      • Pichia pastoris로부터 Toll-like Receptor 9의 세포 내 도메인 단백질의 발현과 순수분리 정제

        Toll-like receptors (TLR) are important components of innate immunity in the defense against pathogens. TLRs recognize pathogen-associated common molecular patterns. TLRs are similar to the receptors involved in defense responses in plants. TLR protein is a type 1 membrane protein, consisting of an extracellular domain containing leucine-rich repeats and a cytoplasmic domain. The cytoplasmic domain delivers ligand recognition signals that result in production of anti-microbial agents. The cytoplasmic domain (amino acid 858-1032) of toll-like receptor 9 has been expressed using methylotrophic yeast Pichia pastoris. The protein expression was confirmed by Western-blot, N-terminal sequencing and MALDI-TOF mass spectrometry. The proteins have been purified by nickel affinity, cation exchange and gel-filtration chromatography.

      • 韓國産 산형과 植物에 關한 本草學的 硏究

        For the purpose of developing Korean Herbaolgy with the plants of Umbelliferae which grow wild and is planted in Korea, the these and writings on herbalogy, from literature of successive generations, have been thoroughly investigative and the results obtained were as follows : 1. There were totaled to 34 genera and 95 species in Korea and among them medicinal plants are 23 genera, 49 species, some 52 % in total but, the number of species may be added because of the similiar plants. 2. According to the oriental names which can be used for medical purpose, the medicinal plants are belonging to the Umbelliferae family were classified as Radix 20, Fructus 8, Herba 7, Rhizoma 3, Foilum and Flos 2. Thus it was noticed that Radix was the main kind. 3. According to sum of 95 species in Umbelliferae family, they were classified into Angelica genera 23, Bupleurum genera 8, Piucedanum genera 7, Hydrocotyle genera 6. Thus it was noticed that Angelica genera was the main kind, some 24% in total. 4. According to the number of species of the origin plants about each chinese materia midicals, they were classified into RADIX BUPLEURI 6, HERBA HYDROCOTYLEI 4, and RADIX ANGELICAE GLGANTIS·RADIX LIGUSTICI TENUISSIMAE·RADIX ANGELICAE DAHURICAE·FRUCTUS TORILIS 3 each. Thus the origin plants was not complexed reletively. 5. According to nature and flavour of medicinal plants, they were classified into warmth, heat: 19 each, cold, cool : 9 each, balance 1 and unidentified 8. Thus it was noticed that warmth and heat is the main in nature and flavour of medicinal plants. 6. According to the Properties and Principal curative action, they were classified into Drugs for diaphoretics or drugs for treating exterior symptom-complex 7. Drugs for relieving pain 6, Antirheumatics, Drugs which have tonifying actions for the treatment of deficiencies of Yin, Yang, Qi and blood and therefore can be classified into four groups 4, Febrifugal and detoxicant drugs 3, Diuretics and hydragogues, Antitussives 2 each. Thus it was noticed that Drugs for diaphoretics or drugs for treating exterior symptom-complex was the main kind. 7. Comparing to whole medicinal plants 49 kinds, toxic durgs include minor toxin were 2 kinds, 4% of the whole. Thus toxic durgs were rare. From this result, It was revealed that the plants for medical purpose in Umbelliferae was 52% of the whole, in which RADIX was mostly abundunt and toxic plants was so rare that it will be used for clinical treatments more easily. It is considered that many experiements and clinical approaches must be continued to use Umbelliferae plants widely.

        This study is intended to provide a more effective curriculum for the In- Service Training Program of industrial technical teachers. The curricula which have been used during last five years were carefully analyzed and the main conclusions are suggested as follows; 1) It must be given to the teachers to take optional subjects. 2) Experiments should be reinforced make up for the understanding of theory. 3) Practical studies and factory investigations have to be provided to the teachers during the In-Service Training Program. In case of needs, the expert engineer of industries become a instructor to the teachers for explaining the up-to-date industrial techniques and theories. 4) The teachers who attending the In- Service Training Program should take an active interest in the program for getting a remarkable results.

        Soil salinity is one of the most serious abiotic stresses limiting the productivity of agricultural crops. To cope with salt stress, plants respond with physiological, developmental and biochemical changes, including the synthesis of a number of proteins and the induction of gene expression. Salicornia herbacea is a halophytic plant that grows in salt marches and on muddy seashores. In order to understand the biochemical and molecular mechanisms of salt tolerance in S. herbacea, we isolated several genes that involved in the salt tolerance by mRNA differential display.Here we report the cloning of a cDNA encoding fructose-1, 6-bisphosphate aldolase, named ShADL, which is 1293 bp long and contains an open reading frame consisted of 359 amino acids with calculated molecular mass of 39 kDa. ShADL protein showed 86% identity with Arabidopsis and 78% with aldolase of common ice plant. Northern blot analysis revealed that the transcript of ShADL gene was increased dramatically depending on the NaCI concentrations.

        Complementary DNA encoding chloroplast outer envelope membrane protein (OEP) from the halophyte Salicornia herbacea has been cloned and sequenced. The full length cDNA is 596 bp and encodes a polypeptide of 91 amino acid residues with a molecular mass of 8.9 kDa. The expression level of ShOEP increased by salt, drought and ABA treatments. ShOEP expression was largely induced in roots and shoots by high salts. The biological function of ShOEP was examined by yeast complementation. ShOEP can suppress Na+ sensitivity of yeast mutant (cnbΔ) in the presence of salt. These results suggest that ShOEP is a salt inducible gene and may have functions in the regulation of plant salt stress.

        류마티스 관절염에서 Methotrexate-Hydroxychloroquine 병합치료와 Methotrexate 단독치료의 비교연구

        Objective : To assess the relative efficacy of combination therapy of methotrexate(MTX) and hydroxychloroquine(HCQ) in rheumatoid arthritis, we performed an open clinical trial for 6 months. Methods : We compared the efficacy of combination therapy of MTX(7.5-15mg/week) and HCQ(300-400mg/ day) with that of MTX(7.5-15mg/week) alone. Twenty six and thirty two patients were allocated in each treatment arm. Disease activity was assessed by visual analogue pain scale(VAPS), duration of morning stiffness(MS), modified health assessment questionnaire (MHAQ), physician's global assessment(PGA), tender joint count(TJC), swollen joint count(SJC), ESR and CRP at 0, 3, 6 months. Adverse effects were monitored monthly. Results : At the end of trial, all parameters were significantly decreased in both group except CRP in combination therapy group. But the extent of response was not significantly different between two groups except VAPS and MHAQ. Mean values of liver enzymes were significantly increased in MTX alone group, but incidence of hepatotoxicity and extent of elevated enzyme were not significantly different between two groups. Conclusion : Both regimens were effective in the treatment of RA patients, but the efficacy of combination therapy was not superior to MTX alone.$quot;

        This study is concerned with the economic aspects of $125,000m^3$ class LNG carriers with different propulsion plant such as conventional steam turbine and slow speed diesel engine with reliquefaction plant. The ship's speed and L/B ratio were optimized with criterion of required freight rate(RFR) by using the PROCAL computer program package. In order to investigate the effect of fuel oil price, round trip distance and boil-off rate on the RFR and the optimum speed, sensitivity analysis were also performed.

        This paper describes a design model for the conceptual design of ships. Existing design models have problems such that their operating mode of batch versions cannot reflect the design procedures in reality. Reliability of the results is low because the performance estimations are based mainly on empirical formulas. To improve the problems of existing design models, a new design model has been developed. The new model consists of an interactive user interface, a database of main engines, a database of particulars of existing ships, and ten modules for performance estimations. To develop such a user-oriented system, the concept of graphical user interface (GUI) is adopted.

        Molecular Analysis of the Interaction between Human PTPN21 and the Oncoprotein E7 from Human Papillomavirus Genotype 18

        Human papillomaviruses (HPVs) cause cellular hyperproliferation-associated abnormalities including cervical cancer. The HPV genome encodes two major viral oncoproteins, E6 and E7, which recruit various host proteins by direct interaction for proteasomal degradation. Recently, we reported the structure of HPV18 E7 conserved region 3 (CR3) bound to the protein tyrosine phosphatase (PTP) domain of PTPN14, a well-defined tumor suppressor, and found that this intermolecular interaction plays a key role in E7-driven transformation and tumorigenesis. In this study, we carried out a molecular analysis of the interaction between CR3 of HPV18 E7 and the PTP domain of PTPN21, a PTP protein that shares high sequence homology with PTPN14 but is putatively oncogenic rather than tumor-suppressive. Through the combined use of biochemical tools, we verified that HPV18 E7 and PTPN21 form a 2:2 complex, with a dissociation constant of 5 nM and a nearly identical binding manner with the HPV18 E7 and PTPN14 complex. Nevertheless, despite the structural similarities, the biological consequences of the E7 interaction were found to differ between the two PTP proteins. Unlike PTPN14, PTPN21 did not appear to be subjected to proteasomal degradation in HPV18-positive HeLa cervical cancer cells. Moreover, knockdown of PTPN21 led to retardation of the migration/invasion of HeLa cells and HPV18 E7-expressing HaCaT keratinocytes, which reflects its protumor activity. In conclusion, the associations of the viral oncoprotein E7 with PTPN14 and PTPN21 are similar at the molecular level but play different physiological roles.

        Prediction of Malignancy With Endoscopic Ultrasonography in Patients With Branch Duct–Type Intraductal Papillary Mucinous Neoplasm

        OBJECTIVE: We evaluated the accuracy of a new endoscopic ultrasonography (EUS) scoring system to predict malignancy in branch duct–type intraductal papillary mucinous neoplasms (BD-IPMNs). METHODS: We performed a retrospective multicenter study of patients who underwent EUS within 3 months before surgery and were diagnosed as having BD-IPMNs by postoperative pathologies at 8 hospitals in Korea from August 2002 to December 2011. To predict the malignancy, we applied the EUS scoring system consisting of pancreatic cyst size, height of mural nodules, main duct dilatation, septal thickening, and patulous orifice. We evaluated the diagnostic accuracy of the new EUS scoring system and compared it with previous individual risk factors. RESULTS: Eighty-four patients (male-to-female ratio, 55:29; mean [SD] age, 64.7 [7.1] years) had 68 benign BD-IPMNs and 16 malignant intraductal papillary mucinous neoplasms. The EUS scoring system showed 75.0% sensitivity and 94.1% specificity at 7 points. This system (area under the curve, 0.939; 95% confidence interval, 0.884–0.994) resulted in a more accurate prediction than the previous known other factors including Sendai criteria, dilatation of ducts, size of the cyst, and presence of septal thickening and mural nodules. CONCLUSIONS: The EUS scoring system predicted BD-IPMN malignancy more accurately than the Sendai criteria and individual risk factors.
