Estudio sobre el caracter de egloga en La Galatea de Cervantes
( Nakwon Choi ) 한국스페인어문학회(구 한국서어서문학회) 2016 스페인어문학 Vol.0 No.81
In the “Prologue” of La Galatea, Cervantes classifies the style of the work not as a plot-oriented novel but rather as an eclogue. Therefore, it is very important to analyze the characteristic of an eclogue that Cervantes identifies as his work`s genre. Cervantes esteemed writing poetry as a primary type of creative activity. Hence, he wished to write his work in the form of a poem, especially an eclogue which had a wide range of contemporaneous readership. The eclogue was a part of the typical classical genre, and it had a flexibility to incorporate other genres including prose. Many remarkable writers chose and wrote this genre as a part of their literary canon. Among these writers, Cervantes mainly modeled himself after Garcilaso de la Vega who had taken the Italian Renaissance poetry and transformed it into his own unique style. However, because of his lack of poetic talent, Cervantes could not write an authentic eclogue like the works of Garcilaso. Cervantes was a novelist, not a poet. As such, his genius as a novelist is amply displayed in his first novel. Betrayal, hatred, murder, et cetera: these negative elements that were hitherto not permitted in traditional pastoral novels add strong realism to Cervantes`s work. Hence La Galatea possesses dualism. The poetic part delivers the morals and the philosophical messages that the author wanted to impart. The prose, with its characteristic realism, brings amusement, vitality, and reality to those messages.

Abstract

Quantitative polymerase chain reaction (qPCR) renders profiling of genes of interest less time-consuming and cost-effective. Recently, multiplex profiling of miRNAs has enabled identifying or investigating predominant miRNAs for various diseases such as cancers and neurodegenerative diseases. Conventional multiplex qPCR technologies mostly use colorimetric measurements in solution phase, yet not only suffer from limited multiplexing capacity but also require target-screening processes due to non-specific binding between targets and primers. Here, we present hydrogel micropost-based qPCR for multiplex detection of miRNAs associated with Alzheimer's disease (AD). Our methodology promises two key advantages compared with the conventional solution-based PCR: 1) nearly no non-specific crosstalks between targets and primers, and 2) practically valuable multiplexing by spatial encoding within a single microchamber. Specifically, we immobilized hydrogel microposts (~ 400µm in diameter) within commercially available polycarbonate PCR chips by multi-step ultraviolet (UV, 365nm) exposure. We optimized this photoimmobilization for thermal cycles of PCR as well. Acrylated forward primers incorporated in polyethylene glycol diacrylate (PEGDA) posts played a crucial role to confine fluorescent signal of cDNA amplification within the PEGDA hydrogel. To demonstrate the potential of our platform, we successfully verified multiplex detection of five miRNAs, which were reported to be highly correlated with AD, from a complex buffer of human plasma.

Highlights

Hydrogel micropost-based multiplex qPCR is reported for detection of miRNAs.

Multi-step UV-exposure allows for immobilization of PEGDA microposts in PCR chips.

Hydrogel-based qPCR results in the significant elimination of non-specific binding.

5-plex detection of miRNAs is presented with excellent specificity & selectivity.

Graphical abstract

[DISPLAY OMISSION]

Most of a driving cycle is used to measure fuel consumption (FC) and emissions for a specified vehicle. A driving cycle was reflected geography and traffic characteristics for each country, also, driving characteristic is affected these parameters such as vehicle dynamics, FC and emissions. Therefore, this study is an attempt to develop a driving cycle for military operational area. The methodology is that the driving cycle is constructed considering important parameters to be affected FC. Therefore, this approach is expected to be a better representation of heterogeneous traffic behavior. The driving cycle is constructed using the proposed methodology for smallest RMSE and information value.

In this study, experimental and numerical investigation for diesel spray evaporation and combustion was carried out in a constant volume combustion chamber(CVCC) on various ambient gas pressure and temperature. In the experiment, the spray was injected into CVCC by common rail system and then images of the spray were acquired by high speed camera and the calculation study was implemented under same condition with experiment by using AVL-Fire. There was a good agreement between experimental and computational results such as spray penetration, ignition delay.
