        Genomics and expression analysis of DHHC-cysteine-rich domain S-acyl transferase protein family in apple

        S-acylation is one of a group of lipid modifications that occurs on eukaryotic proteins, mediated by DHHC-CRD-containing proteins, which plays an important role in regulating the membrane association, trafficking and function of target proteins. Although genome-wide identification of PAT family has been carried out in yeast, mice, humans and Arabidopsis, little is known about apple PAT genes. In this study, a total of 33 putative apple PAT proteins, containing DHHC-CRD by domain analysis, have been identified, and were classified into three groups according to the phylogenetic analysis of PAT proteins in apple and Arabidopsis. More complex TMDs in the most MdPATs revealed the PM location of the gene family. Gene structure, gene chromosomal location and paralogs analysis of MdPAT genes within the apple genome demonstrated that tandem and segmental duplications, as well as whole genome duplications, have likely contributed to the expansion and evolution of the PAT gene family in apple. According to the microarray and expressed sequence tag (ESTs) analysis, the different expression patterns indicate that they may play different roles during fruit development and rootstock-scion interactions process. Moreover, PATs were performed expression profile analyses in different tissues, indicating that the PATs are involved in various aspects of physiological and developmental processes of apple. To our knowledge, this is the first report of a genome-wide analysis of the apple PAT gene family, and this genomic analysis of apple DHHCCRD PAT genes provides the first step towards a functional study of this gene family in apple.

        Black box-assisted fine-grained hierarchical access control scheme for epidemiological survey data

        Epidemiological survey is an important means for the prevention and control of infectious diseases. Due to the particularity of the epidemic survey, 1) epidemiological survey in epidemic prevention and control has a wide range of people involved, a large number of data collected, strong requirements for information disclosure and high timeliness of data processing; 2) the epidemiological survey data need to be disclosed at different institutions and the use of data has different permission requirements. As a result, it easily causes personal privacy disclosure. Therefore, traditional access control technologies are unsuitable for the privacy protection of epidemiological survey data. In view of these situations, we propose a black box-assisted fine-grained hierarchical access control scheme for epidemiological survey data. Firstly, a black box-assisted multi-attribute authority management mechanism without a trusted center is established to avoid authority deception. Meanwhile, the establishment of a master key-free system not only reduces the storage load but also prevents the risk of master key disclosure. Secondly, a sensitivity classification method is proposed according to the confidentiality degree of the institution to which the data belong and the importance of the data properties to set fine-grained access permission. Thirdly, a hierarchical authorization algorithm combined with data sensitivity and hierarchical attribute-based encryption (ABE) technology is proposed to achieve hierarchical access control of epidemiological survey data. Efficiency analysis and experiments show that the scheme meets the security requirements of privacy protection and key management in epidemiological survey.

      • Arginine-Rich Manganese Silicate Nanobubbles as a Ferroptosis-Inducing Agent for Tumor-Targeted Theranostics

        <P>Ferroptosis, an iron-based cell-death pathway, has recently attracted great attention owing to its effectiveness in killing cancer cells. Previous investigations focused on the development of iron-based nanomaterials to induce ferroptosis in cancer cells by the up-regulation of reactive oxygen species (ROS) generated by the well-known Fenton reaction. Herein, we report a ferroptosis-inducing agent based on arginine-rich manganese silicate nanobubbles (AMSNs) that possess highly efficient glutathione (GSH) depletion ability and thereby induce ferroptosis by the inactivation of glutathione-dependent peroxidases 4 (GPX4). The AMSNs were synthesized <I>via</I> a one-pot reaction with arginine (Arg) as the surface ligand for tumor homing. Subsequently, a significant tumor suppression effect can be achieved by GSH depletion-induced ferroptosis. Moreover, the degradation of AMSNs during the GSH depletion contributed to <I>T</I><SUB>1</SUB>-weighted magnetic resonance imaging (MRI) enhancement as well as on-demand chemotherapeutic drug release for synergistic cancer therapy. We anticipate that the GSH-depletion-induced ferroptosis strategy by using manganese-based nanomaterials would provide insights in designing nanomedicines for tumor-targeted theranostics.</P> [FIG OMISSION]</BR>

        Study on the role of gga-miRNA-200a in regulating cell differentiation and proliferation of chicken breast muscle by targeting Grb2

        Growth factor receptor-bound protein 2 (Grb2) have been proved by a lot of studies playing a major role in cell proliferation and cell differentiation. However, the regulation of Grb2 expression by microRNAs (miRNAs) in chicken breast muscle still remains unknown. The expression profile of Grb2 was checked based on our previous RNA sequencing data and the Grb2 relative expression level in breast muscle of aged hens (55-week-old) was validated significantly higher than juvenile hens (20-week-old) using qRT-PCR. miRNAs that interact with Grb2 have been predicted in chicken and the relationship between the potential miRNA and Grb2 was verified using dual luciferase reporter assay in chicken DF1 cells. Dual-luciferase reporter assays results demonstrated that the expression of luciferase reporter gene linked with part sequence of the 3′UTR of chicken Grb2 gene was down-regulated by the overexpression of gga (Gallus Gallus)- miR-200a-3p in the DF1 cells, and the down-regulation behavior was abolished when the ggamiR- 200a-3p binding site in 3′UTR of Grb2 was mutated, indicating that gga-miR-200a can suppress the expression level of its target gene Grb2. Therefore, we concluded that the significantly increased expression level of Grb2 in the breast muscle of aged chicken can (at least partly can) be explained by the decreased expression of miR-200a, which reduced the inhibitory effect on Grb2. Taken together, these findings suggest that gga-miR-200a can suppress the expression level of its target gene Grb2 and might be involved in the cell differentiation and proliferation of chicken breast muscle through binding with the 3’UTR of Grb2.
