        Preservation of Acid Treated Bivoltine Eggs in Silkworm Bombyx mori L.

        The hybrid ($CSR2{\times}CSR4$) eggs treated with acid were taken up for the study with an objective to develop long-term preservation schedule. The hybrid eggs obtained with two mating duration (3 h and 6 h) and oviposition period (6 h and 24 h) with two age groups of eggs (24 h and 36 h) were treated with Hydrochloric acid. These eggs were subjected to preservation at $5^{\circ}C$ in single step refrigeration and at $5^{\circ}C$ and $2.5^{\circ}C$ under double step refrigeration from $10{\sim}120$ days. These eggs were released from the cold storage as per the specified durations and incubated at standard conditions and allowed 2 h for hatching at 450 lux light. Hatchability was found to be significantly higher or on par with the control in three treatments (T1, T2 and T4) where the eggs are preserved continuously at $5^{\circ}C$ up to 30 days. However under double step refrigeration, hatching was not significantly affected in 20+60 day's combination of T1 treatment up to 80 days. Bioassay studies of the promising treatment i.e.. T1 with (20+60) days indicated that early stage loss and cocoon yield was found to be on par with the control. Hence this treatment was recommended for preservation of acid treated new bivoltine hybrid layings. Details of the hatchability and rearing performance of long term preservation of acid treated eggs are discussed.

        In order to study the feasibility of utilizing bivoltine foundation cross (FC2) males of the silkworm as male components with Pure Mysore (PM), six foundation crosses viz,CSR2 x CSR27, CSR27 x CSR2, CSR27 x CSR50, CSR50 x CSR27, CSR2 x CSR50 and CSR50 x CSR2 along with CSR2 pure breed were assessed for performance of parental crop,grainage, rearing and reeling performance. The foundation crosses exhibited better performance than CSR2 as a parental crop and male component which resulted in higher egg recovery. Though there was not much difference between PM crossed with FC2 and CSR2 males pertaining to important bioassay parameters, PM x CSR2 exhibited superiority in reeling traits particularly with reelability and raw silk %. This study reveals that FC2 seed cocoons can be utilized during exigency when there is a dearth for CSR2 seed cocoons but not regularly.

        Vibration and buckling analyses of laminated panels with and without cutouts under compressive and tensile edge loads

        In this study, the influence of centrally placed circular and square cutouts on vibration and buckling characteristics of different ply-oriented laminated panels under the action of compressive and/or tensile types of nonuniform in-plane edge loads are investigated. The panels are inspected under the action of uniaxial compression, uniaxial tension and biaxial, compression-tension, loading configurations. Furthermore, the effects of different degrees of edge restraints and panel aspect ratios are also addressed in this work. Towards this, a nine-node heterosis plate element has been adopted which includes the effect of shear deformation and rotary inertia. According to the results, the tensile buckling loads are higher than that of compressive buckling loads. However, the tensile buckling load continuously reduces with the increased cutout sizes irrespective of ply-orientations. This is also true for compressive buckling loads except for some particular ply-orientations with higher sized cutouts.

        In this paper, we establish three new continued fraction expansions for the ratios of the basic hypergeometric series 2'1 by employing four q-difference equations. As particular case of these, we obtain certain new continued fractions including the continued fractions for the quotient of Ramanujan-G¨ollnitz-Gordon function.

      • Optimum arrangement of stiffener on the buckling behaviour of stiffened composite panels with reinforced elliptical cutouts subjected to non-uniform edge load

        Cutouts in the beams or plates are often unavoidable due to inspection, maintenance, ventilation, structural aesthetics purpose, and sometimes to lighten the structures. Therefore, there will be a substantial reduction in the strength of the structure due to the introduction of the cutouts. However, these cutouts can be reinforced with the different patterns of ribs (stiffener) to enhance the strength of the structure. The present study highlights the influence of the elliptical cutout reinforced with a different pattern of ribs on the stability performance of such stiffened composite panels subjected to non-uniform edge loads by employing the Finite element (FE) technique. In the present formulation, a 9-noded heterosis element is used to model the skin, and a 3-noded isoparametric beam element is used to simulate the rib that is attached around a cutout in different patterns. The displacement compatibility condition is employed between the plate and stiffener, and arbitrary orientations are taken care by introducing respective transformation matrices. The effect of shear deformation and rotary inertia are incorporated in the formulation. A new mesh configuration is developed to house the attached ribs around an elliptical cutout with different patterns. Initially, a study is performed on the panels with different stiffener schemes for various ply orientations and for different stiffener depth to width ratios (ds/bs) to determine an optimal stiffener configuration. Further, various parametric studies are conducted on an obtained optimal stiffened panel to understand the effect of cutout size, cutout orientation, panel aspect ratio, and boundary conditions. Finally, from the analysis, it can be observed that the arrangement of the stiffener attached to a panel has a major impact on the buckling capacity of the stiffened panel. The stiffener’s depth to width ratio also significantly influences the buckling characteristic.

        Cytotoxicity and Proteasome Inhibition by Alkaloid Extract from Murraya koenigii Leaves in Breast Cancer Cells—Molecular Docking Studies

        Murraya koenigii (curry tree) leaves are rich in bioactive compounds such as flavonoids, alkaloids, and coumarins. Alkaloids from M. koenigii leaves have antianalgesic, antiulcerogenic, antiobesity, and antitumor activities. In this study, we tested the cytotoxic and proteasome-inhibitory potential of a total alkaloid extract (TAE) from M. koenigii leaves in the breast cancer cell line MDA-MB-231. The TAE decreased cell viability with an IC50 of 14.4 μg/mL and altered growth kinetics of breast cancer cells. TAE (32 μg/mL) arrested cells (35%) in the “S” phase of the cell cycle and induced apoptosis. The 26S proteasome, a multicatalytic protease complex, promotes tumor cell proliferation and protects tumor cells from apoptosis. The TAE and mahanine, a carbazole alkaloid present in M. koenigii leaves, preferentially inhibited the trypsin-like, but not the chymotrypsin-like proteolytic activity of the proteasome with an IC50 of 162 μg/mL and 287 μM, respectively. In silico analysis of 26 compounds from M. koenigii leaves revealed significant docking scores for mahanine and two other carbazole alkaloids with the β2 and β5 subunits of the catalytic 20S proteasome. Taken together, this study demonstrates that inhibition of the proteasome is an important biological activity of M. koenigii alkaloids, which may lead to cancer cell death.

        Characterization of Microsatellite Markers, Their Transferability to Orphan Legumes and Use in Determination of Genetic Diversity among Chickpea (Cicer arietinum L.) Cultivars

        The microsatellite or simple sequence repeat (SSR) marker is the most preferred marker because of its many desirable properties. It is important to increase the genic and genomic resources particularly in legumes because the SSR markers currently available inchickpea, pigeonpea, horsegram, blackgram, and cowpea are very limited. In the present study, 201 pairs of SSR markers comprisingof 172 genic and 29 genomic SSRs were screened against 11 chickpea genotypes, among which 153 produced monomorphic and 48produced polymorphic bands. The polymorphic information content ranged from 0.152 to 0.373 for both genic and genomic SSRs. Among the polymorphic markers, two-three alleles were detected for genic and two-four alleles for genomic SSRs. A unique bandingpattern could be found for all the genotypes within 48 polymorphic SSR markers and cultivar specific markers could be identified forseed purity test. We have also studied the ability of chickpea genic and genomic SSRs to amplify distantly related but importantlegumes viz., horsegram, blackgram, cowpea, pigeonpea, and soybean. Out of 201 chickpea SSR primer pairs, 66.7% in blackgram,62.2% in horsegram, 61.7% in redgram, 54.7% in cowpea, and 62.7% in soybean produced amplification. The transferability ofabout 60.0% of the chickpea SSRs to distantly related legumes could be considered successful. In the present study, 134, 133, 126,124, and 110 new SSR primers for blackgram, horsegram, soybean, redgram, and cowpea pulse crops, respectively, were identified. Itis an important addition to the already available genomic resources in these crops. In addition, among genic primer pairs, 12 in horsegram,three in soybean, 13 in redgram, and eight in cowpea, and among genomic primer pairs, two in horsegram and four in redgramwere polymorphic even in the two-three genotypes tested indicating their potentially for application in genetic studies and mapping
