<P>The generation of the mammalian heartbeat is a complex and vital function requiring multiple and coordinated ionic channel activities. The functional role of low-voltage activated (LVA) T-type calcium channels in the pacemaker activity of the sinoatrial node (SAN) is, to date, unresolved. Here we show that disruption of the gene coding for CaV3.1/alpha1G T-type calcium channels (cacna1g) abolishes T-type calcium current (I(Ca,T)) in isolated cells from the SAN and the atrioventricular node without affecting the L-type Ca2+ current (I(Ca,L)). By using telemetric electrocardiograms on unrestrained mice and intracardiac recordings, we find that cacna1g inactivation causes bradycardia and delays atrioventricular conduction without affecting the excitability of the right atrium. Consistently, no I(Ca,T) was detected in right atrium myocytes in both wild-type and CaV3.1(-/-) mice. Furthermore, inactivation of cacna1g significantly slowed the intrinsic in vivo heart rate, prolonged the SAN recovery time, and slowed pacemaker activity of individual SAN cells through a reduction of the slope of the diastolic depolarization. Our results demonstrate that CaV3.1/T-type Ca2+ channels contribute to SAN pacemaker activity and atrioventricular conduction.</P>
National Narratives and Archaeology: Thoughts on Koreaness and Hellenism

This article attempts a comparative study of efforts by three countries to construct their respective national narratives (sequences of historical events) and support them with archaeological evidence. Korea, Greece and Cyprus, the last two included within the cultural sphere of Hellenism, are geographically distant and seem unrelated, since their historical destinies never touched before the mid-twentieth century. However, parallel circumstances in which the nation-building processes took place, similar aspirations, and interesting differences make their comparison illuminating. It is argued that all national narratives reflect modern preoccupations rather than historical realities. They are ruled by a more or less common set of parameters and the archaeological record can support these parameters in specific ways. Finally, the ways in which other countries have used archaeology and different narratives to manipulate in their turn the national identities of Greece, Korea and Cyprus are also studied. Use of Amplatzer Vascular Plug to Treat a Biliary Cutaneous Fistula

Several substances have been used in an attempt to sclerose biliary ducts associated with persistent biliary-cutaneous fistula (BCF). The AMPLATZER Vascular Plug (AVP; AGA Medical, USA) system is a recently developed endovascular occlusion device, introduced as an alternative to permanent embolic materials (metallic coils or acrylic glue), in the occlusion of large and medium-calibre arteries and veins. We report a successful use of the AVP to embolize BCF, developed after the removal of an internal-external biliary drainage.

Nowadays, computational fluid dynamics is commonly used by design engineers to evaluate and compare losses in hydraulic components as it is less expensive and less time consuming than model tests. For that purpose, an automatic tool for casing and distributor analysis will be presented in this paper. An in-house mesh generator and a Reynolds Averaged Navier-Stokes equation solver using the standard k-ω shear stress transport (SST) turbulence model will be used to perform all computations. Two solvers based on the C++ OpenFOAM library will be used and compared to a commercial solver. The performance of the new fully coupled block solver developed by the University of Lucerne and Andritz will be compared to the standard 1.6ext segregated simpleFoam solver and to a commercial solver. In this study, relative comparisons of different geometries of casing and distributor will be performed. The present study is thus aimed at validating the block solver and the tool chain and providing design engineers with a faster and more reliable analysis tool that can be integrated into their design process.

How Many Antimicrobial Peptide Molecules Kill a Bacterium? The Case of PMAP-23

<P>Antimicrobial peptides (AMPs) kill bacteria mainly through the perturbation of their membranes and are promising compounds to fight drug resistance. Models of the mechanism of AMPs-induced membrane perturbation were developed based on experiments in liposomes, but their relevance for bacterial killing is debated. We determined the association of an analogue of the AMP PMAP-23 to <I>Escherichia coli</I> cells, under the same experimental conditions used to measure bactericidal activity. Killing took place only when bound peptides completely saturated bacterial membranes (10<SUP>6</SUP>–10<SUP>7</SUP> bound peptides per cell), indicating that the “carpet” model for the perturbation of artificial bilayers is representative of what happens in real bacteria. This finding supports the view that, at least for this peptide, a microbicidal mechanism is possible <I>in vivo</I> only at micromolar total peptide concentrations. <P>Host-defense peptides (HDPs) are promising compounds against multidrug-resistant microbes. In vitro, their bactericidal and toxic concentrations are significantly different, but this might be due to the use of separate assays, with different cell densities. For experiments with a single cell type, the cell-density dependence of the active concentration of the DNS-PMAP23 HDP could be predicted based on the water/cell-membrane partition equilibrium and exhibited a lower bound at low cell counts. On the basis of these data, in the simultaneous presence of both bacteria and an excess of human cells, one would expect no significant toxicity, but also inhibition of the bactericidal activity due to peptide sequestration by host cells. However, this inhibition did not take place in assays with mixed cell populations, showing that for the HDP esculentin-la(1-21)NH2, a range of bactericidal, nontoxic concentrations exists and confirming the effective selectivity of HDPs. Mixed-cell assays might be necessary to effectively asses HDP selectivity.</P>
