        임피던스 방법을 이용한 뇌혈류량 측정기기의 개발(Ⅰ)

        In this study, Rheoencephalography(REG) was designed and constructed. Using the constructed instrument cerebral blood flow(CBF) of left and right brain, CBF change by posture, and reproducibility were studied. Followings are important results. Relative CBF could be measured noninvasively and continuously. Since reproducibility of the constructed REG was found to be quite high (4 %), the accuracy of the instrument itself was proved. REG can be easily applied for testing the effect of drugs for the patients with cerebral vessel diseases since it is quite sensitive to the relative changes of CBF. Various cerebral diseases can be diagnosed by analyzing the REG waveforms. The area of restricted cerebral blood vessels can be located by arranging the electrode configuration. Comparative studies with the standard techniques of measuring CBF are needed to verify the accuracy of REG. Further studies on optimal electrode configuration and various artifacts are also necessary.

        The purpose of this investigation was to develop a new carrier using chitin and glucan as the model for Aspergillus oryzae cell wall. In case of the ratio of chitin and glucan(3/1), the activity of the immobilized protease had higher than that of model(chitin/glucan=1/3) and carriers were formed gel. In case of soluble chitin and glucan treated with calcium carbonate, there were a 1.8-fold increase in activity of immobilized protease compared with untreated chitin. The scanning electron microscope image showed that a pore size of this carrier had a homogenized pore size and a smooth surface.

        To find out the variation of bearing capacity due to the plasticity of soils in case of working the unsymmetrical surcharge of ML, CL, and CH, the existing theoretic expression were investigated and were compared with a series of test results The critical surcharge of soft soils was found as q_(cr)=4.22C_(u) in ML, which is slightly larger than the existing expression, but similar to Darragh's expression. The critical surcharge in CL was found as q_(cr)=3.21C_(u), which is the value between JHI and Jaky and in CH was found as qcr=2.60C_(u), which is a the value between Tachebotarioff and Meyerhof. The ultimate bearing capacity of soft soils was found as q_(uIt) = 11.19C_(u) in ML, which was significantly larger than the existing expression. The ultimate bearing capacity of CL was found as q_(ult)=8.41C_(u), which was slightly bigger than the existing expression, but is close to Meyerhof's expression. The ultimate bearing capacity in CH was found as q_(uIt) =4.73C_(u), which is similar to Parndtl's expression.

      • Both α2,3- and α2,6-Linked Sialic Acids on O-Linked Glycoproteins Act as Functional Receptors for Porcine Sapovirus

        <▼1><P>Sapovirus, a member of the <I>Caliciviridae</I> family, is an important cause of acute gastroenteritis in humans and pigs. Currently, the porcine sapovirus (PSaV) Cowden strain remains the only cultivable member of the <I>Sapovirus</I> genus. While some caliciviruses are known to utilize carbohydrate receptors for entry and infection, a functional receptor for sapovirus is unknown. To characterize the functional receptor of the Cowden strain of PSaV, we undertook a comprehensive series of protein-ligand biochemical assays in mock and PSaV-infected cell culture and/or piglet intestinal tissue sections. PSaV revealed neither hemagglutination activity with red blood cells from any species nor binding activity to synthetic histo-blood group antigens, indicating that PSaV does not use histo-blood group antigens as receptors. Attachment and infection of PSaV were markedly blocked by sialic acid and <I>Vibrio cholerae</I> neuraminidase (NA), suggesting a role for α2,3-linked, α2,6-linked or α2,8-linked sialic acid in virus attachment. However, viral attachment and infection were only partially inhibited by treatment of cells with sialidase S (SS) or <I>Maackia amurensis</I> lectin (MAL), both specific for α2,3-linked sialic acid, or <I>Sambucus nigra</I> lectin (SNL), specific for α2,6-linked sialic acid. These results indicated that PSaV recognizes both α2,3- and α2,6-linked sialic acids for viral attachment and infection. Treatment of cells with proteases or with benzyl 4-O-β-D-galactopyranosyl-β-D-glucopyranoside (benzylGalNAc), which inhibits <I>O</I>-linked glycosylation, also reduced virus binding and infection, whereas inhibition of glycolipd synthesis or <I>N</I>-linked glycosylation had no such effect on virus binding or infection. These data suggest PSaV binds to cellular receptors that consist of α2,3- and α2,6-linked sialic acids on glycoproteins attached via <I>O</I>-linked glycosylation.</P></▼1><▼2><P><B>Author Summary</B></P><P>Although enteropathogenic sapoviruses and noroviruses are leading causes of acute gastroenteritis in both humans and animals, the study of viral pathogenesis and immunity of these ubiquitous pathogens has been hampered due to the lack of a fully permissive cell culture system. Porcine sapovirus Cowden strain provides a suitable system that can be used to identify the molecular mechanisms of viral pathogenesis. Previous studies have shown that carbohydrates and glycolipids play important roles in the attachment of members of the <I>Caliciviridae</I>; histo-blood group antigens (HBGAs) are used by <I>Norovirus</I> genogroups I to IV, as well as members of the <I>Lagovirus</I>, and <I>Recovirus</I> genera, whereas terminal sialic acid is recognized as a receptor for feline calicivirus and murine norovirus. To date, however, the role of carbohydrates in the life cycle of sapoviruses has remained largely unknown. We found that porcine sapovirus binds to susceptible host cells through both α2,3- and α2,6-linked terminal sialic acids which are attached to <I>O</I>-linked glycoproteins. These efforts, findings and insights will significantly contribute to a better understanding of the sapovirus life cycle.</P></▼2>

      • Detection of Potato virus Y (PVY) from a Single Aphid by One-Step Rt-Pcr Using Boiling Extraction Method

        PVY (Potyviridae: potyvirus) is one of the most important potato virus affecting seed potato production and also it is transmitted non-persistently via aphids. For healthy seed potato production, a virus detection system is highly important in addition to aphid monitoring and control. To achieve this detection method, it need to fast and easy to use. About two decades ago RT-PCR based PVY detection method was developed. However that was very time consuming and has low sensitivity. Here, we developed an advanced PVY detection method which a uses the boiling extraction of the viral RNA from aphid stylet and amplification by specific primers located in the viral capsid protein gene. Therefore, it could directly synthesize cDNA of PVY viral capsid gene from extracted RNA of PVY using one-step RT-PCR method in very short time compared to previous methods due to the omission of RNA extraction step. We confirmed this PVY detection method using the two aphid species (Macrosiphum euphorbiae and Aphis gossypii) that known as PVY vectors. The efficiency of this PVY detection method was 60% to 80% from two the aphid species. Hence, this method could be potentially applied to virus free seed potato production programs.

        Intestinal Nematodes from Small Mammals Captured near the Demilitarized Zone, Gyeonggi Province, Republic of Korea

        A total of 1,708 small mammals (1,617 rodents and 91 soricomorphs), including Apodemus agrarius (n = 1,400), Microtus fortis (167), Crocidura lasiura (91), Mus musculus (32), Myodes (= Eothenomys) regulus (9), Micromys minutus (6), and Tscherskia (= Cricetulus) triton (3), were live-trapped at US/Republic of Korea (ROK) military training sites near the demilitarized zone (DMZ) of Paju, Pocheon, and Yeoncheon, Gyeonggi Province from December 2004 to December 2009. Small mammals were examined for their intestinal nematodes by necropsy. A total of 1,617 rodents (100%) and 91 (100%) soricomorphs were infected with at least 1 nematode species, including Nippostrongylus brasiliensis, Heligmosomoides polygyrus, Syphacia obvelata, Heterakis spumosa, Protospirura muris, Capillaria spp., Trichuris muris, Rictularia affinis, and an unidentified species. N. brasiliensis was the most common species infecting small mammals (1,060; 62.1%) followed by H. polygyrus (617; 36.1%), S. obvelata (370; 21.7%), H. spumosa (314; 18.4%), P. muris (123; 7.2%), and Capillaria spp. (59; 3.5%). Low infection rates (0.1-0.8%) were observed for T. muris, R. affinis, and an unidentified species. The number of recovered worms was highest for N. brasiliensis (21,623 worms; mean 20.4 worms/infected specimen) followed by S. obvelata (9,235; 25.0 worms), H. polygyrus (4,122; 6.7 worms), and H. spumosa (1,160; 3.7 worms). A. agrarius demonstrated the highest prevalence for N. brasiliensis (70.9%), followed by M. minutus (50.0%), T. triton (33.3%), M. fortis (28.1%), M. musculus (15.6%), C. lasiura (13.2%), and M. regulus (0%). This is the first report of nematode infections in small mammals captured near the DMZ in ROK.

        Absorbable Plate as a Perpendicular Strut for Acute Saddle Nose Deformities

        Background : Nasal pyramid fractures accompanied by saddle nose deformities are not easily corrected by closed reduction. We used an absorbable plate as a perpendicular strut to support the collapsed "keystone area" and obtained good results. Methods : Between September 2008 and June 2011, 18 patients who had nasal pyramid fractures with saddle nose deformities underwent surgery. Pre- and postoperative facial computed tomographic images and photographs were taken to estimate outcomes. The operative technique included the mucoperichondrial dissection of the nasal septum, insertion of an absorbable plate prepared to an appropriate length to support the "keystone area", and fixation of the absorbable plate strut to the cartilaginous septum. Results : Functional and esthetic outcomes were satisfactory in all patients. Eleven patients assessed the postoperative appearance of the external nose as 'markedly improved' and 7 patients as 'improved'. The 5 surgeons scored the results as a mean of 4.5 on a 5-point scale. Conclusions : The use of an absorbable plate as a perpendicular strut requires no additional procedures because the plate is gradually absorbed. The mechanical strength provided by a buttress between the "keystone area" and the maxillary crest lasts for a long time before the strut is absorbed.

        Outcomes of deceased donor liver transplantation from elderly donors

        Background: Favorable outcomes achieved after deceased donor liver transplantation (DDLT) suggest that use of elderly donors may be an effective way to expand donor pool. Methods: This was a retrospective analysis of adult DDLT using elderly donors. It was a double-arm study that compared posttransplant outcomes to ascertain whether use of elderly donors (aged ≥76 years) has adverse effects on outcome of DDLT. Elderly donor study group included 14 donors aged ≥76 years and elderly donor control group comprised 39 donors aged 66–75 years. Results: Mean donor age of the study and control groups was 78.2±3.1 years and 68.9±2.7 years, respectively (P<0.001). Other clinical parameters were comparable between these two donor groups. The 1-, 3-, and 5-year graft survival rates in the elderly study group were 83.6%, 59.7%, and 59.7%, respectively, and those in the elderly control group were 79.4%, 68.1%, and 59.6%, respectively (P=0.97). The overall 1-, 3-, and 5-year survival rates after donation from elderly study group were 83.6%, 59.7%, and 59.7%, respectively, and those after donation from control group were 79.3%, 72.1%, and 64.1%, respectively (P=0.74). Regarding overall patient survival, univariate analysis identified pretransplant requirement for ventilator support (P=0.021) and pretransplant renal replacement therapy (P=0.025) as statistically significant risk factors; however, neither was significant on multivariate analysis. Conclusions: The results of this study suggest that using an elderly donor graft might not worsen the posttransplant outcomes significantly; thus, advanced age per se may not be an exclusion criterion for organ donation.

The objective of our study was to figure out the ideal conditions which minimize damages of quality of [$^{18}F$]FDG using radical scavenger and controlling temperature of storage.
