        Development of the Droplet Digital PCR Method for the Detection and Quantification of Erwinia pyrifoliae

        Black shoot blight disease caused by Erwinia pyrifoliae has serious impacts on quality and yield in pear production in Korea; therefore, rapid and accurate methods for its detection are needed. However, traditional detection methods require a great deal of time and fail to achieve absolute quantification. In the present study, we developed a droplet digital polymerase chain reaction (ddPCR) method for the detection and absolute quantification of E. pyrifoliae using a pair of species-specific primers. The detection range was 103- 107 copies/ml (DNA templates) and cfu/ml (cell culture templates). This new method exhibited good linearity and repeatability and was validated by absolute quantification of E. pyrifoliae DNA copies from samples of artificially inoculated immature pear fruits. Here, we present the first study of ddPCR assay for the detection and quantification of E. pyrifoliae. This method has potential applications in epidemiology and for the early prediction of black shoot blight outbreaks.

        Effect of Male-Sterile Cytoplasm on the Genetic Performance of Agronomic Traits in $F_1$ Hybrid Rices

        Three different male-sterile cytoplasmic lines and their common maintainer 'Zhenshan 97B' and two elite restorer lines were used to study cytoplasmic effects on agronomic trait manifestation per se under different nitrogen supply levels. The result showed that cytoplasmic effects could be modified by nitrogen environments. The cytoplasmic effect on grain yield under 150 kg N/ha varied depending on crosses, while it was significantly negative in most crosses under both 60 and 330 kg N/ha. The correlation and path-coefficient analyses suggested that it was expected to improve cytoplasmic effects through reducing maximum tillers and increasing the percentage of productive tillers, leading to increased productive tillers and higher yield in hybrid rice by the aid of cultural practice and genetic transformation. This study also revealed that the same cytoplasm in different combinations had differential effect under the same nitrogen environment, indicating that cytoplasmic effect was produced by interaction of nuclear genes with cytoplasm rather than cytoplasm per se. These results indicated the usefulness of evaluating diverse cytoplasmic sources in various nuclear genotypes bred for hybrid rice breeding program. The finding also suggested that negative cytoplasmic effect could be effectively overcome by elite restorer lines through the interaction of nuclear genes with female cytoplasm.

      • Effects of Emodin Extracted from Chinese Herbs on Proliferation of Non-small Cell Lung Cancer and Underlying Mechanisms

        To aim of this was to observe emodin-mediated cytotoxicity and its influence on Rad51 and ERCC1 expressionin non-small cell lung cancer (NSCLC). NSCLC cells were cultured in vitro with emodin at various concentrations (0, 25, 50, 75 and $100\;{\mu}mol/L$) for 48h and the proliferation inhibition rate was determined by the MTT method. Then, NSCLC were treated with emodin (SK-MES-1 $40\;{\mu}mol/L$, A549 $70\;{\mu}mol/L$) or $20\;{\mu}mol/L$ U0126 (an ERK inhibitor) for 48 h, or with various concentrations of emodin for 48 h and the protein and mRNA expressions of ERCC1 and Rad51 were determined by RT-PCR and Western blot assay, respectively. Emodin exerted a suppressive effect on the proliferation of NSCLC in a concentration dependent manner. Protein and mRNA expression of ERCC1 and Rad51 was also significantly decreased with the dose. Vacuolar degeneration was observed in A549 and SK-MES-1 cell lines after emodin treatment by transmission electron microscopy. Emodin may thus inhibited cell proliferation in NSCLC cells by downregulation ERCC1 and Rad51.

      • Benefit of Post-mastectomy Radiotherapy of the Supra-/infraclavicular Lymphatic Drainage Area in Breast Cancer Patients

        Background: This study investigated the survival benefit of radiotherapy (RT) of the supra- and infraclavicular lymphatic drainage area in Chinese women with T1-2N1M0 breast cancer receiving mastectomy. Methods: A total of 593 cases were retrospectively reviewed from 1998 to 2007. The relationship between supra- or infraclavicular fossa relapse (SCFR) and post-operative RT at the supra-/infraclavicular lymphatic drainage area was evaluated. Results: The majority of patients (532/593; 89. 8%) received no RT while 61 patients received RT. The median follow-up was 85 months. Among patients without RT, 54 (10. 2%) developed recurrence in the chest wall or ipsilateral SCFR. However, none of the 61 patients who underwent RT demonstrated SCFR. One patient who received RT (1. 6%) experienced recurrence in the chest wall. Univariate analysis revealed that age and molecular subtype (both P < 0. 05) were two prognostic factors related to supraclavicular and infraclavicular fossa relapse-free survival (SFRFS). Multivariate analysis revealed that only Her-2 positive status (P = 0. 011) was an independent predictor of SFRFS. RT had no influence on distant metastasis (P = 0. 328) or overall survival (P = 0. 541). SCFR significantly affected probability of distant metastasis (P < 0. 001) and overall survival (P < 0. 001). Conclusion: Although RT was not significantly associated with SFRFS, postoperative RT was significantly associated with a lower locoregional (i. e., supraclavicular/infraclavicular and chest wall) recurrence rate. SCFR significantly influenced distant metastasis-free survival, which significantly influenced the overall survival of T1-2N1M0 breast cancer patients after mastectomy. Thus, prophylactic RT is recommended in T1-2N1M0 breast cancer patients, especially those who have Her-2 positive lesions.

      • Construction of sports engineering structures with high resistance to improve the quality of sports training

        The textile industry has benefited from nanotechnology in various fields of application as the use of nanomaterials, and nanotechnology is multiplying. Nanoparticles can increase the performance of textiles by up to 100 times when used in finishing, coating, and dyeing techniques, providing them with capabilities they did not previously possess. Nanotechnology is used in the textile chemical industry to produce sports mats with stain resistance, flame resistance, wrinkle resistance, moisture management, antimicrobial quality, and UV protection. The incorporation of nanomaterials into fabrics can have a significant effect on their properties, including shrinkage, strength, electrical conductivity, and flammability. Various inventions and innovations may result from nano-processed textiles in the future, thus leading to the advancement of science. This article presents the construction of sports engineering structures with high resistance to improve the quality of sports training. The mechanical properties of sports mats are improved with the help of nanotechnology. Strength, elasticity, and tear resistance are among these properties. This method enables the production of elastic, durable, and tear-resistant sports mats.

      • Formation of heterostructures via direct growth CN on <i>h</i>-BN porous nanosheets for metal-free photocatalysis

        <P><B>Abstract</B></P> <P>Many recent advances in creating heterostructures based on 2D materials have opened new possibilities in catalysis. This study prepared a new 2D hybrid photocatalyst (CBN-x) consisting of CN and <I>h-</I>BN using low-cost precursors (urea and boric acid). The effects of CN loading on the structure, surface chemistry and photocatalytic activities were systematically investigated. The photocatalytic activities of CBN-x samples were tested for the production of H<SUB>2</SUB> and H<SUB>2</SUB>O<SUB>2</SUB>, which demonstrated markedly enhanced activities without the need of noble metal co-catalysts. However, CBN-x activities for the photocatalytic oxidation of organic compound were not better than that of h-BN. Loading CN on <I>h-</I>BN sheets, C 2p and N 2p orbitals from CN introduce new valence and conduction band edges, which gradually narrowed the bandgap and enhanced light absorption efficiency of the hybrid photocatalysts. In such metal-free systems, electrons generated in CN transfer to h-BN, while photogenerated holes on <I>h-</I>BN transfer to CN, which enhances the charge separation through the heterojunction interface (CN/BN). Therefore, increasing the CN loading enhances the overall efficiency of photocatalysis until excessive loading of CN covers the active sites on h-BN.</P> <P><B>Highlights</B></P> <P> <UL> <LI> In this work, we introduced a new kind of 2D heterojunction created by loading CN on h-BN nanosheets. </LI> <LI> The effects of CN loading on the surface chemistry and photocatalytic activities were systematically investigated. </LI> <LI> Photocatalytic H<SUB>2</SUB> and H<SUB>2</SUB>O<SUB>2</SUB> production activities of hybrid CBN-x samples were proven to be enhanced. </LI> <LI> We carried out first-principle calculations which revealed the unique physical features of CN/BN heterostructures. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Draft Genome Sequence of Aureobasidium pullulans Strain MHAU2101, a Biological Control Agent against Fire Blight from Korea

        In this study, we present the draft genome of Aureobasidium pullulans strain MHAU2101, which is the first strain to effectively control fire blight caused by Erwinia amylovora in Korea. The genome of strain MHAU2101 was composed of 28,669,322 base pairs, with a C+G content of 50.4%. The assembly comprised 17 contigs and had 99.22% completeness. The results of this study will be a valuable resource for future research on the biocontrol mechanism of A. pullulans strain MHAU2101.

      • PIK3CA and AKT Gene Polymorphisms in Susceptibility to Osteosarcoma in a Chinese Population

        Purpose: To explore the association between PIK3CA and AKT single nucleotide polymorphisms(SNP) and osteosarcoma susceptibility. Methods: TaqMan polymerase chain reaction(PCR) was used to detect the genotypes of SNPs (rs7646409, rs6973569 and rs9866361) in peripheral blood samples from 59 patients with osteosarcoma and from 63 healthy controls. Unconditional logistic regression was used to analyze the correlation between SNPs and osteosarcoma risk. Results: No statistically significant difference was found between osteosarcoma patients and healthy controls in the genotype of AKT rs6973569 (P=0.7). However, after stratified analysis, the genotype AA of AKT rs6973569 carried a higher risk of osteosarcoma metastasis (OR:2.94, 95%CL:1.00-8.59); the difference of rs7646409 genotype distributions between the case and control groups was statistically significant (P=0.032). Taking genotype TT as a reference, the risk of osteosarcoma increased three fold in patients with genotype CC (OR:3.47, 95%CL:1.26-9.56). A statistically significant difference was found between the alleles C and T (P=0.005). Further analysis showed that the risk factor was more pronounced in male patients with Enneking's stage IIB and osteoblastic osteosarcoma. PIK3CA rs9866361 did not fit Hardy-Weinberg equilibrium (P<0.05). Conclusions: Genotype CC in locus PIK3CA rs7646409 may increase the risk of osteosarcoma in the Chinese population.
