Neuraminidase Treatment Enhances Allogeneic Stimulation of Unprimed CD8+ T Cells

Many cell types are known to stimulate CD8^+ T cells in allogeneic recognition such as mixed lymphocyte reaction (MLR). Whereas dendritic cells are most potent among them. T cells are usually considered very poor in stimulating CD8^+ T cells although there are some tumor cells that are weakly stimulatory. T cells, as a stimulator. cultured in the presence of concanavalin A that were otherwise nonstimulatory to CD8^+ T cells appeared to stimulate CD8^+ T cells strongly when they were pretreated with neuraminidase. The enhancement of MLR by neuraminidase could be achieved by treating either the stimulators or responders with neuraminidase. Removal of negatively-charged sialic acid moieties from the cell surface, which reduced electrostatic repulsion between responders and stimulators to give better cell-cell contact might be responsible for the enhanced MLR. In addition. neuraminidase treatment also appeared to deliver activation signal to responding T cells since it could activate CD8^+ T cells in synergy with phorbol myristate acetate. The maximal responses were observed when both responders and stimulators were treated with neuraminidase.
Differential Activation of T Cells by T - Cell Receptor Ligand Analogs

Although CD4 T cell responses to protein-derived antigen have well been understood, the epitopes recognized by hapten-specific CD4 T cells have not been fully defined. In this study, we characterized the response of a T cell hybridoma (5Di0.1B8) which is specific for a hapten. Nhydroxysuccinimidyl-4-azidobenzoate (HSAB) restricted by MHC class II I-A^d. Using three different antigen presenting cells (APCs) expressing I-A^d. the role of class II MHC proteins in haptenic antigen presentation and subsequent activation of 5D10.1B8 has been examined. Activation of 5D10.1B8 T cells by HSAB analogs was also performed. Our results show that each APC activated T cells differentially and that interleukin-2 (IL2) augmented antigen-presenting ability of all the APCs, suggesting that increased expression of class II MHC protein by IL-2 played an important role in HSAB presentation and T cell activation. Finally, early T cell receptor-dependent signals induced by HSAB or its analogs were examined by phosphotyrosine immunoblot analysis, and showed that tyrosine phosphorylation level of a 18-20 kD protein increased upon stimulation.

The purpose of this study was to establish an in vitro culture method of tumor-specific T cells, and determine the efficacy of the cultured tumor-specific cytotoxic T-lymphocytes (CTL) as an agent of anti-tumor immunotherapy against a murine lymphoma, TIMI.4. Tumor-specific T-lymphocytes derived from C57BL/6 mice (thy-1.2) immune to TIMI.4 were activated by in vitro stimulation with the irradiated TIMI.4 cells, and expanded by restimulation with TIMI.4 in the presence of the concanavalin A-stimulated rat spleen culture supernatant, and splenic antigen-presenting cells. In vitro restimulation enhanced markedly the proportion of $CD8^+$, a predominant surface marker of CTL and the cytotoxic activity in the cultured immune T cell population. The resulting TIMI.4-specific T cells were adoptively transferred into nude mice. The tumor cells residing in the host after 7 days of adoptive transfer to B6.PL (thy-1.1) mice were quantified by use of an antibody directed to the thy-1.2 allele. The TIMI.4 cells in the recipient nude mice were decreased in a dose-dependent manner. Anti-tumor activity of the TIMI.4-specific T cells was also demonstrated by a survival test, where the tumor-bearing nu/nu mice which received the activated T-cells survived about 30% longer than the control mice which received the tumor cells alone. These suggest that adoptive transfer of TIMI.4-specific T cells could be a candidate for effective therapy of the murine lymphoma.

Two component polyurethane (PU) flame retardant coatings were prepared by blending trichloro modified polyesters (TCMPs) and isophorone diisocyanate isocyanurate. TCMPs were synthesized by polycondensation of trichlorobenzoic acid (TCBA), a flame retardant component, with adipic acid, 1,4 butanediol, and trimethylolpropane. The content of TCBA was varied in 10, 20, and 30 wt% for the reaction. Theses new flame retardant coatings showed various properties comparable to other non flame retardant coatings. Moreover, we carried out the combustion test and the flammability test for our flame retardant coatings. The results of vertical burning test for the coatings containing more than 20 wt% of TCBA were determined as no burn. The results of flammability test for the coatings with 20 wt% and 30 wt% of TCBA contents indicated the limiting oxygen index (LOI) values of 26% and 29% respectively, which implied relatively good flame retardancy.
Ferromagnetism and p-type Conductivity in Laser-deposited (Zn,Mn)O Thin Films Codoped by Mg and P

We report on the observation of p-type conductivity and ferromagnetism in diluted magnetic semiconductor (Zn0.97Mg0.01Mn0.02)O:P films grown on SiO₂/Si substrates by pulsed laser deposition. The p-type conduction with hole concentration over 1018 ㎝<SUP>?3</SUP> is obtained by codoping of Mg and P followed by rapid thermal annealing in an O₂atmosphere. Structural and compositional analyses for the p-type (Zn0.97Mg0.01Mn0.02)O:P films annealed at 800 ℃ indicates that highly c-axis oriented homogeneous films were grown without any detectable formation of secondary phases. The films were found to be transparent in the visible range. The magnetic measurements clearly revealed an enhancement of room temperature ferromagnetism by p-type doping.
Effect of Tin Codoping on Transport and Magnetic Properties of Chromium-doped Indium Oxide Films

This study examined the effect of Sn co-doping on the transport and magnetic properties of Cr-doped In₂O₃ thin films grown on (100) silicon substrates by pulsed laser deposition. The experimental results showed that Sn co-doping enhances the magnetization and appearance of the anomalous Hall effect, and increases the carrier (electron) concentration. These results suggest that the conduction carrier plays an important role in enhancing the ferromagnetism of a laser-deposited Cr-doped In₂O₃ film, which may have applications in transparent oxide semiconductor spin electronics devices.

Leiomyosarcoma (LMS) of the small intestine is a rare tumor, accounting for about 1% of all malignant mesenchymal lesions in the gastrointestinal tract. Since small bowel tumors are initially asymptomatic and nonspecific, delayed diagnosis and treatment are common. We found that a 44-year-old male patient who came in for lower abdominal pain had partial obstruction in the small bowel. Multiple ascites and ileal tumors involving peritoneal seeding were observed from his abdominal computed tomography. He was diagnosed as epithelioid LMS involving peritoneal transition after surgical resection, and the outpatient department has planned for a conservative therapy with observation. To the best of our knowledge, ileal epithelioid LMS accompanying a huge omental mass, with a size of 18 cm in dimension, and peritoneal seeding has not yet been reported in Korea. We report this rare case with literature review.
