        Short-term exposure to dim light at night disrupts rhythmic behaviors and causes neurodegeneration in fly models of tauopathy and Alzheimer's disease

        <P><B>Abstract</B></P> <P>The accumulation and aggregation of phosphorylated tau proteins in the brain are the hallmarks for the onset of Alzheimer's disease (AD). In addition, disruptions in circadian rhythms (CRs) with altered sleep-wake cycles, dysregulation of locomotion, and increased memory defects have been reported in patients with AD. <I>Drosophila</I> flies that have an overexpression of human tau protein in neurons exhibit most of the symptoms of human patients with AD, including locomotion defects and neurodegeneration. Using the fly model for tauopathy/AD, we investigated the effects of an exposure to dim light at night on AD symptoms. We used a light intensity of 10 lux, which is considered the lower limit of light pollution in many countries. After the tauopathy flies were exposed to the dim light at night for 3 days, the flies showed disrupted CRs, altered sleep-wake cycles due to increased pTau proteins and neurodegeneration, in the brains of the AD flies. The results indicate that the nighttime exposure of tauopathy/AD model <I>Drosophila</I> flies to dim light disrupted CR and sleep-wake behavior and promoted neurodegeneration.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Dim light disturbs circadian rhythm and sleep behaviors. </LI> <LI> Internal clock is maintained in <I>Drosophila</I> model of tauopathy Alzheimer's disease. </LI> <LI> Short-term disturbance of circadian rhythm by dim light increases formation of phosphorylated Tau in AD model flies. </LI> <LI> Dim light exposure at night promotes neurodegeneration in AD model flies. </LI> </UL> </P>

      • The association of serum leptin with the reduction of food intake and body weight during electroacupuncture in rats

        Previous studies indicate that acupuncture or electroacupuncture (EA) treatment reduces body weight and food intake in rats by increasing the level of anoretic peptides and decreasing that of orexigenic peptides in the hypothalamus. Considering a well-established role of leptin as a major regulator for feeding behavior in the hypothalamus, we hypothesized that EA might exert its effect via increasing serum leptin levels. In this study, we tested our hypothesis by evaluating the effects of EA on food intake and body weight, as well as on serum leptin levels in rats. Rats were randomly divided into 3 groups: AL (fed ad libitum with no treatments), "Holder (fed ad libitum with daily holder restraint) and EA (fed ad libitum with daily holder restraint and 100 Hz EA stimulation) groups. During the four-week experimental period, daily food intake and body weight were measured. At the end of the experiment, levels of serum leptin and corticosterone, and plasma epinephrine (Epi) and norepinephrine (NE) were determined. Here we demonstrate that EA treatment indeed led to reduction of food intake and body weight, and to an increase of serum leptin levels. The level of Epi, NE, and corticosterone increased in the Holder group, but such increase in the level of aforementioned stress hormones was not observed in the EA group. Overall, our results suggest that EA treatment reduces food intake and body weight in rats possibly through increasing leptin levels, and that this effect of EA is not due to the stress caused by the daily holder restraint.

        COSMOS: A Middleware for Integrated Data Processing over Heterogeneous Sensor Networks

        With the increasing need for intelligent environment monitoring applications and the decreasing cost of manufacturing sensor devices, it is likely that a wide variety of sensor networks will be deployed in the near future. In this environment, the way to access heterogeneous sensor networks and the way to integrate various sensor data are very important. This paper proposes the common system for middleware of sensor networks (COSMOS), which provides integrated data processing over multiple heterogeneous sensor networks based on sensor network abstraction called the sensor network common interface. Specifically, this paper introduces the sensor network common interface which defines a standardized communication protocol and message formats used between the COSMOS and sensor networks.

      • Effects of pH alteration on the pathogenesis of medication-related osteonecrosis of the jaw

        <P><B>Abstract</B></P> <P><B>Introduction</B></P> <P>An acidic environment has been recognized to increase catabolic activities and inhibit osteoblastic deposition, and also exhibited in the pathogenesis of various bone diseases. The aim of the study was to investigate the role of systemic and local pH alteration in the pathogenesis of medication-related osteonecrosis of the jaw (MRONJ).</P> <P><B>Material and methods</B></P> <P>Initially, MRONJ was induced in 54 Sprague-Dawley rats via subcutaneous bisphosphonate injections, once a week for 8 weeks. A week prior to bisphosphonate termination, surgical intervention was performed and rats were divided into 3 groups—alkalotic, acidic and control group, wherein each received NaHCO<SUB>3</SUB>, NH<SUB>4</SUB>Cl and normal saline, respectively for 8 weeks. Upon sacrifice, blood was sent for arterial blood pH analysis and their mandibles were subjected to histomorphometric and μCT analyses. ONJ was histologically defined as necrotic bone persisting for eight weeks after surgical intervention.</P> <P><B>Results</B></P> <P>Each intervention exemplified its expected outcome wherein each group exhibited a borderline alkalotic (7.43 ± 0.05) and acidic state (7.27 ± 37), respectively (<I>P</I> < 0.05). Acidic group showed a higher occurrence of MRONJ (95%) compared to that of alkalotic group (60%) and control (76.9%). Histomorphometric and microstructural evaluation revealed that acidic group presented deteriorated bone architectures with significantly higher necrotic bone fraction, clusters of empty lacunae, N.Oc/B.Pm and lower B.Ar./T.Ar, BV/TV, Tb.Th (<I>P</I> < 0.05). Alkalotic group showed possible protective effects against ONJ versus acidic group, however these trends were not statistically significant.</P> <P><B>Conclusions</B></P> <P>An acidic milieu aggravated ONJ development in an animal model.       • Flow-enhanced electrochemical immunosensors on centrifugal microfluidic platforms.

        <P>We present a novel fully integrated centrifugal microfluidic device with features for target antigen capture from biological samples, via a bead-based enzyme-linked immune-sorbent assay, and flow-enhanced electrochemical detection. The limit of detection (LOD) of our device for the C-reactive protein (CRP) was determined to be 4.9 pg mL(-1), a 17-fold improvement over quantification by optical density. The complete sample-to-answer protocol of our device is fully automated and takes less than 20 min. Overall, the presented microfluidic disc adds to the comparatively small number of fully integrated microfluidic-based platforms that utilize electrochemical detection and exemplifies how electrochemical detection can be enhanced by flow to successfully detect very low levels of biomarkers (e.g. pg mL(-1)).</P>

      • Anisotropic and amphoteric characteristics of diverse carbenes

        <P>Despite its key importance in carbene chemistry, the amphoteric (<I>i.e.</I>, both nucleophilic and electrophilic) behavior of the divalent carbon atom (:C) in carbenes is not well understood. The electrostatic potential (EP) around :C is often incorrectly described by simple isotropic atomic charges (particularly, as in singlet CF2); therefore, it should be described by the multipole model, which can illustrate both negative and positive EPs, favoring the positively and negatively charged species that are often present around :C. This amphotericity is much stronger in the singlet state, which has more conspicuous anisotropic charge distribution than the triplet state; this is validated by the complexation structures of carbenes interacting with Na<SUP>+</SUP>, Cl<SUP>−</SUP>, H2O, and Ag<SUP>+</SUP>. From the study of diverse carbenes [including CH2, CLi2/CNa2, CBe2/CMg2, CF2/CCl2, C(BH2)2/C(AlH2)2, C(CH3)2/C(SiH3)2, C(NH2)2/C(PH2)2, cyclic systems of C(CH2)2/C(CH)2, C(BHCH)2, C(CH2CH)2/C(CHCH)2, and C(NHCH)2/C(NCH)2], we elucidate the relationships between the electron configurations, electron accepting/donating strengths of atoms attached to :C, π conjugation, singlet-triplet energy gaps, anisotropic hard wall radii, anisotropic electrostatic potentials, and amphotericities of carbenes, which are vital to carbene chemistry. The (σ<SUP>2</SUP>, π<SUP>2</SUP> or σπ) electronic configuration associated with :C on the :CA2 plane (where A is an adjacent atom) in singlet and triplet carbenes largely governs the amphoteric behaviors along the :C tip and :C face-on directions. The :C tip and :C face-on sites of σ<SUP>2</SUP> singlet carbenes tend to show negative and positive EPs, favoring nucleophiles and electrophiles, respectively; meanwhile, those of π<SUP>2</SUP> singlet carbenes, such as very highly π-conjugated 5-membered cyclic C(NCH)2, tend to show the opposite behavior. Open-shell σπ singlet (such as highly π-conjugated 5-membered cyclic C(CHCH)2) and triplet carbenes show less anisotropic and amphoteric behaviors.</P>

      • Robust Self-presentation: When and How Do People Present Themselves on Social Networking Sites (SNSs)?

        User of social networking sites (SNSs) often encounter boundary complexity, as users interact with multitude of people of different social groups with diverse interests and perspectives in one common space. User actions (e.g. postings, dialogs, pictures, other online materials) on SNSs are, by nature, shared with a broad audience, even contacts who have incompatible views with the focal user. To act effectively, credibly, and reliably interact with their contacts, users craft their own ways in presenting themselves on SNSs. To explore the behavioral responses to boundary complexity on SNSs, we incorporate the theory on self-presentation and robust action and argue that individuals were able to better achieve a desired presentation of the self through structural reformation of the platform or rhetorical formulation through the platform. We conduct 30 in-depth interviews from those who have used a specific platform (i.e. Instagram). With the interviews, we first specify the bases of boundary complexities into Audience, Interest, and Attention and given this, we classify two processes of boundary management: authentic self-presentation and robust self-presentation. While users of SNSs respond to boundary complexities in various ways to make their self-presentation effective, the methods are based on the ongoing interactions with the platform. SNSs provide particular functions to the users to reconcile issues of boundary complexity. This suggests that the authentic self-presentation is performatively devised through interacting with their platform as well as their contacts.
