Sarcopenia represents the progressive loss of skeletal muscles, which occurs as a result of aging. Plant-derived phytochemicals have the potential ability to manage sarcopenial conditions. The randomized, placebo-controlled, double-blind clinical study involving thirty subjects evaluated the efficacy of Cureit™ supplementation in the management of sarcopenial conditions by measuring the variables, such as hand grip strength, weight lift strength, time/distance before feeling tired after cycling, walking and climbing stairs, and Karnofsky performance scale index along with effects on general fitness, such as protein, urea, oxidative stress, and hematology parameters. The results showed that Cureit supplementation resulted in a significant increase of 1.43% (P < .001) in the handgrip strength compared with placebo. The weight-lifting capacity of subjects supplemented with Cureit showed an increase of 6.08%, whereas placebo showed a 4.54% decrease after the end of the study period. The results demonstrated that the Cureit tended to have a positive impact on distance covered before feeling tired as shown by an increase (P = .09) of 5.51%, compared with placebo group, which showed an increase of 2.29%. The time taken to walk the same distance was reduced in the Cureit group (1.15%), whereas in the placebo group, it was increased (2.02%). Cureit plays a significant role in the management of sarcopenia by anti-inflammatory action, increased hand grip strength, antifatigue effects, and muscle protein management. BackgroundPlasma cell leukemia (PCL) is a rare and aggressive plasma cell neoplasm. In PCL, clonal plasma cells comprise ≥20% of the peripheral blood (PB) leukocytes and/or the absolute clonal PB plasma cell count is ≥2×109/L. Primary PCL (PPCL) originates de novo, whereas, secondary PCL (SPCL) evolves from pre-existing multiple myeloma.MethodsClinicohematological features, immunophenotypic profile, and survival of PCL patients were analyzed retrospectively.ResultsBetween January 2007 and December 2014, ten PPCL and four SPCL patients were inves-tigated (8 PPCLs and 3 SPCLs had complete clinical data). All were North Indians, sharing common geography and ethnicity. Our cohort showed less frequent renal failure, more frequent hepatomegaly, and non-secretory type disease. In contrast to western literature, flow cytometric immunophenotyping of our cohort revealed altered expression of CD138 (67%), CD56 (33%), and CD20 (0%). With novel therapeutic agents, these PPCL patients had a median overall survival of 15 months.ConclusionWe highlight that our PPCL patients from North India had distinct clinicohematological and immunophenotypic profiles. The significance of our findings must be tested in a larger patient cohort and must be supported by molecular and cytogenetic investigations to un-mask possible significant effects on pathogenesis.

Rheumatoid arthritis (RA) is an autoimmune, chronic systemic inflammatory disorder. The long-term use of currently available drugs for the treatment of RA has many potential side effects. Natural phytonutrients may serve as alternative strategies for the safe and effective treatment of RA, and curcuminoids have been used in Ayurvedic medicine for the treatment of inflammatory conditions for centuries. In this study, a novel, highly bioavailable form of curcumin in a completely natural turmeric matrix was evaluated for its ability to improve the clinical symptoms of RA. A randomized, double-blind, placebo-controlled, three-arm, parallel-group study was conducted to evaluate the comparative efficacy of two different doses of curcumin with that of a placebo in active RA patients. Twelve patients in each group received placebo, 250 or 500 mg of the curcumin product twice daily for 90 days. The responses of the patients were assessed using the American College of Rheumatology (ACR) response, visual analog scale (VAS), C-reactive protein (CRP), Disease Activity Score 28 (DAS28), erythrocyte sedimentation rate (ESR), and rheumatoid factor (RF) values. RA patients who received the curcumin product at both low and high doses reported statistically significant changes in their clinical symptoms at the end of the study. These observations were confirmed by significant changes in ESR, CPR, and RF values in patients receiving the study product compared to baseline and placebo. The results indicate that this novel curcumin in a turmeric matrix acts as an analgesic and anti-inflammatory agent for the management of RA at a dose as low as 250 mg twice daily as evidenced by significant improvement in the ESR, CRP, VAS, RF, DAS28, and ACR responses compared to placebo. Both doses of the study product were well tolerated and without side effects.
