        Single Nucleotide Polymorphisms of the GnRHR Gene Associated with Reproductive Traits of Japanese Flounder (Paralichthys olivaceus)

        Gonadotropin-releasing hormone receptor (GnRHR) gene is expressed at the anterior pituitary gland and plays a key role in gonad development. This study aimed to investigate molecular genetic characteristics of the GnRHR gene and elucidate the effects of single nucleotide polymorphisms (SNPs) of GnRHR gene on sex steroid level in Japanese flounder (Paralichthys olivaceus). We used polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) and sequencing of the GnRHR gene in 75 individuals. We identified three SNPs in the GnRHR gene: P1 locus (C759A and C830T) in the coding region of exon2 which were both linked together and P2 locus (G984T) in the coding region of exon3, which added a new transcript factor (ADR1) and a new methylation site (CG). Only C830T of P1 leads to amino acid changes Thr266Ile. Statistical analysis showed that P1 was significantly associated with $17{\beta}$-estradiol ($E_2$) level (p<0.01) and gonadosomatic index (GSI) (p<0.05). Individuals with genotype BB of P1 had significantly higher serum $E_2$ levels (p<0.01) and GSI (p<0.05) than those of genotype AA or AB. Another SNP, P2, synonymous mutation, was significantly associated with GSI (p<0.05). Individuals with genotype AB of P2 had significantly higher GSI (p<0.05) than that of genotype AA. In addition, there was a significant association between one diplotype based on three SNPs and reproductive traits. The genetic effects for both serum $E_2$ level and GSI of diplotype D4 were super diplotypes (p<0.05). These results suggest that the SNPs in Japanese Flounder GnRHR are associated with $E_2$ level and GSI.

      • A Multicast Search Scheme Based on Bipartite Graph Matching Model

        This paper proposes an efficient multicast search scheme based on bipartite graph matching model, aiming at one of the most important problems in the multicast service, how can the wireless network track and locate the mobile uses in the idle state, under the tight bandwidth and delay constraints,. By quantifying the location uncertainty of mobile users with Shannon's entropy, the scheme adopts the LZ78 compression algorithm to design location update scheme and predict the location probability, in order to reduce the cost of location update. The multicast search system need to guarantee the maximum total probability that each user resides at the assigned paging area, for the purpose of an optimal performance on both expected paging delay and paging cost. Therefore, the bigraph-based multicast paging scheme (BMPS) firstly builds the bipartite graph model of multicast search problem, through converting the location probability into weight of the edge. BMPS decides the optimal allocation between the mobile users and location areas, which is mainly achieved by the maximum weight perfect matching in bipartite graph, while modifying the weights dynamically. Simulation results show that BMPS yields a low search delay as well as a low search cost, and reduces the impact of user collision.

        Risk assessment of water inrush in karst tunnels based on a modified grey evaluation model: Sample as Shangjiawan Tunnel

        A modified grey clustering method is presented to systematically evaluate the risk of water inrush in karst tunnels. Based on the center triangle whitenization weight function and upper and lower limit measure whitenization weight function, the modified grey evaluation model doesn't have the crossing properties of grey cluster and meets the standard well. By adsorbing and integrating the previous research results, seven influence factors are selected as evaluation indexes. A couple of evaluation indexes are modified and quantitatively graded according to four risk grades through expert evaluation method. The weights of evaluation indexes are rationally distributed by the comprehensive assignment method. It is integrated by the subjective factors and the objective factors. Subjective weight is given based on analytical hierarchy process, and objective weight obtained from simple dependent function. The modified grey evaluation model is validated by Jigongling Tunnel. Finally, the water inrush risk of Shangjiawan Tunnel is evaluated by using the established model, and the evaluation result obtained from the proposed method is agrees well with practical situation. This risk assessment methodology provides a powerful tool with which planners and engineers can systematically assess the risk of water inrush in karst tunnels.

      • Early Efficacy of Endostar Combined with Chemoradiotherapy for Advanced Cervical Cancers

        The aim of this study was to investigate the early outcome of Endostar combined with chemoradiotherapy for advanced cervical cancer. Fifty-two cases (FIGO IIb to IVa) were divided randomly into two groups, receiving chemoradiotherapy alone (CRT group) and Endostar combined with chemoradiotherapy (CRT+E group). For the patients in the CRT+E group, Endostar was administered daily with the dosage of 7.5 $mg/m^2$, and cisplatin was administered weekly with the dosage of 20 $mg/m^2$ during the radiation. The regimens lasted for 4 weeks with no difference in chemoradiotherapy between the two groups. The early outcome complete remission rate was 73.1%, partial remission rate was 23.1% and the total response rate was 96.2% in CRT+E group, a significant improvement on the 34.6%, 42.3% and 76.9%, respectively, in the CRT group. One year survive rates were 100% and 84.6% in the CRT+E group and CRT groups, the difference being significant. Endostar combined with chemoradiotherapy can improve the early outcome of the advanced cervical cancer, and adverse effects were not encountered.

        Antinociceptive Effects of Prim-O-Glucosylcimifugin in Inflammatory Nociception via Reducing Spinal COX-2

        We measured anti-nociceptive activity of prim-o-glucosylcimifugin (POG), a molecule from Saposhnikovia divaricate (Turcz) Schischk. Anti-nociceptive or anti-inflammatory effects of POG on a formalin-induced tonic nociceptive response and a complete Freund``s adjuvant (CFA) inoculation-induced rat arthritis pain model were studied. Single subcutaneous injections of POG produced potent anti-nociception in both models that was comparable to indomethacin analgesia. Anti-nociceptive activity of POG was dose-dependent, maximally reducing pain 56.6% with an ED50 of 1.6 mg. Rats given POG over time did not develop tolerance. POG also time-dependently reduced serum TNFα, IL-1β and IL-6 in arthritic rats and both POG and indomethacin reduced spinal prostaglandin E2 (PGE2). Like indomethacin which inhibits cyclooxygenase-2 (COX-2) activity, POG dose-dependently decreased spinal COX-2 content in arthritic rats. Additionally, POG, and its metabolite cimifugin, downregulated COX-2 expression in vitro. Thus, POG produced potent anti-nociception by downregulating spinal COX-2 expression.

        Cross-Corresponding Preparation of Organometallic Acetylferrocene Micro-Nanostructures in Bidirection Using Top-Down and Bottom-Up Approaches

        The larch-fruit-like acetylferrocene micro-nanomaterials and beltlike acetylferrocene micro-nanomaterials have been cross-correspondingly prepared in bidirection by the top-down and bottom-up approach, respectively, and characterized by scanning electron microscope, transmission electron microscope and X-ray diffractometer. UV-Vis absorption spectra show that the redshift was found in the acetylferrocene micro-nanomaterials compared to that in solution. The formation mechanism of the acetylferrocene micro-nanomaterials is also proposed. This work provides a new strategy for the material synthesis and preparation and may realize the reuse of organometallic materials.

      • Early Efficacy of Taxotere and Cisplatin Chemo-Radiotherapy for Advanced Cervical Cancer

        The aim of this study was to investigate the early outcome of the taxotere and cisplatin chemoradiotherapy for advanced cervical cancer. Fifty-six cases (FIGO II b to IVa) were divided randomly into two groups: radiotherapy alone (28 cases) and radiation plus chemotherapy (TP) group. There was no difference in radiotherapy between the two groups. The RT+C cases who received TP regimen during the radiation, and DDP once weekly injection of vain, according to 20$mg/m^2$ and taxotere once weekly iv according to 35 $mg/m^2$. These regimens were given for 4~5weeks, and some medicines to control vomiting were available for the RT+C cases. The two groups received an oral medicine MA 160mg every day during the treatment. Regarding early outcome, the complete remission rate was 64.3% and partial remission rate was 35.7% in RT+C. The complete remission rate was 32.1% and partial remission rate was 39.3% in RT. The total response rate and complete remission in the RT+C group were higher than that in the RT group. We conclude that taxotere and cisplatin chemoradiotherapy can improve the early outcome of the advanced cervical cancer, the adverse effects being endurable.

        Detection of the expression of a Bombyx mori Atypical Protein Kinase C in BmPLV-Infected Larval Midgut

        Protein kinase C (PKC) is involved in many cellular signaling pathways, it participates in many physiological processes, such as cell cycle, growth, proliferation, differentiation and apoptosis. To investigate the effect of PKC on the silkworm midgut tissue infection of Bombyx mori parvo-like virus (BmPLV), a B. mori atypical protein kinase C (BmaPKC) gene was cloned from larval midgut tissue, expressed in E. coli and purified. Additionally, the BmPLV susceptible silkworm strain and resistant silkworm strain were used to test the effect of the B. mori infection on BmPLV. The result showed that BmaPKC encodes a predicted 586 amino acid protein, which contains a C-terminal kinase domain and an N-terminal regulatory domain. The maximum expression amount of the soluble (His)6-tagged fusion protein was detected after 0.8 mmol/L IPTG was added and cultured at 21˚C. The (His) 6-tagged fusion protein revealed about 73 kDa molecular weight which confirmed by western blot and mass spectrography. Furthermore BmaPKC protein were detected at 0-72 h post-infection in BmPLVinfected larval midgut tissue, western blot showed that as time went on, the expression of BmaPKC increased gradually in susceptible strain, the expression quantity on 72 h is 5 times of 0 h. However, in resistant strain, the expression quantity is slightly lower than susceptible strain. But no significant change in resistant strain was observed as time went on. The available data suggest that BmaPKC may involve in the regulation of BmPLV proliferation.

        The Three-wavelength PR<SUP>3+</SUP>:YLF Laser at 604 nm 607 nm and 640 nm with Fabry-Perot Etalon

        A three-wavelength Pr<SUP>3+</SUP>:YLF laser at 604 nm, 607 nm and 640 nm simultaneously output by Fabry-Perot (F-P) etalon has been obtained. A 444 nm blue laser diode is used for pumping the Pr<SUP>3+</SUP>:YLF crystal, and a 0.1 mm F-P etalon is inserted in the resonator to select wavelength. The theoretical model of three-wavelength Pr<SUP>3+</SUP>:YLF laser is established, by adjusting the tilt angle of the etalon, the transmittances of the different wavelengths can be controlled, and the threshold values can be made to equalize by controlling the loss among different wavelengths. In the experiment, when the tilt angle of etalon is 9° and the optimized length of resonator is 48 mm, the total output power of 25 mW at the three-wavelength is achieved at incident pump power of 7.5 W.
