PLGA/DBP 필름에서 DBP 함량에 따른 슈반씨세포와 후각초성세포의 부착과 성장에 미치는 영향

Poly(lactic-co-glycolic acid)(PLGA) is a biodegradable synthetic polymer with acceptable mechanical strength and well-controlled degradation rate. Also, it can be easily fabricated into many shapes. Demineralized bone particle(DBP) contains powerful bioactive molecules. In this study, we fabricated natural/synthetic biomaterial hybrid films using 0, 10, 20, 40 and 80 % of DBP. Schwann cells(SCs) and olfactory ensheathing cells(OECs) were seeded on PLGA/DBP film and confirmed the effects of adhesion and proliferation on SCs and OECs according to content of DBP. PLGA/DBP composite films are easily fabricated and provided bioactive environment that enhance the cell adhesion and proliferation. In this study, we confirmed that specific 10~20% DBP in PLGA/DBP films was superior to adhesion and proliferation of SCs and OECs and content of DBP such as 40% and 80% could not help cell growth and proliferation. These results suggest that DBP, natural material, provide suitable surface to neural cells and proper content affect on culture condition to improve cell adhesion and proliferation. In addition, these information would contribute to design of 3-dimentional PLGA/DBP scaffold for tissue engineered spinal cord regeneration.
Effects of poly‐γ‐glutamic acid on the physicochemical characteristics of skim milk yoghurt

<P>The physicochemical and sensory properties of skim milk yoghurts containing poly‐γ‐glutamic acid (PGA) at different levels (0.0025, 0.005 and 0.01%) were evaluated. Addition of PGA up to 0.01% to reconstituted skim milk (11%, w/v) did not affect the growth of lactic acid bacteria or the development of titratable acidity in yoghurt, whereas full‐fat control yoghurt had reduced acid production. No changes were found in viable cell counts of PGA yoghurts during storage (4 weeks at 4 °C). The addition of PGA (0.005%) significantly decreased syneresis in skim milk yoghurt and did not cause any undesirable effects in sensory acceptability.</P>

Demineralized bone particle(DBP) contains powerful bioactive molecules. Bone marrow mesenchymal stem cells(BMSCs) are important progenitor and supporting cells that have the intrinsic ability to self-renewal and differentiate into multiple functional cells. In this study, we fabricated natural/ synthetic hybrid PLGA scaffolds included 20% of DBP(PLGA/DBP). Schwann cells(SCs) and olfactory ensheathing cells(OECs) were seeded on PLGA/DBP scaffolds. We confirmed that effects of PLGA/DBP scaffolds seeded OEC and SC on proliferation and differentiation of BMSCs. PLGA/DBP scaffolds seeded OECs and SCs were superior to differentiation into neuronal cell proliferation of BMSCs. This phenomenon was expected due to cytokine secreted by OECs and SCs promote proliferation and differentiation of BMSCs.

Olfactory ensheathing cells(OEC) have been reported to migrate long distances and to guide axonal regeneration after spinal cord injury(SCI). Bone morphogenetic proteins(BMPs), members of the TGF-β family of signaling molecules, have numerous functions in nervous system development. In this study, Therefore, we investigated interaction of OEC with poly(lactide-co-glycolide)(PLGA)/demineralized bone particle(DBP) scaffold according to content of DBP. We manufactured PLGA/DBP scaffold by solvent casting/salt leaching method. Cell proliferation seeded into the scaffold were assayed MTT test and NF, S-100, P75 and BDNF were expressed in OEC cultured PLGA/DBP scaffold according to content of DBP. We examined the cell adhesion and proliferation by scanning electron microscopy observation. We confirmed that OEC adhesion and proliferation was increased on PLGA/ DBP scaffold of higher content of DBP. In these results, we recognized that PLGA/DBP scaffold provide the suitable environment for improvement of cell proliferation and formation of extra cellular matrix.
궤양성 대장염 활성도 평가에서 Erythrocyte Sedimentation Rate과 C-reactive Protein의 유용성 비교

목적: 임상에서 질병 활성도를 객관적으로 평가하기 위해 흔히 사용하는 Truelove & Witts의 활성도 분류는 염증의 지표로 검사실 소견 중 erythrocyte sedimentation rate (ESR)를 사용하고 있으나, C-reactive protein (CRP)도 염증의 지표로 자주 사용하고 있다. 저자들은 임상에서 궤양성 대장염 환자의 추적 검사 중 ESR과 CRP가 일치하지 않는 경우를 드물지 않게 경험하여 두 검사실 소견 중 어느 것이 환자들이 보고하는 임상 증상과 잘 일치하는지 알아보기 위해 후향 연구를 시행하였다. 대상 및 방법: 궤양성 대장염으로 진단되어 추적 관찰 중인 155명의 환자(541 회의 임상 증상 평가)를 대상으로 환자들의 의무기록을 조사하여 진단 당시의 질환 범위, 이환 기간과 환자들이 기록하는 임상 증상 설문지를 통해 배변 횟수, 혈변의 정도, 긴박감, 전신상태, 복통, 발열, 식욕부진, 구역, 구토 유무를 확인하였다. 그리고 검사실 소견으로 ESR, CRP, 백혈구, 혈색소, 알부민, 혈소판을 확인하였다. 상관 분석은 Pearson`s correlation coefficient를 이용하여 분석하였다. 결과: 전체 155명 환자, 541회 평가에서 ESR과 CRP의 Pearson`s correlation coefficient가 0.403 (p=0.000)으로 높은 상관성을 보였고, 임상 증상과 ESR과 CRP의 각각의 Pearson`s correlation coefficient는 0.376, 0.258 이었다. 질병 활성도가 중증인 경우 correlation coefficient는 ESR 0.629, CRP 0.516으로 질병 활성도가 높을수록 임상 증상 정도를 ESR과 CRP가 잘 반영하였다. 총 131예(24.2%)에서 ESR과 CRP의 불일치를 보였으며, ESR과CRP의 correlation coefficient는 각각 0.338, 0.034이었다. 불일치군을 high ESR/low CRP군과 low ESR/high CRP군으로 구분하여 임상 증상과의 상관성을 보았을 때 high ESR/low CRP군에서는 correlation coefficient가 ESR 0.420, CRP 0.226으로 ESR과 더 잘 일치되었고, low ESR/high CRP군에서도 correlation coefficient가 ESR 0.333 (p=0.038), CRP 0.068 (p>0.01)로 ESR이 임상 증상과 더 잘 일치하였다. 결론: 궤양성 대장염 환자의 질병 활성도를 평가하는 경우 ESR이 더 유용한 검사실 소견이었다. 특히 ESR과 CRP 상승이 차이를 보일 때는 ESR의 상승이 임상 증상을 더욱 잘 반영하였다. Background/Aims: Although erythrocyte sedimentation rate (ESR) is included as a laboratory parameter in Truelove and Witts` classification, C-reactive protein (CRP) is also used for severity assessment in ulcerative colitis (UC). Frequently, the discordance between ESR and CRP is observed in clinical practice. The aim of this study was to determine which parameter is more related with clinical activity in UC patients. Methods: A total of 155 patients with UC were identified from January 2004 to March 2005. Their medical records were reviewed within these patients, a total of 541 assessments of disease activity were made. Correlation of clinical activity and laboratory tests were evaluated by Pearson`s correlation coefficient. Results: Pearson`s correlation coefficients of ESR and CRP with clinical symptoms were 0.376 and 0.258, respectively. The correlation coefficient between ESR and CRP was 0.403 (p=0.000). A total of 131 (24.2%) assessments revealed discordance between ESR and CRP. When discordance occurred, the correlation coefficients with clinical symptoms were 0.338 for ESR (p=0.000) and 0.034 for CRP (p>0.01). Dividing discordant patients into high ESR/low CRP group and low ESR/high CRP group, the coefficients were 0.420 for ESR and 0.226 for CRP in high ESR/low CRP group, and 0.333 for ESR and 0.068 for CRP in low ESR/high CRP group. Conclusions: The correlation analysis indicates that ESR appears to be a more reliable laboratory parameter of disease activity than CRP in assessing the severity of UC. In particular, when the level of ESR and CRP is discordant, ESR is more useful in assessing the disease activity in UC patients. (Korean J Gastroenterol 2006;48:313-320)
Indirect Reduction Technique in Proximal Humeral Fractures Stabilized by Locking Plates

Background: Indirect reduction technique offers a valid option in the treatment of proximal humerus fracture. The purpose of this study is to evaluate the functional outcome and the complication rate after indirect reduction and internal fixation of unstable proximal humeral fractures with use of a locking plate. Methods: Twenty four patients with acute proximal humerus fracture were managed with indirect reduction and internal fixation with a locking plate. The mean follow-up period was 15.5 months. Results: The anatomical reduction of the medial cortex buttress was seen in 16 patients (66%) of the Group A and the non-anatomical reduction was seen in 8 patients (33%) of the Group B. Mean union time was 3.2 ± 1.9 months; it was 2.2 ± 0.6 months in the Group A and 5.3 ± 2.2 months in the Group B (p < 0.05). In our series, there were 6 cases of complications and these include 2 cases of varus malunion, 2 cases of shoulder stiffness, 1 case of heterotrophic ossification, 2 cases of screw perforation and 1 case of impingement. Conclusions: We conclude from our studies that indirect reduction and internal fixation using locking plate for acute proximal humerus fracture can give good results with bony union and predictable good overall functional outcome. If the medial cortex buttress is well maintained, a better anatomical reduction would be achieved, the union would be prompted, the pain would be further reduced and the range of the motion would be recovered more promptly.
