        Effect of Garcinia indica Powder on Metabolic Parameters and Body Composition in Genetically Mutant WNIN/GR-Ob Rats

        Obesity is associated with metabolic diseases, including insulin resistance, type 2 diabetes and dyslipidemia. Antiobesity drugs are available but have side effects. Hydroxy citric acid (HCA) with ATP-citrate lyase enzyme-inhibiting activity has been identified as a safe potential supplement for weight management and as an antiobesity agent. In the present study, we aim to test the antiobesity potential of the fruit rind powder of G. indica (a plant rich in HCA) in genetically obese rats. Forty-five-day-old Male WNIN/GR-Ob rats were divided equally into four groups with each group having six rats. Group 1 was fed with a standard powder diet (SPD), whereas Groups 2, 3, and 4 were fed with SPD containing 1%, 3%, and 5%, respectively, of G. indica powder for 12 weeks. Food intake, body composition, oral glucose tolerance test, plasma insulin, alanine aminotransferase (ALT), aspartate aminotransferase (AST), lipid profile, hepatic glycogen, ATP-citrate lyase, and Glucose-6-Phosphate dehydrogenase (G6PDH) were measured. Histological analysis of vital organs and Immunohistochemistry (IHC) analysis was carried out in liver sections for citrate lyase score. G. indica significantly decreased food intake, body weight, body fat %, hepatic and circulating triglycerides, cholesterol, and liver steatosis. In addition, G6PDH and ATP-citrate lyase enzyme activities were decreased along with an increase in liver glycogen. The IHC scores of citrate lyase were lower in treated groups. The results indicate that G. indica exerts favorable effects on obesity with a possible mechanism of anorectic effects, suppressed citrate lyase enzyme activity, and improved insulin sensitivity due to the modulation of carbohydrate metabolism by the phytochemicals and secondary metabolites.

        Study of microstructural and mechanical properties of Aluminium LM14 hybridized with reinforcement of Nanoparticles of Titanium Diboride and Alumina

        The reinforcing impact of TiB2 nanoparticles (TiB2np) and alumina nanoparticles (Al2O3np) in an aluminum (LM14) alloybase matrix composite was studied. Stir casting was used to create the hybrid aluminum matrix-based nanocomposites(AMNC) specimens. When nanoparticles are mixed into a matrix, the resulting material is stronger than LM14 aluminumalloy. The results of the EDAX and SEM studies clearly illustrate the distribution of reinforcing particle sizes. Compressionand density tests on AMNC specimens are contrasted with those on LM14 aluminum alloy tests. The increase in theirmechanical characteristics is influenced by the proportion of reinforcements in the matrix phase. Nanocomposites based onan aluminum matrix outperform the metal in terms of compressive strength, density, and abrasion resistance.

        Congestion Management in Deregulated Power System by Optimal Choice and Allocation of FACTS Controllers Using Multi-Objective Genetic Algorithm

        Congestion management is one of the technical challenges in power system deregulation. This paper presents single objective and multi-objective optimization approaches for optimal choice, location and size of Static Var Compensators (SVC) and Thyristor Controlled Series Capacitors (TCSC) in deregulated power system to improve branch loading (minimize congestion), improve voltage stability and reduce line losses. Though FACTS controllers offer many advantages, their installation cost is very high. Hence Independent System Operator (ISO) has to locate them optimally to satisfy a desired objective. This paper presents optimal location of FACTS controllers considering branch loading (BL), voltage stability (VS) and loss minimization (LM) as objectives at once using GA. It is observed that the locations that are most favorable with respect to one objective are not suitable locations with respect to other two objectives. Later these competing objectives are optimized simultaneously considering two and three objectives at a time using multi-objective Strength Pareto Evolutionary Algorithms (SPEA). The developed algorithms are tested on IEEE 30 bus system. Various cases like i) uniform line loading ii) line outage iii) bilateral and multilateral transactions between source and sink nodes have been considered to create congestion in the system. The developed algorithms show effective locations for all the cases considered for both single and multiobjective optimization studies.

        Wind has been a rapidly growing renewable power source for the last twenty years. Since wind behavior is chaotic in nature, its forecasting is not easy. At the same time, developing an accurate forecasting method is essential when wind farms are integrated into the power grid. In fact, wind speed forecasting tools can solve issues related to grid stability and reserve allocation. In this paper 30 hours ahead wind speed profile forecast is proposed using Adaptive Wavelet Neural Network (AWNN). The implemented AWNN uses a Mexican hat mother Wavelet, and Morlet Mother Wavelet for seven, eight and nine levels decompositions. For wind speed forecasting, the time series data on wind speed has been gathered from the National Renewable Energy Laboratory (NREL) website. In this work, hourly averaged 10-min wind speed data sets for the year 2004 in the Midwest ISO region (site number 7263) is taken for analysis. Data sets are normalized in the range of [-1, 1] to improve the training performance of forecasting models. Total 8760 samples were taken for this forecasting analysis. After the forecasting phase, statistical parameters are calculated to evaluate system accuracy, comparing different configurations.

      • TP53 Codon 72 Polymorphism and Risk of Acute Leukemia

        TP53 is the mostly commonly mutated gene in many cancers and the P53 tumor suppressor protein is involved in multiple cellular processes, including transcription, DNA repair, genomic stability, senescence, cell cycle control and apoptosis. A common single nucleotide polymorphism located within the proline rich region of TP53 gene at codon 72 in exon 4 encodes either proline or arginine. TP53 Arg 72 is more active than TP53 Pro 72 in inducing apoptosis. The aim of this study was to understand the association of the 72 codon polymorphism with acute leukemia development and prognosis. A total of 288 acute leukemia cases comprising 147 acute lymphocytic leukemia (ALL) and 141 acute myeloid leukemia (AML), as well as 245 controls were recruited for analysis of the TP53 72 polymorphism using PCR-RFLP method. Significant association of homozygous arginine genotype with AML was observed (${\chi}^2$- 133.53; df-2, p < 0.001. When data were analyzed with respect to clinical variables, elevation in mean WBC, blast %, LDH levels and slight reduction in DFS in ALL cases with the arginine genotype was observed. In contrast, AML patients with Pro/Pro had elevated WBC, Blast%, LDH levels with slightly reduced DFS. Our study indicates that Arg/Arg genotype might confer increased risk to development of acute myeloid leukemia.

      • Deletion of GSTM1 and T1 Genes as a Risk Factor for Development of Acute Leukemia

        The glutathione S-transferases (GSTs) are a family of enzymes involved in the detoxification of a wide range of chemicals, including important environmental carcinogens, as well as chemotherapeutic agents. In the present study 294 acute leukemia cases, comprising 152 of acute lymphocytic leukemia (ALL) and 142 of acute myeloid leukemia, and 251 control samples were analyzed for GSTM1 and GSTT1 polymorphisms through multiplex PCR methods. Significantly increased frequencies of GSTM1 null genotype (M0), GSTT1 null genotype (T0) and GST double null genotype (T0M0) were observed in the both ALL and AML cases as compared to controls. When data were analyzed with respect to clinical variables, increased mean levels of WBC, Blast %, LDH and significant reduction in DFS were observed in both ALL and AML cases with T0 genotype. In conclusion, absence of both GST M & GST T might confer increased risk of developing ALL or AML. The absence of GST enzyme might lead to oxidative stress and subsequent DNA damage resulting in genomic instability, a hallmark of acute leukemia. The GST enzyme deficiency might also exert impact on clinical prognosis leading to poorer DFS. Hence GST genotyping can be made mandatory in management of acute leukemia so that more aggressive therapy such as allogenic stem cell transplantation may be planned in the case of patients with a null genotype.

      • Role of the MDM2 Promoter Polymorphism (-309T>G) in Acute Myeloid Leukemia Development

        Background: The human homologue of the mouse double minute 2 (MDM2) gene is a negative regulator of Tp53. MDM2-309T>G a functional promoter polymorphism was found to be associated with overexpression thereby attenuation of Tp53 stress response and increased cancer susceptibility. We have planned to evaluate the possible role of MDM2-309T>G polymorphism with risk and response to chemotherapy in AML. Materials and Methods: A total of 223 de novo AML cases and 304 age and sex matched healthy controls were genotyped for the MDM2-309T>G polymorphism through the tetra-primer amplification refractory mutation system (ARMS)-PCR method. In order to assess the functional relationship of -309T>G SNP with MDM2 expression level, we quantified MDM2 mRNA in 30 primary AML blood samples through quantitative RT-PCR. Both the (-309T>G) genotypes and the MDM2 expression were correlated with disease free survival (DFS) rates among patients who have achieved complete remission (CR) after first induction chemotherapy. Results: MDM2-309T>G polymorphism was significantly associated with AML development (p<0.0001). The presence of either GG genotype or G allele at MDM2-309 confered 1.79 (95% CI: 1.12-2.86; p<0.001) and 1.46 fold (95%CI: 1.14-1.86; p= 0.003) increased AML risk. Survival analysis revealed that CR+ve cases with GG genotype had significantly increased DFS rates (16months, p=0.05) compared to CR+ve TT (11 months) and TG (9 months) genotype groups. Further, MDM2 expression was also found to be significantly elevated in GG genotype patients (p=0.0039) and among CR+ve cases (p=0.0036). Conclusions: The MDM2-309T>G polymorphism might be involved in AML development and also serve as a good prognostic indicator.

      • Induction of Th1-Biased T Follicular Helper (Tfh) Cells in Lymphoid Tissues during Chronic Simian Immunodeficiency Virus Infection Defines Functionally Distinct Germinal Center Tfh Cells

        <P>Chronic HIV infection is associated with accumulation of germinal center (GC) T follicular helper (Tfh) cells in the lymphoid tissue. The GC Tfh cells can be heterogeneous based on the expression of chemokine receptors associated with T helper lineages, such as CXCR3 (Th1), CCR4 (Th2), and CCR6 (Th17). However, the heterogeneous nature of GC Tfh cells in the lymphoid tissue and its association with viral persistence and Ab production during chronic SIV/HIV infection are not known. To address this, we characterized the expression of CXCR3, CCR4, and CCR6 on GC Tfh cells in lymph nodes following SIVmac251 infection in rhesus macaques. In SIV-naive rhesus macaques, only a small fraction of GC Tfh cells expressed CXCR3, CCR4, and CCR6. However, during chronic SIV infection, the majority of GC Tfh cells expressed CXCR3, whereas the proportion of CCR4(+) cells did not change, and CCR6(+) cells decreased. CXCR3(+), but not CXCR3(-), GC Tfh cells produced IFN-gamma (Th1 cytokine) and IL-21 (Tfh cytokine), whereas both subsets expressed CD40L following stimulation. Immunohistochemistry analysis demonstrated an accumulation of CD4(+) IFN-gamma(+) T cells within the hyperplastic follicles during chronic SIV infection. CXCR3(+) GC Tfh cells also expressed higher levels of ICOS, CCR5, and alpha 4 beta 7 and contained more copies of SIV DNA compared with CXCR3(-) GC Tfh cells. However, CXCR3(+) and CXCR3(-) GC Tfh cells delivered help to B cells in vitro for production of IgG. These data demonstrate that chronic SIV infection promotes expansion of Th1-biased GC Tfh cells, which are phenotypically and functionally distinct from conventional GC Tfh cells and contribute to hypergammaglobulinemia and viral reservoirs.</P>
