        Appropriate Nouns with Obligatory Modifiers

        The notion of appropriate sequence as introduced by Z. Harris provides a powerful syntactic way of analysing the detailed meaning of various sentences, including ambiguous ones. In an adjectival sentence like The leather was yellow, the introduction of an appropriate noun, here colour, specifies which quality the adjective describes. In some other adjectival sentences with an appropriate noun, that noun plays the same part as colour and seems to be relevant to the description of the adjective. These appropriate nouns can usually be used in elementary sentences like The leather had some colour, but in many cases they have a more or less obligatory modifier. For example, you can hardly mention that an object has a colour without qulifying that colour at all. About 300 French nouns are appropriate in at least one adjectival sentence and have an obligatory modifier. They enter in a number of sentence structures related by several syntactic transformations. The appropriateness of the noun and the fact that the modifier is obligatory are reflected in these transformations. The description of these syntactic phenomena provides a basis for a classfication of these nouns. It also concerns the lexical properties of thousands of predicative adjectives, and in particular the relations between the sentence without the noun; The leather was yellow and the adjectival sentence with the noun: The colour of the leather was yellow.

      • β-Arrestin/AP-2 Interaction in G Protein-coupled Receptor Internalization : IDENTIFICATION OF A β-ARRESTIN BINDING SITE IN β_2-ADAPTIN

        β-Arrestins, proteins involved in the turn-off of G protein-coupled receptor (GPCR) activation, bind to the β_2-adaptin subunit of the clathrin adaptor AP-2. The interaction of β_2-adaptin with β-Arrestin involves critical arginine residues in the C-terminal domain of β-Arrestin and plays an important role in initiating clathrin-mediated endocytosis of the β_2-adrenergic receptor(β_2AR) (Laporte, S. A., Oakley, R. H., Holt, J. A., Barak, L. S., and Caron, M. G. (2000) J. Biol. Chem. 276, 23120-23126). However, the β-arrestin-binding site in β_2-adaptin has not been identified, and little is known about the role of β-arrestin/AP-2 interaction in the endocytosis of other GPCRs. Using in vitro binding assays, we have identified two glutamate residues (Glu-849 and Glu-902) in β_2-adaptin that are important in β-arrestin binding. These residues are located in the platform subdomain of the C terminus β_2-adaptin, where accessory/adapter endocytic proteins for other classes of receptors interact, distinct from the main site where clathrin interact. The functional significance of the β-arrestin/AP-2/clathrin complex in the endocytosis of GPCRs such as the β_2AR and vasopressin type Ⅱ receptor was evaluated using mutant constructs of the β_2-adaptin C terminus containing either the clathrin and the β-arrestin binding domains or the β-arrestin-binding domain alone. When expressed in human embryonic kidney 293 cells, both constructs acted as dominant negatives inhibiting the agonist-induced internalization of the β_2AR and the vasopressin type Ⅱ receptor. In addition, although the β_2-adaptin construct containing both the clathrin and β-arrestin binding domains was able to block the endocytosis of transferrin receptors, a β_2-adaptin construct capable of assoociating with β-arrestin but lacking its high affinity clathrin interaction did not interfere with transferrin receptor endocytosis. These results suggest that the interaction of β-arrestin with β_2-adaptin represents a selective endocytic trigger for several members of the GPCR family.

      • The CCAAT box-binding transcription factor NF-YA1 controls rhizobial infection

        <P>Symbiosis between legume plants and soil rhizobia culminates in the formation of a novel root organ, the ‘nodule’, containing bacteria differentiated as facultative nitrogen-fixing organelles. MtNF-YA1 is a <I>Medicago truncatula</I> CCAAT box-binding transcription factor (TF), formerly called HAP2-1, highly expressed in mature nodules and required for nodule meristem function and persistence. Here a role for MtNF-YA1 during early nodule development is demonstrated. Detailed expression analysis based on RNA sequencing, quantitiative real-time PCR (qRT-PCR), as well as promoter–β-glucuronidase (GUS) fusions reveal that <I>MtNF-YA1</I> is first induced at the onset of symbiotic development during preparation for, and initiation and progression of, symbiotic infection. Moreover, using a new knock-out mutant, <I>Mtnf-ya1-1</I>, it is shown that <I>MtNF-YA1</I> controls infection thread (IT) progression from initial root infection through colonization of nodule tissues. Extensive confocal and electronic microscopic observations suggest that the bulbous and erratic IT growth phenotypes observed in <I>Mtnf-ya1-1</I> could be a consequence of the fact that walls of ITs in this mutant are thinner and less coherent than in the wild type. It is proposed that <I>MtNF-YA1</I> controls rhizobial infection progression by regulating the formation and the wall of ITs.</P>

        Genome wide identification of Staufen2-bound mRNAs in embryonic rat brains

        Messenger ribonucleoprotein particles (mRNPs) are used to transport mRNAs along neuronal dendrites to their site of translation. Staufen2 is an mRNA-binding protein expressed in the cell bodies and cellular processes of different brain cells. It is notably involved in the transport of dendritic mRNAs along microtubules. Its knockdown expression was shown to change spine morphology and impair synaptic functions. However, the identity of Staufen2-bound mRNAs in brain cells is still completely unknown. As a mean to identify these mRNAs, we immunoprecipitated Staufen2-containing mRNPs from embryonic rat brains and used a genome wide approach to identify Staufen2-associated mRNAs. The genome wide approach identified 1780 mRNAs in Staufen2-containing mRNPs that code for proteins involved in cellular processes such as post-translational protein modifications, RNA metabolism, intracellular transport and translation. These results represent an additional and important step in the characterization of Staufen2- mediated neuronal functions in rat brains. [BMB reports 2010; 43(5): 344-348]

        Location of Major Determinants for the Differential Expression Between aceA and aceK in the Glyoxylate Bypass Operon of E . coli

        Although the three enzymes unique to the glyoxylate cycle are expressed from a single ace operon in E. coli, the intracellular level of isocitrate lyase expressed from aceA is more than 100-fold higher than that of isocitrate dehydrogenase (IDH) kinase/phosphatase, the product of aceK. In an effort to elucidate the mechanism(s) for the differential expression, a sequence in the aceA/aceK intergenic region which can form a very stable secondary structure (ΔG= -54 ㎉/㏖) when transcribed was deleted by site-directed mutagenesis. Determination of enzyme activities and maxicell labeling experiment with the deletion mutant and wild type operon clone indicated that the secondary structure plays little role in the downregulation of aceK expression. However, a comparison of β-galactosidase activities of several aceA::lacZ and aceK::lacZ fusion genes suggested that some 20-fold downshift occurs still in the aceA/aceK intergenic region, although the secondary structure-forming sequence within the region proved not to be involved. Another 8-fold drop in the expression of lacZ was observed from two fusion genes which had their fusion points within the coding sequence of aceK, suggesting one of the major determinants for the differential expression between aceA and aceK is located within the structural part of aceK.

        Global Networking of Cancer and NCD Professionals Using Internet Technologies: The Supercourse and mHealth Applications

        Cancer is a leading cause of death around the world. Education is at the core of cancer prevention activities, especially programs targeting empowering existing public health workforce. In the past 10 years, researchers at the University of Pittsburgh have been building the Global Health Network Supercourse project, a library of over 4500 online lectures and a network of nearly 50000 public health professionals in 174 countries. As of November 2010, the overall number of Supercourse participants from Asia exceeds 7000 participants. The Supercourse network has been investigating methods for Internet based recruitment of cancer prevention professionals in order to network cancer experts locally and globally, including the use of mHealth technologies for cancer research education and for NCD registries. Supercourse is a tool that can offer a solution to the challenges of information sharing, especially in the field of NCDs and cancer. In this paper, we highlight the need for the development of Cancer Supercourse with Satellite in Asia and encourage faculty members from Asia to join the network.
