Based on comprehensive spectroscopic analyses including IR, MS, 1D- and 2D NMR (COSY, HSQC, and HMBC), their structures were elucidated as 3-hydroxyl-5-methyphenol-1-O-[${\beta}$-D-glucopyranosyl-($1{\rightarrow}3$)-${\beta}$-D-glucopyranoside (1) and 1',3'-dimethoxyl-4-hydroxyalangifolioside (2).

Effects of sucrose fatty acid esters (SEs) with hydrophilic-lipophilic balance (HLB) values of 5 (S-570), 9 (S-970), and 15 (S-1570) on the pasting and rheological properties of waxy rice flour (WRF) were investigated. Rapid Visco Analyzer (RVA) showed that addition of SEs affected the pasting properties of WRF. Rheological experiment includes the steady shear flow characteristics, and dynamic viscoelastic properties were also determined using a controlled-stress rheometer. The steady shear tests demonstrated that the viscosity data fitted well with the power law model (R2≥0.976) and all WRF pastes exhibited typical pseudoplastic and shear-thinning properties. Dynamic rheological measurements revealed that the addition of S-970 and S-1570 significantly increased the values of G' and G'' of samples, whereas the addition of S-570 decreased these values. The addition of S-1570 reduced the value of tanδ, whereas S-570 and S-970 increased it.

A new elliptical one-sided bolt was applied for connection of the steel square tubular column and the I-shaped beam joint. This new bolt is simple in structure and easy to install. Besides, as the bolt head and bolt hole are both elliptical, the new one is quite diff erent from the traditional circular bolts lacking bending performance studies of the joint. Therefore, experiments and fi nite element analysis on ultimate bearing capacity of the joint were conducted, monotonic static loading of six joint specimens were tested, and the moment-rotation curves were obtained in this study. The eff ects of various factors such as axial load ratio and bolt size on bearing capacity were analyzed. Finite element simulation was carried out, and it fi tted tests well. Both tests and simulation showed that failure mode of joints is fi rstly yielding at the endplate. The larger the bolt size, the better the joint ultimate bearing capacity can be enhanced. The M16 group indicated a pattern that the larger axial load on column, the less of ultimate bearing capacity of the joint. However, the M20 group comes to a diff erent pattern which contradicts the assumption, thus further studies are needed in this regard.

Purpose: To investigate the effect of deacetylase inhibitory trichostatin A (TSA) on anti HepG2 liver carcinoma cells and explore the underlying mechanisms. Materials and Methods: HepG2 cells exposed to different concentrations of TSA for 24, 48, or 72h were examined for cell growth inhibition using CCK8, changes in cell cycle distribution with flow cytometry, cell apoptosis with annexin V-FTIC/PI double staining, and cell morphology changes under an inverted microscope. Expression of ${\beta}$-catenin, HDAC1, HDAC3, H3K9, CyclinD1 and Bax proteins was tested by Western blotting. Gene expression for ${\beta}$-catenin, HDAC1and HDAC3 was tested by q-PCR. ${\beta}$-catenin and H3K9 proteins were also tested by immunofluorescence. Activity of Renilla luciferase (pTCF/LEF-luc) was assessed using the Luciferase Reporter Assay system reagent. The activity of total HDACs was detected with a HDACs colorimetric kit. Results: Exposure to TSA caused significant dose-and time-dependent inhibition of HepG2 cell proliferation (p<0.05) and resulted in increased cell percentages in G0/G1 and G2/M phases and decrease in the S phase. The apoptotic index in the control group was $6.22{\pm}0.25%$, which increased to $7.17{\pm}0.20%$ and $18.1{\pm}0.42%$ in the treatment group. Exposure to 250 and 500nmol/L TSA also caused cell morphology changes with numerous floating cells. Expression of ${\beta}$-catenin, H3K9and Bax proteins was significantly increased, expression levels of CyclinD1, HDAC1, HDAC3 were decreased. Expression of ${\beta}$-catenin at the genetic level was significantly increased, with no significant difference in HDAC1and HDAC3 genes. In the cytoplasm, expression of ${\beta}$-catenin fluorescence protein was not obvious changed and in the nucleus, small amounts of green fluorescence were observed. H3K9 fluorescence protein were increased. Expression levels of the transcription factor TCF werealso increased in HepG2 cells following induction by TSA, whikle the activity of total HDACs was decreased. Conclusions: TSA inhibits HDAC activity, promotes histone acetylation, and activates Wnt/${\beta}$-catenin signaling to inhibit proliferation of HepG2 cell, arrest cell cycling and induce apoptosis.
Hydrogels with diffusion-facilitated porous network for improved adsorption performance

Porous alginate-based hydrogel beads (porous ABH) have been prepared through a facile and sustainable template-assisted method using nano-calcium carbonate and nano-CaCO3 as pore-directing agent for the efficient capture of methylene blue (MB). The materials were characterized by various techniques. The sorption capacities of ABH towards MB were compared with pure sodium alginate (ABH-1:0) in batch and fixed-bed column adsorption studies. The obtained adsorbent (ABH-1:3) has a higher BET surface area and a smaller average pore diameter. The maximum adsorption capacity of ABH-1:3 obtained from Langmuir model was as high as 1,426.0mg g1. The kinetics strictly followed pseudo-second order rate equation and the adsorption reaction was effectively facilitated, approximately 50minutes to achieve adsorption equilibrium, which was significantly shorter than that of ABH-1:0. The thermodynamic parameters revealed that the adsorption was spontaneous and exothermic. Thomas model fitted well with the breakthrough curves and could describe the dynamic behavior of the column. More significantly, the uptake capacity of ABH-1:3 was still higher than 75% of the maximum adsorption capacity even after ten cycles, indicating that this novel adsorbent can be a promising adsorptive material for removal of MB from aqueous solution under batch and continuous systems.
KLF4 Expression Correlates with the Degree of Differentiation in Colorectal Cancer

Background/Aims: Kruppel-like factor 4 (KLF4) is an epithelial-specifi c transcription factor primarily expressed in the gastrointestinal tract that mediates growth arrest in the colonic epithelium. We tried to find whether KLF4 expression is associated with the progression and differentiation of colorectal cancer. Methods: We detected KLF4 expression in 109 colorectal specimens (40 normal appearing mucosa, 7 adenomas, and 62 carcinomas) by immunohistochemistry using a tissue microarray. Western blot and RT-PCR analyses were also performed. Results: The upregulation of KLF4 expression in carcinoma tissue was statistically significant (p<0.05) when compared to normal appearing mucosa. The negative and weak positive staining rates in normal appearing mucosa, adenoma, and carcinoma were 42.5%, 71.4%, and 82.3%, respectively, indicating a decreased degree of KLF4 expression over the course of progressive transformation of normal cells into malignant derivatives. KLF4 protein levels showed no correlation with sex, age, or metastatic state (p>0.05), while KLF4 protein expression correlated with the diagnostic stage (p<0.05). Furthermore, strong KLF4 staining was detected in 22.9% (11/48) and 0% (0/14) of well/moderately and poorly differentiated colorectal cancers, respectively. Our results clearly indicate that KLF4 protein expression signifi cantly correlates with the degree of differentiation in colorectal cancers (p<0.05). KLF4 expression in RKO cells is also upregulated by butyrate, an inducer of differentiation. Conclusions: Downregulation of KLF4 expression may lead to more poorly differentiated tumors.
