Purpose: The clinical interpretation of children sensitized to allergens is challenging, particularly in children with food allergies. We aimed to examine clinical differences between children with monosensitization and those with polysensitization to common food allergens and to determine risk factors for polysensitization in young children <10 years of age with immediate-type food allergies. Methods: The study included children <10 years of age with signs and symptoms indicative of immediate-type food allergies. Serum total IgE level was measured, and ImmunoCAP analysis for food allergens was performed. Results: The mean age of the study subjects was $1.6{\pm}1.6$ years (75 boys and 51 girls). Thirty-eight children (30.2%) were monosensitized and 88 children (69.8%) were polysensitized. Multivariate logistic regression analysis showed that the development of polysensitization to common food allergens was positively associated with a parental history of allergic rhinitis (adjusted odds ratio [aOR], 6.28; 95% confidence interval [CI], 1.78-22.13; P=0.004), season of birth (summer/fall) (aOR, 3.10; 95% CI, 1.10-8.79; P=0.033), and exclusive breastfeeding in the first 6 months of age (aOR, 3.51; 95% CI, 1.20-10.25; P=0.022). Conclusion: We found significant clinical differences between children with monosensitization and those with polysensitization to common food allergens and identified risk factors for the development of polysensitization in young children with immediate-type food allergies. Clinicians should consider these clinical risk factors when evaluating, counseling, treating, and monitoring young children with food allergies.

We investigated the antioxidant and cholinesterase inhibitory activities of the aqueous extract of rainbow trout Oncorhynchus mykiss. The antioxidant activity of O. mykiss aqueous extract was determined by in vitro peroxynitrite scavenging activity and reducing power assays. The aqueous extract of O. mykiss showed potent peroxynitrite radical scavenging activity ($IC_{50}=0.12{\pm}0.001\;mg/mL$) and reducing power (absorbance=$0.47{\pm}0.001$) at the concentration of 1 mg/mL. The in vitro cholinesterase inhibitory activity of O. mykiss aqueous extract was examined using spectrophotometric analyses of acetyl- and butyrylcholinesterase. The aqueous extract of O. mykiss showed acetylcholinesterase inhibitory activity ($IC_{50}=1.61{\pm}0.13\;mg/mL$), but did not exhibit inhibitory activity against butyrylcholinesterase. These results suggest that O. mykiss possesses antioxidant and acetylcholinesterase inhibitory activities and provide scientific evidence for the health benefits of O. mykiss aqueous extract.
Prognostic factors for classifying extranodal NK/T cell lymphoma, nasal type, as lymphoid neoplasia

<P>Abstract</P><P>This study evaluated the applicability of prognostic factors commonly used for diagnosis of classical lymphoma outcomes to extranodal NK/T cell lymphoma, nasal type (NTCL). Clinical features and their associations with lactate dehydrogenase (LDH) were evaluated in 70 patients. RLDH was defined as the ratio of LDH to the upper normal limit. RLDH was associated with stage (I–II vs. III–IV), lymph node involvement (LNI), and International Prognostic Index score (<2 vs. ≥2). Poor performance status and advanced stage were common in patients with local tumor invasiveness (LTI). LDH level, classified into three levels (low, high, and very high) was associated with survival (<I>P</I> < 0.001). In multivariate analysis, the predictive values of LDH level, B symptom, performance status, and stage remained significant whereas those of LTI and LNI did not. Scoring was performed by weighting each factor with 0.5 or 1.0 according to its hazard ratio. Scores were classified into four groups. Groups with high scores were associated with unfavorable outcomes (<I>P</I> < 0.001). Current study suggests that prognostic factors for NHL may be useful to predict the outcome of NTCL but the model should take LDH level and the prognostic weight of each factor into account.</P>
Effects of Culture Dimensions on Maintenance of Porcine Inner Cell Mass-Derived Cell Self-Renewal

Despite the fact that porcine embryonic stem cells (ESCs) are a practical study tool, in vitro long-term maintenance of these cells is difficult in a two-dimensional (2D) microenvironment using cellular niche or extracellular matrix proteins. However, a three-dimensional (3D) microenvironment, similar to that enclosing the inner cell mass of the blastocyst, may improve in vitro maintenance of self-renewal. Accordingly, as a first step toward constructing a 3D microenvironment optimized to maintain porcine ESC self-renewal, we investigated different culture dimensions for porcine ICM-derived cells to enhance the maintenance of self-renewal. Porcine ICM-derived cells were cultured in agarose-based 3D hydrogel with self-renewal-friendly mechanics and in 2D culture plates with or without feeder cells. Subsequently, the effects of the 3D microenvironment on maintenance of self-renewal were identified by analyzing colony formation and morphology, alkaline phosphatase (AP) activity, and transcriptional and translational regulation of self-renewal-related genes. The 3D microenvironment using a 1.5% (w/v) agarose-based 3D hydrogel resulted in significantly more colonies with stereoscopic morphology, significantly improved AP activity, and increased protein expression of self-renewal-related genes compared to those in the 2D microenvironment. These results demonstrate that self-renewal of porcine ICM-derived cells can be maintained more effectively in a 3D microenvironment than in a 2D microenvironment. These results will help develop novel culture systems for ICM-derived cells derived from diverse species, which will contribute to stimulating basic and applicable studies related to ESCs.

<P><B>Abstract</B></P> <P>SIRT1, an NAD<SUP>+</SUP>-dependent histone/protein deacetylase, has diverse physiological actions. Recent studies have demonstrated that SIRT1 is overexpressed in colorectal cancer, suggesting its oncogenic potential. However, the molecular mechanisms by which overexpressed SIRT1 induces the progression of colorectal cancer and its inhibition remain largely unknown. Curcumin (diferuloymethane), a major component of the spice turmeric derived from the plant <I>Curcuma longa</I> L., has been reported to exert chemopreventive and anti-carcinogenic effects on colon carcinogenesis. In the present study, we found that curcumin reduced the expression of SIRT1 protein without influencing its mRNA expression in human colon cancer cells, suggesting posttranslational regulation of SIRT1 by this phytochemical. Notably, ubiquitination and subsequent proteasomal degradation of SIRT1 were induced by curcumin treatment. Results of nano-LC-ESI-MS/MS revealed the direct binding of curcumin to cysteine 67 of SIRT1. In line with this result, the protein stability and clonogenicity of a mutant SIRT1 in which cysteine 67 was substituted by alanine were unaffected by curcumin. Taken together, these observations suggest that curcumin facilitates the proteasomal degradation of oncogenic SIRT1 through covalent modification of SIRT1 at the cysteine 67 residue.</P> <P><B>Highlights</B></P> <P> <UL> <LI> SIRT1 is overexpressed in colorectal cancer tissues compared with normal colon tissues. </LI> <LI> SIRT1 knockdown attenuates viability and migration of human colon cancer cells. </LI> <LI> Silencing of SIRT1 suppressed the growth of HCT-116 derived tumor xenografts. </LI> <LI> Curcumin inhibits migration and growth of HCT-116 cells. </LI> <LI> Curcumin covalently modifies SIRT1 on the cysteine 67 residue, thereby stimulating its proteasomal degradation. </LI> </UL> </P>

<P><B>Abstract</B></P> <P>Leptin, a representative adipokine secreted from the white adipose tissue, is considered as a potential linker between obesity and cancer. SIRT1 is an NAD<SUP>+</SUP>-dependent histone/protein deacetylase speculated to function as an oncogene. In the present study, we found that leptin signaling-defective <I>ob/ob</I> and <I>db/db</I> mice had lower colonic expression of SIRT1 compared with leptin signaling-intact C57BL/6J mice, implying that leptin signaling is crucial for SIRT1 expression in vivo. Moreover, leptin induced up-regulation of SIRT1 in human colon cancer (HCT-116) cells. Leptin stimulated migration and invasion of cultured HCT-116 cells and tumor growth in the xenograft assay, and these effects were abrogated by a SIRT1 inhibitor sirtinol, suggesting that SIRT1 plays a role in leptin-induced colon carcinogenesis. Leptin-induced SIRT1 expression was regulated by the redox-sensitive transcription factor NF-E2-related factor 2 (Nrf2). Leptin stimulated nuclear accumulation of Nrf2 as well as its binding to the antioxidant response elements located in the <I>SIRT1</I> promoter. Moreover, siRNA knockdown of <I>Nrf2</I> abrogated the leptin-induced SIRT1 expression. Notably, SIRT1 was significantly reduced in colon tissues of <I>Nrf2</I>-null mice, lending further support to Nrf2-dependent SIRT1 expression. Expression of leptin, Nrf2 and SIRT1 was coordinately increased in human colon tumor tissues. In conclusion, leptin might play a role in colon carcinogenesis by inducing Nrf2-dependent SIRT1 overexpression.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

IT 기술과 더불어 전자상거래는 많은 발전을 하고 있다. 특히 포탈, 엔터테인먼트, 인터넷쇼핑몰 등은 IT 발전이 만들어낸 결과라고 볼 수 있다. 하지만 관련 업체들은 외형적 성장에도 불구하고 경쟁업체의 증가로 인한 경쟁의 격화 및 수익성 논란, 고객 이탈의 증가 등과 같은 부정적 요소들도 많이 부각되고 있다. 따라서 웹 사이트 운영에 있어서 환경변화에 신속히 대응하고 경쟁우위의 확보를 위한 다양한 충성고객 확보전략 및 연구가 필요하다. 이런 관점에서 본 연구는 관계마케팅 관점에서 웹사이트 특성이 고객가치, 고객만족 및 고객 충성도에 미치는 영향을 파악하였다. 연구결과 첫째, 인터넷 웹 사이트 특성이 고객가치와 고객만족에 유의한 영향을 미치는 것으로 나타났다. 둘째, 고객가치와 고객만족도, 고객충성도에 유의한 영향을 미치는 것으로 나타났다. 이는 고객만족 요인이 고객가치에 비해 고객충성도에 미치는 영향력이 큰 것을 의미한다. 셋째, 고객가치와 고객만족은 상호 영향을 미치는 것으로 나타났다. 넷째, 판매촉진은 웹 사이트 특성과 부수적 고객가치 사이에서만 조절역할을 하는 것으로 분석되었으나 웹 사이트 특성과 기본적 고객가치, 웹 사이트 특성과 고객만족 사이에서는 유의하지 않은 결과를 보였다. 따라서 웹 사이트 제공자들은 고객의 기대에 맞는 적절한 판매촉진 기법을 다각도로 연구해야 할 것이다. This study took the perspective of relationship marketing in investigating the impacts of the characteristics of a web site on customer value, satisfaction, and loyalty according to sales promotions of portals, entertainment sites, and Internet shopping malls. In addition, it aimed to examine the mutual influences between the characteristics and elements of sales promotion, as well as their impacts on customer value, satisfaction, and loyalty and to suggest an idea of what kind of factors were needed for ongoing customer creation and retention.
