        A Cooperative Use of Stochastic Dynamic Programming and Non-Linear Programming for Optimization of Reservoir Operation

        Stochastic Dynamic Programming (SDP) is a conventional procedure to extract optimal reservoir operating policies. SDP appliesexhaustive search based on the total combinations of discrete values of state variables. The accuracy of SDP can be improved byincreasing the number of intervals of inflow or reservoir volume domain; however, increasing the inflow intervals is limited andincreasing the reservoir volume intervals does not guarantee the regular improvement of results. This study attempts to determine thenear optimum discrete reservoir volume values by cooperating SDP and non-linear programming (CSDP) optimization modules, sothe acceptable performance is achieved by limited intervals of the state variables. Towards this end, the objective function is utilizedwith a hedging policy to control the amount of the monthly reservoir release. The non-linear programming optimization moduleembodied in SDP, determines the proper values of reservoir volume taking into account the hedging policy and available constrains. Considering various numbers of inflow and reservoir volume intervals, the conventional SDP results are implemented and comparedto CSDP approach. Our comparisons indicate an improved reliability performance obtained in the CSDP technique. A DistanceBased Interpolation Formula (DBIF) is also illustrated and applied to employ optimal operating polices for reservoir operationmanagement. Findings show better performance of DBIF as compared to conventional methods like Multiple Linear Regression(MLR) during long-time simulation of reservoir operation.

        Experimental Study on Mechanical Properties of Cement-Stabilized Soil Blended with Crushed Stone Waste

        Waste generated by the processing of natural stone poses environmental and economic problems. One promising use for stone waste is soil stabilization. In this paper, a number of tests including unconfined compressive strength (UCS) and California bearing ratio (CBR) of crushed stone were performed to study the effect of curing time, soaked and unsoaked conditions and freeze-thaw cycles on the geotechnical characteristics of cement-stabilized specimens. The results indicate that crushed stone waste can be combined with cement as a stabilizer to improve soil properties. Also the optimum moisture content (OMC) and maximum dry density (MDD) are dependent on the cement content and almost independent of the crushed stone waste content. The CBR increased significantly as the crushed stone waste and cement contents increased under both soaked and unsoaked conditions, but these had little effect on the UCS at curing times of less than 14 days. Cement-stabilized specimens with 10% crushed stone waste showed a significant increase in UCS. The CBR and UCS of unsoaked specimens were higher than for soaked specimens at different crushed stone waste and cement contents. The CBR and UCS values increased with an increase in curing time. However, the curing time had a more obvious effect on UCS than on CBR.

      • High Expression of Stem Cell Marker ALDH1 is Associated with Reduced BRCA1 in Invasive Breast Carcinomas

        Background: Cancer stem cells (CSC) have been described in a variety of malignancies, including breast carcinomas. Among several markers, aldehyde dehydrogenase 1 (ALDH1) has been identified as reliable for breast cancer stem cells. Knockdown of BRCA1 in primary breast epithelial cells leads to an increase in cells expressing ALDH1. Methods: We examined 127 breast carcinomas for expression of ALDH1, using immunohistochemistry and correlated with clinicopathological parameters as well as the BRAC1 status. Results: Comparing the results for both ALDH1 and BRCA1 expression showed a significant inverse association between the two, indicating that reduced BRCA1 was more often seen in breast cancer cells expressing ALDH1 (p-value = 0.044). A total of 24/110 (22%) of tumours displayed the ALDH1 + / BRCA1 -/low phenotype, which showed a trend for a relation with a high grade (p-value= 0.056). Cytoplasmic expression of ALDH1 was not correlated with tumour characteristics. Conclusion: Taken together, our findings suggest that increased ALDH1 is inversely correlated with decreased BRCA1 in a series of unselected breast carcinomas. Therefore, ALDH1 positive (cancer stem) cells with reduced BRCA1 phenotype may indicate a subset of patients for whom specific targeting of the CSC marker ALDH1 and more aggressive adjuvant treatment is appropriate.
