      • Haplotype and mutation analysis for newly suggested Y-STRs in Korean father-son pairs

        In this study, 363 Korean father-son haplotype transfers in 351 families were analyzed using an in-house multiplex PCR system for 14 Y-STRs (DYS385a/b, DYF387S1, DYS391, DYS449, DYS460, DYS481, DYS518, DYS533, DYS549, DYS570, DYS576, DYS627 and DYS643), that included 11 loci newly added to the PowerPlex Y23 system or the Yfiler Plus system. The Y-STRs showed gene diversity values ranging from 0.2499 to 0.9612; the multicopy Y-STR loci DYS385 and DYF387S1 had high gene diversity of 0.9612 and 0.9457, respectively. In addition, DYF387S1, which has two copies, showed three alleles in seven individuals, and micro-variant alleles were observed in 14 individuals at four loci (DYS448, DYS518, DYS570 and DYS627). Among 351 haplotypes for the 11 newly added Y-STRs, 350 different haplotypes were observed, with an overall haplotype diversity of 0.9999 and discrimination capacity of 99.72%. In 363 haplotype transfers from 351 pedigrees, 29 single-step mutations were observed at 11 Y-STRs. Locus-specific mutation rate estimates varied from 0.0 to 1.93x10<SUP>-2</SUP>, with an average estimated mutation rate of 6.66x10<SUP>-3</SUP>. Two father-son pairs had mutations at two different loci in 11 Y-STRs. The number of pairs with mutations at multiple loci increased to five when the mutation event was investigated for haplotype transfer at 28 Y-STRs including 17 Yfiler loci and 11 Y-STRs examined in this study: four father-son pairs had mutations at two loci, and one pair had mutations at three loci. Overall, mutations were frequently observed at DYS449, DYS576 and DYS627 loci, which are known to be rapidly mutating Y-STRs. Mutation rate estimates at most loci were not significantly different from rates in other populations, but estimates for DYF387S1, DYS518 and DYS570 were considerably lower in the Korean population than in other populations.

© 2010 John Wiley & Sons A/S.</P><P><B>Background – </B> It has not been clarified whether the disparity in ischemic stroke outcome between populations is caused by ethnic and geographic differences or by variations in case mix. Propensity score matching (PSM) analysis can overcome some analytical problems but is rarely used in stroke outcome research. This study was to compare the ischemic stroke case‐fatality between two PSM cohorts of Sweden and Korea.</P><P><B>Methods – </B> Prognostic variables related to baseline characteristics and stroke care were included in our PSM model. Then, we selected 7675 Swedish and 1220 Korean patients with ischemic stroke from each stroke registers and performed one‐to‐one matching based on propensity scores of each patient.</P><P><B>Results – </B> After PSM, all measured variables were well balanced in 1163 matched subjects, and the 90‐day case‐fatality was identical 6.2% (HR 0.997, 95%CI 0.905–1.099) in Sweden and Korea.</P><P><B>Conclusions – </B> No difference is found in the 90‐day case‐fatality in propensity score‐matched Swedish and Korean patients with ischemic stroke.</P>

        Overview of KSTAR research progress and future plans toward ITER and K-DEMO

        <P>A decade-long operation of the Korean Superconducting Tokamak Advanced Research (KSTAR) has contributed significantly to the operation of superconducting tokamak devices and the advancement of tokamak physics which will be beneficial for the ITER and K-DEMO programs. Even with limited heating capability, various conventional as well as new operating regimes have been explored and have achieved improved performance. As examples, a long pulse high-confinement mode operation with and without an edge-localized mode (ELM) crash was well over 70 and 30 s, respectively. The unique capabilities of KSTAR allowed it to improve the capability of controlling harmful instabilities, and they have been instrumental in uncovering much new physics. The highlights are that the L/H transition threshold power is sensitive to the resonant magnetic perturbation (RMP) and insensitive to non-resonant magnetic perturbation. Co-<I>I</I> <SUB>p</SUB> offset rotation dominated by an electron channel predicted by general neoclassical toroidal viscosity theory was confirmed. Improved heat dispersal in a divertor system using three rows of rotating RMP was demonstrated and predictive control of the ELM-crash with <I>a priori</I> modeling was successfully tested. In magnetohydrodynamic physics, validation of the full reconnection model (i.e. <I>q</I> <SUB>0</SUB>  >  1 right after the sawtooth crash) and self-consistent validation of the anisotropic distribution of turbulence amplitude and flow in the presence of the 2/1 island with theoretical models were achieved. The turbulence amplitude induced by RMP was linearly increased with the slow RMP coil current ramp-up time (i.e. the magnetic diffusion time scale). The <I>D</I> <SUB> <I>α</I> </SUB> spikes (i.e. ELM-crash amplitude) was linearly decreased with the turbulence amplitude and not correlated with the perpendicular electron flow. In the turbulence area, a non-diffusive ‘avalanche’ transport event and the role of a quiescent coherent mode in confinement were studied. To accommodate the anticipation of a higher performance of the KSTAR plasmas with the increased heating powers, a new divertor/internal interface with a full active cooling system will be implemented after a full test of the new heating (neutral beam injection II and electron cyclotron heating) and current drive (CD) (Helicon and lower hybrid CD) systems. An upgrade plan for the internal hardware, heating systems and efficient CD system may allow for a long pulse operation of higher performance plasmas at <I>β</I> <SUB>N</SUB>  >  3.0 with <I>f</I> <SUB>bs</SUB> ~ 0.5 and <I>T</I> <SUB>i</SUB>  >  10 keV.</P>

      • A multiplex PCR system for 13 RM Y-STRs with separate amplification of two different repeat motif structures in DYF403S1a

        <P>In forensic science and human genetics, Y-chromosomal short tandem repeats (Y-STRs) have been used as very useful markers. Recently, more Y-STR markers have been analyzed to enhance the resolution power in haplotype analysis, and 13 rapidly mutating (RM) Y-STRs have been suggested as revolutionary tools that can widen Y-chromosomal application from paternal lineage differentiation to male individualization. We have constructed two multiplex PCR sets for the amplification of 13 RM Y-STRs, which yield small-sized amplicons (<400 bp) and a more balanced PCR efficiency with minimum PCR cycling. In particular, with the developed multiplex PCR system, we could separate three copies of DYF403S1a into two copies of DYF403S1a and one of DYF403S1b1. This is because DYF403S1b1 possesses distinguishable sequences from DYF403S1a at both the front and rear flanking regions of the repeat motif; therefore, the locus could be separately amplified using sequence-specific primers. In addition, the other copy, defined as DYF403S1b by Ballantyne et al., was renamed DYF403S1b2 because of its similar flanking region sequence to DYF403S1b1. By redefining DYF403S1 with the developed multiplex system, all genotypes of four copies could be successfully typed and more diverse haplotypes were obtained. We analyzed haplotype distributions in 705 Korean males based on four different Y-STR subsets: Yfiler, PowerPlex Y23, Yfiler Plus, and RM Y-STRs.       • Eugenol Inhibits K<sup>+</sup> Currents in Trigeminal Ganglion Neurons

        <P>Eugenol, a natural capsaicin congener, is widely used in dentistry. Eugenol inhibits voltage-activated Na<SUP>+</SUP> and Ca<SUP>2+</SUP> channels in a transient receptor potential vanilloid 1 (TRPV1)-independent manner. We hypothesized that eugenol also inhibits voltage-gated K<SUP>+</SUP> currents, and investigated this in rat trigeminal ganglion neurons and in a heterologous system using whole-cell patch clamping. Eugenol inhibited voltage-gated K<SUP>+</SUP> currents, and the inhibitory effects of eugenol were observed in both capsaicin-sensitive and capsaicin-insensitive neurons. Pre-treatment with capsazepine, a well-known antagonist of TRPV1, failed to block the inhibitory effects of eugenol on K<SUP>+</SUP> currents, suggesting no involvement of TRPV1. Eugenol inhibited human Kv1.5 currents stably expressed in <I>Ltk</I><SUP>−</SUP> cells, where TRPV1 is not endogenously expressed. We conclude that eugenol inhibits voltage-gated K<SUP>+</SUP> currents in a TRPV1-independent manner. The inhibition of voltage-gated K<SUP>+</SUP> currents is likely to contribute to the irritable action of eugenol.       • Enhanced photoluminescence due to Bi<sup>3+</sup> → Eu<sup>3+</sup> energy transfer and re-precipitation of RE doped homogeneous sized Y<sub>2</sub>O<sub>3</sub> nanophosphors

        Precipitation and re-precipitation of metal ions has been carried out from original and supernatant solution producing Bi<SUP>3+</SUP> - Eu<SUP>3+</SUP> doped Y<SUB>2</SUB>O<SUB>3</SUB>:Eu<SUP>3+</SUP> and Eu<SUP>3+</SUP>/Tb<SUP>3+</SUP> doped Y<SUB>2</SUB>O<SUB>3</SUB>, respectively. Shorter reaction time is unable to consume all metal ions present in the solution which can be utilized through re-precipitation process. The doping of Bi<SUP>3+</SUP> - Eu<SUP>3+</SUP> in Y<SUB>2</SUB>O<SUB>3</SUB> helps to absorb maximum UV light. The activation of Y<SUB>2</SUB>O<SUB>3</SUB> matrix by Bi<SUP>3+</SUP> and Eu<SUP>3+</SUP> ions, together and separately, were studied considering the excitation energy transfer to the luminescence centers. The successful replacement of Y<SUP>3+</SUP> by RE<SUP>3+</SUP> ions can help for fine tuning of emission wavelength. Re-precipitation of supernatant solution by adding terbium precursor can successively produce uniform sized Tb<SUP>3+</SUP> doped Y<SUB>2</SUB>O<SUB>3</SUB>:Eu<SUP>3+</SUP>. The re-precipitation of the supernatant solution ensures maximum consumption of metal ions for higher product yield and possible fine tuning of emission wavelengths.
