Enhancement of Haloacetate Dehalogenase Production by Strain Mutation and Condition Optimization

Enhancement of the activity of an inducible chloroacetate dehalogenase was carried out by efficient and safe mutation with UV and microwave irradiation along with optimization of culture conditions. First, a stable mutant of Pseudomonas sp. CGMCC 3267-MW6 with chloroacetate dehalogenase activity of 2.77 U/mL (3-fold higher activity than the wild strain) was produced by mutation. The maximum activity of this inducible enzyme was measured as 29.41 U/mL when Pseudomonas sp. CGMCC 3267-MW6 was cultured with 4 g/L 3-hydroxybutyrate for 12 h followed by 40 mM 3-chlorobutyrate for an additional 20 h. Production of the enzyme was found to be associated with growth of the bacterium. According to these results, we determined that the optimum inducer of chloroacetate dehalogenase activity would be a hard degradable substrate. The optimum auxiliary carbon source would be the primary metabolite of the substrate or the precursor of the metabolite. The optimum time of inducer supplementation would be during the middle stage of exponential phase. The optimum concentration of substrate would be sufficient but would not induce inhibition. Finally, the optimum collection time would be at the later stage of exponential phase. This work provides further knowledge of chloroacetate dehalogenase and the optimization of inducible enzyme production.
Tazarotene-Induced Gene 1 Interacts with DNAJC8 and Regulates Glycolysis in Cervical Cancer Cells

The tazarotene-induced gene 1 (TIG1) protein is a retinoid-inducible growth regulator and is considered a tumor suppressor. Here, we show that DnaJ heat shock protein family member C8 (DNAJC8) is a TIG1 target that regulates glycolysis. Ectopic DNAJC8 expression induced the translocation of pyruvate kinase M2 (PKM2) into the nucleus, subsequently inducing glucose transporter 1 (GLUT1) expression to promote glucose uptake. Silencing either DNAJC8 or PKM2 alleviated the upregulation of GLUT1 expression and glucose uptake induced by ectopic DNAJC8 expression. TIG1 interacted with DNAJC8 in the cytosol, and this interaction completely blocked DNAJC8-mediated PKM2 translocation and inhibited glucose uptake. Furthermore, increased glycose uptake was observed in cells in which TIG1 was silenced. In conclusion, TIG1 acts as a pivotal repressor of DNAJC8 to enhance glucose uptake by partially regulating PKM2 translocation.
Chronic Kidney Disease and Associated Cardiovascular Risk Factors in Chinese with Type 2 Diabetes

Background: To determine the frequency of chronic kidney disease (CKD) and its associated risk factors in Chinese type 2 diabetic patients, we conducted a cross-sectional study in Nanjing, China, in the period between January 2008 and December 2009. Methods: Patients with type 2 diabetes under the care by Jiangsu Province Official Hospital, Nanjing, China were invited for assessment. CKD was defined as the presence of albuminuria or estimated glomerular filtration rate <60 mL/min/1.73 m2. Albuminuria was defined as urinary albumin-to-creatinine ratio ≥30 mg/g. Results: We recruited 1,521 urban Chinese patients with type 2 diabetes (mean age, 63.9±12.0 years). The frequency of CKD and albuminuria was 31.0% and 28.9%, respectively. After adjusted by age and sex, hypertension, anemia and duration of diabetes were significantly associated with CKD with odds ratio (95% confidence interval) being 1.93 (1.28 to 2.93), 1.70 (1.09 to 2.64), and 1.03 (1.00 to 1.06), respectively. Conclusion: In conclusion, CKD was common in the urban Nanjing Chinese with type 2 diabetes. Strategies to prevent or delay progression of kidney disease in diabetes should be carried out at the early disease course of type 2 diabetes.

Fisetin is an effective compound extracted from lacquer which has been used in the treatment of various diseases. Preliminary data indicate that it also exerts specific anti-cancer effects. However, the manner in which fisetin regulates cancer growth remains unknown. In this study, we elucidated interference of fisetin with targets of the nuclear factor ${\kappa}B$ signal transduction pathway activated by Epstein-Barr virus encoding latent membrane protein 1 (LMP1)in nasopharyngeal carcinoma (NPC) cells, Results showed that fisetin inhibited the survival rate of CNE-LMP1 cells and NF-${\kappa}B$ activation caused by LMP1. Fisetin also suppressed nuclear translocation of NF-${\kappa}B$ (p65) and $I{\kappa}B{\alpha}$ phosphorylation, while inhibiting CyclinD1, all key targets of the NF-${\kappa}B$ signal transduction pathway. It was suggested that interference effects of fisetin with signal transduction activated by LMP1 encoded by the Epstein-Barr virus may play an important role in its anticancer potential.

Matching Reconstruction Algorithms Performance Comparison based on Compressed Sensing in GPR Imaging

Compressed sensing (CS) provides a new solution for the problems of requiring large amount of measurements data and long data acquisition time in radar application, and both issues also exist in ground penetrating radar (GPR). Aiming at this problem, we adopt impulse radar with CS framework, and transform the GPR imaging into sparse constraint optimization problem performed on time-domain sub-sampling in this paper. Specifically, it focuses on the impulse GPR imaging method based on CS under double underground targets condition containing noise and abundant clutter. Furthermore, the performance of matching reconstruction algorithms under the different signal to noise ratios (SNR), measurement dimensions and sparseness values is also presented. The experimental results show that CS algorithms based on matching reconstruction can obviously reduce measurement data, improve the image quality and make a better anti-noise performance. When SNR of measurement data is 1dB, the probability of accurate imaging can still reach 95%. So we may reasonably conclude that the regularized orthogonal matching pursuit algorithm has a better performance than the other matching algorithms.
A Novel Control Strategy for Input-Parallel-Output-Series Inverter System

This paper presents a topology structure and control method for an input-parallel-output-series(IPOS) inverter system which is suitable for high input current, high output voltage, and high power applications. In order to ensure the normal operation of the IPOS inverter system, the control method should achieve input current sharing(ICS) and output voltage sharing(OVS) among constituent modules. Through the analysis in this paper, ICS is automatically achieved as long as OVS is controlled. The IPOS inverter system is controlled by a three-loop control system which is composed of an outer common-output voltage loop, inner current loops and voltage sharing loops. Simulation results show that this control strategy can achieve low total harmonic distortion(THD) in the system output voltage, fast dynamic response, and good output voltage sharing performance.
