        DETECTION AND COUNTING OF FLOWERS BASED ON DIGITAL IMAGES USING COMPUTER VISION AND A CONCAVE POINT DETECTION TECHNIQUE

        In this paper we propose a new algorithm for detecting and counting flowers in a complex background based on digital images. The algorithm mainly includes the following parts: edge contour extraction of flowers, edge contour determination of overlapped flowers and flower counting. We use a contour detection technique in Computer Vision (CV) to extract the edge contours of flowers and propose an improved algorithm with a concave point detection technique to find accurate segmentation for overlapped flowers. In this process, we first use the polygon approximation to smooth edge contours and then adopt the second-order central moments to fit ellipse contours to determine whether edge contours overlap. To obtain accurate segmentation points, we calculate the curvature of each pixel point on the edge contours with an improved Curvature Scale Space (CSS) corner detector. Finally, we successively give three adaptive judgment criteria to detect and count flowers accurately and automatically. Both ex- perimental results and the proposed evaluation indicators reveal that the proposed algorithm is more efficient for flower counting.

        The genus Achlya Billberg, 1820 is reported for the first time from China as A. jezoensis (Matsumura, 1927), which is a species collected from Maorshan, Heilongjiang Province.         Production of Transgenic Pigs with an Introduced Missense Mutation of the Bone Morphogenetic Protein Receptor Type IB Gene Related to Prolificacy

        In the last few decades, transgenic animal technology has witnessed an increasingly wide application in animal breeding. Reproductive traits are economically important to the pig industry. It has been shown that the bone morphogenetic protein receptor type IB (BMPR1B) A746G polymorphism is responsible for the fertility in sheep. However, this causal mutation exits exclusively in sheep and goat. In this study, we attempted to create transgenic pigs by introducing this mutation with the aim to improve reproductive traits in pigs. We successfully constructed a vector containing porcine BMPR1B coding sequence (CDS) with the mutant G allele of A746G mutation. In total, we obtained 24 cloned male piglets using handmade cloning (HMC) technique, and 12 individuals survived till maturation. A set of polymerase chain reactions indicated that 11 of 12 matured boars were transgene-positive individuals, and that the transgenic vector was most likely disrupted during cloning. Of 11 positive pigs, one (No. 11) lost a part of the terminator region but had the intact promoter and the CDS regions. cDNA sequencing showed that the introduced allele (746G) was expressed in multiple tissues of transgene-positive offspring of No.11. Western blot analysis revealed that BMPR1B protein expression in multiple tissues of transgene-positive $F_1$ piglets was 0.5 to 2-fold higher than that in the transgene-negative siblings. The No. 11 boar showed normal litter size performance as normal pigs from the same breed. Transgene-positive $F_1$ boars produced by No. 11 had higher semen volume, sperm concentration and total sperm per ejaculate than the negative siblings, although the differences did not reached statistical significance. Transgene-positive $F_1$ sows had similar litter size performance to the negative siblings, and more data are needed to adequately assess the litter size performance. In conclusion, we obtained 24 cloned transgenic pigs with the modified porcine BMPR1B CDS using HMC. cDNA sequencing and western blot indicated that the exogenous BMPR1B CDS was successfully expressed in host pigs. The transgenic pigs showed normal litter size performance. However, no significant differences in litter size were found between transgene-positive and negative sows. Our study provides new insight into producing cloned transgenic livestock related to reproductive traits.

      • The GSTT1 Null Genotype Contributes to Increased Risk of Prostate Cancer in Asians: a Meta-analysis

        Background: Many studies have investigated the association between glutathione S-transferase T 1 (GSTT1) null genotype and risk of prostate cancer, but the impact of GSTT1 null genotype in Asians is still unclear owing to inconsistencies across results. Thie present meta-analysis aimed to quantify the strength of the association between GSTT1 null genotype and risk of prostate cancer. Methods: We searched the PubMed, Embase and Wangfang databases for studies of associations between the GSTT1 null genotype and risk of prostate cancer in Asians and estimated summary odds ratio (OR) with their 95% confidence interval (95% CI). Results: A total of 11 case-control studies with 3,118 subjects were included in this meta-analysis, which showed the GSTT1 null genotype to be significantly associated with increased risk of prostate cancer in Asians (random-effects OR = 1.49, 95% CI 1.15-1.92, P = 0.002), also after adjustment for heterogeneity (fixed-effects OR = 1.45, 95% CI 1.23-1.70, P < 0.001). No evidence of publication bias was observed. Conclusions: This meta-analysis of available data suggested the GSTT1 null genotype does contribute to increased risk of prostate cancer in Asians.

      • Functional Investigation on Aromatase in Endometrial Hyperplasia in Polycystic Ovary Syndrome Cases

        Objective: To explore the possible significance of aromatase P450 in endometrial hyperplasia with a background of polycystic ovary syndrome (PCOS). Methods: Immunohistochemistry was used to determine the expression of aromatase P450 in endometrium of PCOS patients. Semiquantitative analysis of aromatase P450 expression of mRNA and protein level wasalso carried out by real-time quantitative RT-PCR method. After endometrial cells were stimulated by testosterone and letrozole in vitro, the estradiol ($E_2$) level was determined, and the expression of cell aromatase P450 mRNA was assessed. Results: The aromatase P450 mRNA level was increased in endometria of PCOS patients. When endometrial cells were cultured with $10^{-6}M$ testosterone, the $E_2$ level in the culture medium increased. An inhibitory effect on $E_2$ generation and expression of aromatase P450 mRNA was observed when the endometrial cells were treated with $10^{-5}M$ letrozole. Conclusions: There is an increased expression of aromatase P450 in PCOS patient endometrium. Androgen stimulation could enhance the synthesis of aromatase P450 mRNA and the production of $E_2$ in endometrial cells in vitro while letrozole could do the reverse.

        COUNTING OF FLOWERS BASED ON K-MEANS CLUSTERING AND WATERSHED SEGMENTATION

        This paper proposes a hybrid algorithm combining K-means clustering and wa- tershed algorithms for flower segmentation and counting. We use the K-means clustering al- gorithm to obtain the main colors in a complex background according to the cluster centers and then take a color space transformation to extract pixel values for the hue, saturation, and value of flower color. Next, we apply the threshold segmentation technique to segment flowers precisely and obtain the binary image of flowers. Based on this, we take the Euclidean dis- tance transformation to obtain the distance map and apply it to find the local maxima of the connected components. Afterward, the proposed algorithm adaptively determines a minimum distance between each peak and apply it to label connected components using the watershed segmentation with eight-connectivity. On a dataset of 30 images, the test results reveal that the proposed method is more efficient and precise for the counting of overlapped flowers ignoring the degree of overlap, number of overlap, and relatively irregular shape.

        Selecting the Optimal Hidden Layer of Extreme Learning Machine Using Multiple Kernel Learning

        Extreme learning machine (ELM) is emerging as a powerful machine learning method in a variety of application scenarios due to its promising advantages of high accuracy, fast learning speed and easy of implementation. However, how to select the optimal hidden layer of ELM is still an open question in the ELM community. Basically, the number of hidden layer nodes is a sensitive hyperparameter that significantly affects the performance of ELM. To address this challenging problem, we propose to adopt multiple kernel learning (MKL) to design a multi-hidden-layer-kernel ELM (MHLK-ELM). Specifically, we first integrate kernel functions with random feature mapping of ELM to design a hidden-layer-kernel ELM (HLK-ELM), which serves as the base of MHLK-ELM. Then, we utilize the MKL method to propose two versions of MHLK-ELMs, called sparse and non-sparse MHLK-ELMs. Both two types of MHLK-ELMs can effectively find out the optimal linear combination of multiple HLK-ELMs for different classification and regression problems. Experimental results on seven data sets, among which three data sets are relevant to classification and four ones are relevant to regression, demonstrate that the proposed MHLK-ELM achieves superior performance compared with conventional ELM and basic HLK-ELM.

        Effect of welding residual stress on operating stress of nuclear turbine low pressure rotor

        The purpose of this study is to investigate the effect of welding residual stress on operating stress in designing a nuclear turbine welded rotor. A two-dimensional axisymmetric finite element model is employed to calculate the residual stress before and after post weld heat treatment (PWHT), and then the superposition of residual stress after PWHT and operating stress at normal speed and overspeed were discussed. The investigated results show that operating stress can be affected significantly by welding residual stress, and the distribution trend of superposition stress at the weld area is mainly determined by welding residual stress. The superposition of residual stress and operating stress is linear superposition, and the hoop stress distribution of superposition stress is similar with the distribution of residual stress. With the increasing overspeed, the distribution pattern of the hoop superimposed stress remains almost unchanged, while the stress level increases

        Examining the Economic Effects of Logistics Infrastructure: The Case of New Western Land-Sea Corridor in China

        Purpose: To examine the economic effects of logistics under the influence of policies. Research design, data and methodology: This study is the first to use the panel data of 31 provinces and municipalities in China from 2012 to 2021, and use the OLS and DID models to evaluate whether the New Western Land-Sea Corridor (NWLSC) has promoted the economic development of the regions along the corridor. Results: The NWLSC has stimulated local economic growth by promoting the development of transportation, postal, and telecommunications industries along the corridor. Further, considering the locational differences of the regions along the NWLSC, we examined the differences in economic effects between regions along the Yangtze River and those not along the Yangtze River under the background of NWLSC implementation. We found that waterway and airway transport located along the NWLSC and in the Yangtze River Economic Belt (YREB) region can significantly promote economic growth. However, for regions located along the NWLSC but not in the YREB region, the impact of roadway, railway, and airway transport in these regions on economic growth is more significant. Conclusions: This study has important reference value on how to use logistics to promote the economic and cross-border commerce development of landlocked countries or regions.
