        Depletion of Janus kinase-2 promotes neuronal differentiation of mouse embryonic stem cells

        Janus kinase 2 (JAK2), a non-receptor tyrosine kinase, is a criticalcomponent of cytokine and growth factor signaling pathwaysregulating hematopoietic cell proliferation. JAK2 mutations areassociated with multiple myeloproliferative neoplasms. Althoughphysiological and pathological functions of JAK2 in hematopoietictissues are well-known, such functions of JAK2 in thenervous system are not well studied yet. The present study demonstratedthat JAK2 could negatively regulate neuronal differentiationof mouse embryonic stem cells (ESCs). Depletion ofJAK2 stimulated neuronal differentiation of mouse ESCs andactivated glycogen synthase kinase 3ꞵ, Fyn, and cyclin-dependentkinase 5. Knockdown of JAK2 resulted in accumulation of GTPboundRac1, a Rho GTPase implicated in the regulation of cytoskeletaldynamics. These findings suggest that JAK2 might negativelyregulate neuronal differentiation by suppressing the GSK-3β/Fyn/CDK5 signaling pathway responsible for morphological maturation.

        일본의 불임치료와 일의 양립지원정책에 관한 연구

        일본은 2012년부터 합계출산율이 상승하였으나 2019년 1.36을 기점으로 다시 하락 추세를 보인다. 이로 인해 만혼화 등을 배경으로 지금까지 불임 치료를 받고 태어난 아이가 해마다 증가하고 있다는 점에 관심을 두게 되었다. 유럽 등의 선진국에서 저출산 극복을 위해 시작된 불임 지원사업은 전 세계적으로 시행되고 있는 국가 수준의 정책 중 하나이다. 이러한 관점에서 일본의 불임 치료지원정책에 대해 고찰하였다. 연구 결과 불임 치료지원을 받기 위한 나이 제한, 소득수준, 혼인 관계 등에 규제가 많았다. 특히 불임치료 중 고액의 비용부담 외 추가로 발생하는 비용도 많았다. 또한, 직장 내 환경이 불임치료에 대한 지식과 이해가 미흡하여 불임 치료와 일의 양립이 어려운 현실이다. 일본은 이러한 문제점을 해결하기 위해 공적 지원 확대 방안을 마련하고 있다. 이와같은 일본의 불임 치료지원에서 나타나는 문제점과 대책 마련 과정이 최근 불임지원정책에 관심을 두는 한국에도 여러 시사점이 될 것이라 기대한다. In Japan, the total fertility rate has risen since 2012, but it has been on the decline again since 1.36 in 2019. As a result, attention has been paid to the fact that the number of children born after infertility treatment is increasing year by year against the backdrop of late marriage. The fertility support project, which was initiated to overcome low birth rates in developed countries such as Europe, is one of the national-level policies being implemented worldwide. From this point of view, Japan"s infertility treatment support policy was considered. As a result of the study, there were many regulations on age restrictions, income levels, and marital relationships to receive support for infertility treatment. In particular, there were many additional costs incurred during infertility treatment other than the high cost burden. In addition, the environment in the workplace lacks knowledge and understanding of infertility treatment, making it difficult to balance work with infertility treatment. Japan is preparing measures to expand public support to solve these problems. I think the problems that appear in infertility treatment support and the process of preparing countermeasures will have many implications for Korea.

        Recombinant Human Epidermal Growth Factor (rhEGF) Protects Radiation-Induced Intestine Injury in Murine System

        <P>This study was to investigate whether rhEGF protects radiation induced intestine injury without compromising antitumor effect of radiation in murine system. A radiation induced intestinal injury model was established in mice by whole body irradiation. Using this model, 4 groups were set; control, rhEGF (100 μg/kg intraperitoneally), radiation (10 Gy), and a combination (rhEGF and radiation). The level of apoptosis and proliferation were analyzed by TUNEL assay and proliferation cell nuclear antigen (PCNA) immunohistochemical staining, respectively, as well as observation of survival and body weight change. A tumor growth delay assay was performed using murine syngeneic tumors; one radioresistant tumor, HCa-I and one radiosensitive tumor, MCa-K. In the radiation induced intestinal injury model, the 10 Gy group had significantly more weight loss with less number of crypt cells and higher apoptosis than the 8 Gy group. Using 10 Gy model, radioprotective effect of rhEGF was tested. Addition of rhEGF improved not only the body weight loss but also survival following radiation. It also induced suppression of apoptosis as well as increase of PCNA expression and recovery of villi. rhEGF did not enhance the tumor growth after radiation exposure in the tested tumors. These findings suggest that combination of exogenous rhEGF and radiation can be a new anticancer strategy by protecting radiation-induced intestinal injury without alleviating antitumor effect of radiation.</P>

      • CNS Cholinergic Innervation to the Hippocampus in the Rat Using pseudorabies Virus as a Neurotracer

        The hippocampus is a central area of the memory-related neural system. Combined irnmunohistochemistry against choline acetyl transferase and retrograde transneuronal labelling of the pseudorabies virus were used to identify cholinergic neurons in the central nervous system projecting to the hippocampal formation of the rat. Five to ten μL of Bartha strain of pseudorabies virus were injected into the dentate gyrus, CA1 and CA3 of the hippocampus of 20 Sprague Dawley rats using stereotaxic instrument. Forty eight to 96 hr after the injection, the brains were removed and the tissue sections were processed for double immunofluorescence procedure using polyclonal antibodies against pseudorabies virus or choline acetyl transferase. The double labelled neurons were distributed at several different nuclei and the labelling patterns of three different areas of the hippocampus were similar. These data suggests that the cholinergic innervation to the hippocampus were distributed in a transsynaptic manner throughout the whole brain area.

        토론 기반 대학 융합교양교육 설계 및 운영 전략 탐색 : S대학 융합 주제 교양교과목 사례를 중심으로

        본 연구는 토론 기반 대학 융합교양교육을 효과적으로 설계하고 운영하는 전략을 탐색하는 것을 목적으로 한다. 이를 위해 인문·사회계열, 이공계열 교수들이 참여하여 교과목 차원에서의 융합교육과정을 구성 및 운영한 S대학교의 토론 기반 융합교양교과목 4개의 교육과정을 분석하고, 담당 교수와 조교를 면담하는 질적 사례 연구를 수행하였다. 반복적 비교분석법을 활용하여 개방 코딩과 범주화 과정을 거친 연구결과는 다음과 같다. 첫째, 토론 기반 융합교양교육의 목표는 융합적 사고력을 육성하는 것이다. 이를 위해 교수자들은 주제 중심으로 학문 내용을 화학적으로 결합한 교육 모듈 뿐만 아니라 각 학문별 이슈를 연결하여 토론하는 종합토론을 설계하고 운영하였다. 둘째, 교육 효과를 높이기 위해 교수, 조교, 학부생 세 집단의 의견을 수렴하여 교육과정을 조정했다. 셋째, 토론 파트를 효과적으로 운영하기 위해 하나의 관점으로는 해결할 수 없는 문제 상황을 토론 주제로 선정하였으며, 토론 퍼실리테이터로서 조교의 역량을 강화했다. 또한 배경 정보가 다양한 구성원들이 참여하는 방식으로 토론 그룹을 구성하고, 학습자들이 자유롭게 의견을 개진할 수 있도록 개방적인 분위기를 형성하는 전략을 사용했다. 동일한 이슈에 대하여 서로 다른 전공 배경을 가진 학습자들이 각자 자신의 전공학문의 관점을 소개하도록 함으로써 다양한 관점이 공존할 수 있음을 인지하도록 했으며, 교수자들은 직접 융합연구자들이 효과적으로 소통하는 방식을 보여주기도 했다. 이를 통해 학습자들은 융합적 사고력 및 소통력을 증진할 수 있었고, 교수자들의 융합 마인드도 배양되었다. 마지막으로 연구자들은 바람직한 방향으로 대학 융합교육이 확산되도록 하기 위해서 교수자들의 융합역량을 확보할 수 있도록 제도적 지원이 필요함을 제안하였다. This study aims to explore strategies for designing and operating effective discussion-based college interdisciplinary liberal education. Thereby, a qualitative case study was conducted by analyzing the curricula of four discussion-based interdisciplinary liberal courses at S University, where professors of humanities, social sciences, and science chemically combine the contents of each discipline, and interviewing the professors and teaching assistants. The results of the open coding and categorization process using the iterative comparative analysis method are presented as follows. The goal of discussion-based interdisciplinary liberal education is to foster convergent thinking. To effectively foster convergent thinking, instructors designed education modules that chemically combined the contents of the disciplines around a theme, and comprehensive discussion section. In addition, the curriculum was not implemented as initially designed, but was finalized by making adjustments based on the opinions of three groups: professors, teaching assistants, and students. To foster convergent thinking, professors selected problematic situations that could not be solved from a single point of view as discussion topics and strengthened the capabilities of teaching assistants as discussion facilitators. The instructors also organized discussion groups with diverse members and respected their opinions, thus creating an open atmosphere. By introducing different disciplinary perspectives on the same issue, the students could become aware of the coexistence of different viewpoints, and the instructors were shown debating to demonstrate how convergence researchers communicate effectively. This helped learners improve their convergent thinking and communication skills, and also led to a change in the way instructors recognized the importance of the convergent mindset. In future, institutional support is needed to secure the convergent capabilities of instructors.

      • Possible Role of Phosphoinositide-3-Kinase in Mx1 Protein Translation and Antiviral Activity of Interferon-Omega-Stimulated HeLa Cells

        <P>Interferon ω (IFN-ω), a cytokine released during innate immune activation, is well known for promoting direct antiviral responses; however, the possible signal pathways that are initiated by IFN-ω binding to the type I IFN receptors have not been fully studied. Here, we provide evidence that activation of phosphoinositide-3-kinase/protein kinase B (PI3K/Akt) signaling plays a pivotal role in the generation of IFN-ω-mediated biological responses. We found that LY294002 (PI3K inhibitor)-attenuated antiviral activities are induced by IFN-ω treatment. Although such effects of LY294002 are unrelated to regulatory activities on IFN-ω-dependent Mx1 (myxovirus resistance 1) or Mx2 gene transcriptional regulation, translation of Mx1 protein, which was known as a key mediator of cell-autonomous antiviral resistance, was significantly reduced by PI3K inhibition. Further studies showed that PI3K inhibition using LY294002 leads to a decrease in PI3K substrate Akt and mitogen-activated protein kinase extracellular signal-regulated kinase and p38 phosphorylation/activation. In addition, although LY294002 was not able to reduce STAT1 activation, we found that the mammalian target of rapamycin (mTOR)/p70 S6 kinase pathway was significantly attenuated by inhibition of the PI3K/Akt signaling pathway. These results indicate that the PI3K/Akt pathway is a common and central integrator for antiviral responses in IFN-ω signaling via its regulatory effects on mTOR that are required for initiation of mRNA translation of Mx genes.</P><P>Copyright © 2011 S.
