      • 헤모글로빈의 Microencapsulation에 의한 인공적혈구의 산소전달과 용혈도에 관한 연구

        In this study, we tried to improve oxygen transfer rate and reduce pressure drop on intravascular lung assist device with microencapsulation of red blood cells, because microcapsulation of red cells reduce blood hemolysis. Hemoglobin was purified from outdated human red blood cells used. Phospholipid was purified from egg yolk. The hemosome was prepared by encapsulation of hemoglobin with phospholipid multilayer, using rotary vacuum evaporator. The function of hemosome and hemosome/blood mixed solution was tested by measuring oxygen dissociation curve using blood gas analyzer. Also we were measured about hemolysis of hemosome, hemosome/blood mixed solution and blood by absorptivity method. As a results, the oxygen dissociation curve of hemosome and hemosome/blood mixed solution showed the same figure as the curve of normal red blood cells. Also, the hemolysis of hemosome and hemosome/blood mixed solution was measured less than normal red blood cell. Therefore, the oxygen transfer rate showed high use of hemosome, and hemosome/blood mixed solution rather than normal blood. Because microen- capsulation of normal red blood cells prevent hemolysis of normal red blood cells.

        When the Microporous hollow fiber membrane(MHFM) was contacted with whole blood, the serum leakage(membrane fouling) was occurred in the MFHM. In general, the MHFM was in use at most intravascular lung assist device(IVALAD) which was often occurred the serum leakage, and these serum leakage were reduced the gas exchange. In this work, we tried to improve gas exchange on the IVALAD by using vibration technique. As a result, it was showed that the vibrating method of this module was very effective due to the reduction of serum leakage on MFHM. Therefore, this method was increased gas exchange, respectively.

      • 혈관 내 폐 보조장치에서의 기체교환속도의 향상

        We intended to develope an vibrating intravascular lung assist device(VIVLAD) in order to exchange of efficiently gas transfer of oxygen and carbon dioxide exchange requirements for end-stage acute respiratory distress syndrome (ARDS) patients. However, the clinical study, concluded that more gas exchange was needed for intravascular oxygenation to be clinically effective in ARDS treatment. In this study, we tried to enhance gas exchange on the VIVLAD by using microencapsulation of hemoglobin, as the microencapsulation of hemoglobin reduces blood hemolysis, and perfluorocarbon emulsion(PFC emulsion). As a result, it was shown that the oxygen transfer of hemosome and blood/hemosome mixed solution were higher than that of whole blood. Also, it showed that the carbon dioxide transfer of whole blood/PFC emulsion mixed solution was higher than that of others. Therefore, we determined that hemosome and PFC emulsion could increase oxygen transfer and carbon dioxide transfer, respectively.

        Chemical Composition of Seed in Medicinal Soybean Collected in Korea

        Production of medicinal soybean [Glycine max (L.) Merrill], characterized with black seed, white stripe at hilum border, yellow cotyledon and small seed, is increasing with increasing consumption. The objective of this study was to investigate the chemical composition of medicinal soybean seed and to provide basic information, for the characterization of these soybeans among genetic resources. Forty-four lines of medicinal soybeans collected from Korea and two control cultivars, 'Hwangkeumkong' (Yellow seed coat) and 'Geomjeongkong l' (Black seed coat) were planted at the Research Farm of the College of Natural Resources, Korea University, located at Namyangju City on May 25, 1996. Seeds of these lines were harvested at full maturity and analyzed for protein, oil, sugar, starch and mineral contents. Mean protein and oil content of the medicinal line seeds were 42.6 and 16.1%, respectively, and those of the control cultivars were in the middle range for protein and oil content. However, sugar and starch content of the medicinal line seeds appeared to be in the lower range of the distributions compared to the control cultivars and were 10.0 and 1.68%, respectively. Mean P, K, Ca, and Mg contents of the seeds of medicinal soybean lines were 15.9, 21.5, 3.11, and 2.81 mg/g, respectively, indicating that these lines had higher P, K, and Mg and lower Ca contents when compared to the control cultivars. Mean Na and Fe contents were 671 and 224 mg/kg, respectively, showing lower Na and similar Fe contents. The observed results provided that chemical compositions of medicinal soybean were, on average, different from those of the general soybean cultivars.

        Responses of Soybean Cultivars to Excessive Soil Moisture Imposed at Different Growth Stages

        Soybean [Glycine max (L.) Merrill] crops, grown in a rice soybean rotation, can suffer when grown in soil with excessive moisture. The objective of this work were to determine the reduction in growth and yield, responses of vegetative and reproductive growth of soybean to excessive soil moisture achieved by prolonged irrigation. Responses of different cultivars were determined at growth stages from V6 to R8 to clarify the sensitive growth stages or characteristics to excessive soil moisture. Cultivar differences in response to excessive soil moisture condition were conspicuous in seed dry weight and harvest index (HI) but not in the response of seed number or pod number per plant. The timing of irrigation causing the condition of excessive soil moisture influenced the vegetative or reproductive traits. Soybean plants were more affected by irrigation commencing at the pre-flowering than at the post-flowering stage. Post-flowering irrigation did not reduce growth of vegetative organs significantly; in fact the growth of stems and leaves was facilitated by the prolonged irrigation commencing at flowering. Differences between cultivar response to prolonged irrigation were assumed to relate to the reduced amount of assimilates translocated to the reproductive organ.

        Effects of Temperature and Moisture Level during Preconditioning on Germination and Seedling Elongation of Soybean Seeds with and without Osmoconditioning

        삼투처리와 무처리의 대두종자(품종 :Williams)에 대하여 두가지 온도(15, 3$0^{\circ}C$)에서 두가지의 종자수분함량(30%, 50%)으로 조절된 조건으로 0. 2, 4 및 8일간 전처리함에 있어서 시간별 세세구배치법 4반복으로 배치하여 발아와 묘신장을 조사하였던 바 주요결과는 다음과 같다. 1. 콩의 묘신장은 30에서 2일간 전처리함으로서 촉진되었으나 같은 온도에서 전처리기간이 길수록 억제되었다. 2. 삼투처리후 전처리된 종자는 수분흡수와 묘신장이 떨어졌다. 3. 종자수분함량이 50%로 조절된 뒤 전처리된 것은 체의 수분함량을 증가시켰으며 4일간까지의 전처리는 묘신장을 크게 하였다. 4. 묘의 건물중에서는 전처리온도가 3$0^{\circ}C$ 일때와 종자수분함량이 50%로 조절되어 8일간 전처리하였을 때 감소되었다. Germination and seedling length of soybean seeds (cultivar: Williams) with and without polyethylene glycol-8000 (PEG) treatments were measured following preconditioning at two temperatures (15 and 30$^{\circ}C$) and two moisture levels 〔low (30 percent) and high (50 percent seed water content)〕 for 0, 2, 4, or 8 days. A split-split plot in time was used with four replications. Observations were made after two days of germination at 30$^{\circ}C$ Seedling growth accelerated with two days of preconditioning at 30$^{\circ}C$, but was reduced as preconditioning duration increased up to eight days at the same temperature. PEG treated preconditioned seeds exhibited reduced moisture uptake and seedling growth. Preconditioning at a high moisture level increased seedling moisture content and also increased seedling length until four days of preconditioning duration. Seedling dry weight decreased when preconditioning temperature was 30$^{\circ}C$ and when the high moisture level of preconditioning was continued for eight days.

        Effects of Ridge Height in Dry Paddy Field on Growth and Seed Yield of Soybean Cultivars

        Excessive soil water in paddy field induces growth losses during the vegetative stages of soybean plants. Our objectives were to know growth responses of soybean as affected by the level of ridge heights. A field experiment was conducted at the Research Farm of Korea University near Seoul in 1996. Ten cultivars of soybean (Glycine max (L.) Merrill) were planted at the ridge heights of 10, 30, and 50cm in paddy soil on 27 June. Total dry matter accumulation at the 10cm ridge height was severely decreased until the growth stage of R5 as compared to the ridge heights of 30 and 50cm, and this decrement was mainly due to both reduced leaf and stem dry weights. However, seed dry weight was not significantly decreased at the lower ridge height with the increment of harvest index. Cultivar differences were found on seed dry weight, crop growth rate from R1 to R5, and leaf/stem ratio of R5. From the above results, soybean growth responses to excessive soil water in paddy field were different between seed filling stage and late vegetative to early reproductive growth stages
