumjang. In this study, the anti-obesity activities of sigumjang extracts (SEs) during 3T3-L1 differentiation into adipocytes were investigated. SEs inhibited adipocyte differentiation by suppressing the CCAAT/enhancer binding $protein-{\beta}$ and sterol regulatory element binding protein-1c expression in the early stage of differentiation, followed by the suppression of the peroxisome proliferator-activated $receptor-{\gamma}$, CCAAT/enhancer binding $protein-{\alpha}$, and adiponectin. These changes in adipogenic markers induced inhibition of lipogenesis via down-regulation of mainly fatty acid synthase, acetyl-CoA carboxylase, fatty acid binding protein 4, and perilipin. These results were more significant in the extract of sigumjang fermented with isolated Bacillus amyloliquefaciens MFST compared to naturally fermented sigumjang group. SEs can be considered as a useful material for developing food with health benefits and anti-obesity properties.

Oxidative stress is one of common cause of fatty changes in the liver. Antioxidant capacity was confirmed in various vegetables including black radish (Raphanus sativus L. var niger). Fermentation of vegetables using Lactobacillus plantarum has been known to generate bioactive components. This study was conducted to determine if fermented black radish (FBR) ameliorates oxidative liver injury induced by CCl4 in rats. To accomplish this, FBR (250 and 500 mg/ kg) was orally administered to rats for 7 consecutive days, single CCl₄ (1.5 mL/kg) treatment or no treatment orally. Serum chemistry at 24 hours after CCl₄ injury showed that FBR (500 mg/kg) significantly reduced the level of both alanine aminotransferase and aspartate aminotransferase in CCl₄ exposed rats. Moreover, FBR treatment significantly increased radical-scavenging effects in livers with the reduction of lipid peroxidation in CCl₄ exposed rats. Histopathologic findings including Kupffer cell activation in the liver of each group matched those of serum chemistry. Collectively, black radish, through fermentation, exerts hepatoprotective capacity in CCl₄ induced liver injury in rats through anti-oxidation.
