MiR-449a attenuates autophagy of T-cell lymphoma cells by downregulating ATG4B expression

Increasing evidence suggests the role of miR-449a in the regulation of tumorigenesis and autophagy. Autophagy plays an important role in the malignancy of T-cell lymphoma. However, it is still unknown whether miR-449a is associated with autophagy to regulate the malignancy of T-cell lymp homa. In this study, we for the first time demonstrated that miR-449a enhanced apoptosis of T-cell lymphoma cells by decreasing the degree of autophagy. Further, miR-449a downregulated autophagy- associated 4B (ATG4B) expression, which subsequently reduced the autophagy of T-cell lymphoma cells. Mechanistically, miR-449a decreased ATG4B protein level by binding to its mRNA 3’UTR, thus reducing the mRNA stability. In addition, studies with nude mice showed that miR-449a significantly inhibited lymphoma characteristics in vivo. In conclusion, our results demonstrated that the “miR-449a/ATG4B/autophagy” pathway played a vital role in the malignancy of T-cell lymphoma, suggesting a novel therapeutic target.

Chemosensory system plays important roles in insect behaviors, some key genes governing chemosensation can potentially be used as novel targets for pest control. Nysius ericae (Hemiptera: Lygaeidae) is a polyphagous insect pest found throughout Eurasia countries which causes damage to many important crops. Little is known about the chemosensory mechanisms of host location and mate choice in N. ericae at themolecular level. Here, we assembled the antennal transcriptome of N. ericae using Illumina sequencing technology. Analysis of the transcriptome data identified 148 candidate chemosensory genes, including 28 for odorant binding proteins (OBPs), 16 for chemosensory proteins (CSPs), 2 for sensory neuron membrane proteins (SNMPs), 83 odorant receptors (ORs), 12 ionotropic receptors (IRs), and 7 gustatory receptors (GRs). Our results greatly improve the gene inventory for N. ericae and provide valuable resources for analyzing the functions of some key genes. This information will aid in the development of better understanding of chemosensation in N. ericae, and indicate novel targets for managing its impact on agriculture. The m6A reader IGF2BP3 promotes acute myeloid leukemia progression by enhancing RCC2 stability

N6-methyladenosine (m6A) is the most abundant posttranscriptional modification of mRNA in eukaryotes. Recent evidence suggests that dysregulated m6A-associated proteins and m6A modifications play a pivotal role in the initiation and progression of diseases such as cancer. Here, we identified that IGF2BP3 is specifically overexpressed in acute myeloid leukemia (AML), a subtype of leukemia associated with poor prognosis and high genetic risk. IGF2BP3 is required for maintaining AML cell survival in an m6A-dependent manner, and knockdown of IGF2BP3 dramatically suppresses the apoptosis, reduces the proliferation, and impairs the leukemic capacity of AML cells in vitro and in vivo. Mechanistically, IGF2BP3 interacts with RCC2 mRNA and stabilizes the expression of m6A-modified RNA. Thus, we provided compelling evidence demonstrating that the m6A reader IGF2BP3 contributes to tumorigenesis and poor prognosis in AML and can serve as a target for the development of cancer therapeutics.
