As the majority of side effects of current chemotherapies stems from toxicity due to excessive dosing of anticancer drugs, minimizing the amount of drug while maximizing drug efficacy is essential to increase the life-quality of chemotherapy patients. This study demonstrated that the intracellular delivery of amide linked doxorubicin on carbon nanotube can nullify the efflux of cancer cells by achieving prolonged endolysosome delivery and can induce burst release of doxorubicin in an acidic hydrolase environment and, ultimately, can reduce the amount of anticancer drug by 10-fold compared to conventional effective drug dose. The clearance of accumulated carbon nanotubes in the liver was observed after 4 weeks, and analysis of liver toxicity markers showed no significant changes in GOT and GPT levels and release of pro-inflammatory cytokines across both short- and long-term periods.

Graphic Abstract

ACS Electronic Supporting Info

A rapid antibiotic susceptibility test (AST) is desperately needed in clinical settings for fast and appropriate antibiotic administration. Traditional ASTs, which rely on cell culture, are not suitable for urgent cases of bacterial infection and antibiotic resistance owing to their relatively long test times. We describe a novel AST called single-cell morphological analysis (SCMA) that can determine antimicrobial susceptibility by automatically analyzing and categorizing morphological changes in single bacterial cells under various antimicrobial conditions. The SCMA was tested with four Clinical and Laboratory Standards Institute standard bacterial strains and 189 clinical samples, including extended-spectrum β-lactamase–positive Escherichia coli and Klebsiella pneumoniae, imipenem-resistant Pseudomonas aeruginosa, methicillin-resistant Staphylococcus aureus, and vancomycin-resistant Enterococci from hospitals. The results were compared with the gold standard broth microdilution test. The SCMA results were obtained in less than 4 hours, with 91.5% categorical agreement and 6.51% minor, 2.56% major, and 1.49% very major discrepancies. Thus, SCMA provides rapid and accurate antimicrobial susceptibility data that satisfy the recommended performance of the U.S. Food and Drug Administration.

Wall-modeled LES (WMLES) is promising to simulate a turbulent flow at high Reynolds number with a reasonable cost because the required computational cost of large-eddy simulation in the near-wall region is proportional to approximately square of the friction Reynolds number (Reτ). An equilibrium stress model is the most widely used method due to the high efficiency. However, this method is still required to enhance the accuracy and applicability because of the limitations of the equilibrium assumption. In the present study, an artificial neural network (ANN) is used to obtain the wall shear stress for WMLES. The proposed method shows good prediction on the profiles of mean velocity and Reynolds stresses compared to equilibrium model when direct numerical simulation dataset of turbulent channel flows is trained at Reτ=395, 540, 930 and 2000. In addition, the present method predicts well the turbulent statistics at Reτ=640 and 5200, which are untrained by the ANN.

In this study, we report the optimal extraction conditions for obtaining organosulfur compounds, such as cycloalliin, from garlic by using principal component analysis (PCA). Extraction variables including temperature ($40{\sim}80^{\circ}C$), time (0.5~12 h), and pH (4~12) were investigated for the highest cycloalliin yields. The cycloalliin yield (5.5 mmol/mL) at pH 10 was enhanced by ~40% relative to those (~3.9 mmol/mL) at pH 4 and pH 6. The cycloalliin level at $80^{\circ}C$ showed the highest yield among the tested temperatures (5.05 mmol/mL). Prolonged extraction times also increased cycloalliin yield; the yield after 12 h was enhanced ~2-fold (4 mmol/mL) compared to the control. Isoalliin and cycloalliin levels were inversely correlated, whereas a direct correlation between polyphenol and cycloalliin levels was observed. In storage for 30 days, garlic stored at $60^{\circ}C$ (11 mmol/mL) showed higher levels of cycloalliin and polyphenols than those at $40^{\circ}C$, with the maximum cycloalliin level (13 mmol/mL) on day 15. Based on the PCA analysis, the isoalliin level depended on the extraction time, while cycloalliin amounts were influenced not only by extraction time, but also by pH and temperature. Taken together, extraction of garlic at $80^{\circ}C$, with an incubation time of 12 h, at pH 10 afforded the maximum yield of cycloalliin.

Purpose: The purpose of this study is to quantitatively identify the characteristics of lower extremity movements before and after taping during the closed change step motion in waltz for dance sports experts by using the functional taping, which is widely known for the prevention and treatment of the sports injuries, and also provide coaches and athletes with the appropriate posture and injury prevention by offering the basic data for the technical performance. Method: As for the subjects of this study, 8 female experts with 5 or more years of experience in dance sports in the metropolitan city of B were selected and experimented. Prior to entering the experiment, a consent was secured from each subject, and the method of the experiment was explained, while the kinematic data were collected by using 5 digital cameras. As for the statistical processing of this study, the joint angle data were calculated before and after taping during the closed change step motion by using the SPSS 24.0. First, the mean and standard deviation were obtained, and in order to analyze the difference before and after taping, and time and phase, a corresponding sample t-test was conducted. The level of significance was set to α=.05. Results: As a result of the experiment, it turned out that taping did stabilize the joint balance rather than causing problems for the mobility of the lower extremity joints via a biomechanical analysis of the lower extremities during the closed change step in motion waltz, and it is determined that it may have significance in that it provided the basic data required for the injury prevention as well as for the efficient performance of movements. Conclusion: Gathering the conclusions as such, it is determined that taping may have significance in that it stabilizes the joint balance during the closed change step motion in waltz and provides the basic data required for the efficient performance of movement and the injury prevention.

This research was carried out to develop the Hanji(Korean paper) sludge ·wood particle composite utilizing the waste sludges occurring from the making process of Hanji(Korean paper). At the research, four mixing ratios of white or black sludge to wood particle(10:90, 20:80, 30:70 and 40:60), three types of the resin adhesives(PMDI, urea and phenol resin) and three levels of the densities(0.60, 0.75 and 0.90) were designed to investigate the mechanical properties of Hanji(Korean paper) sludge ·wood particle composite. In the white and black sludge ·wood particle composites, bending properties(MOR, MOE) showed the decreasing tendency according to the increase of sludge additive, but it was clearly increased with the increase of specific gravity. Also tensile strength had the same tendency as in these bending properties. The internal bond strength of white sludge ·wood particle composite had no tendency, but that of black sludge ·wood particle composite was decreased as an increase of Hanji sludge additive.

Transmembrane protein 165 (TMEM165), a Golgi protein, functions in ion homeostasis and vesicular trafficking in the Golgi apparatus. While mutations in TMEM165 are known to cause human ‘congenital disorders of glycosylation’, a recessive autosomal metabolic disease, the potential association of this protein with human cancer development has not been explored to date. In the present study, we revealed that TMEM165 is overexpressed in HCC and its depletion weakens the invasive activity of cancer cells through suppression of matrix metalloproteinase-2 (MMP-2) expression. Levels of TMEM165 mRNA and protein were clearly increased in HCC patient tissues and cell cultures. Quantitative real-time RT-PCR analysis of fresh HCC tissues (n=88) revealed association of TMEM165 overexpression with more frequent macroscopic vascular invasion, microscopic serosal invasion and higher α-fetoprotein levels. Notably, depletion of TMEM165 led to a marked decrease in the invasive activity of two different HCC cell types, Huh7 and SNU475, accompanied by downregulation of MMP-2. Our collective findings clearly indicated that TMEM165 contributed to the progression of HCC by promoting invasive activity, supporting its utility as a novel biomarker and therapeutic target for cancer.

Automatic Name Line Detection for Person Indexing Based on Overlay Text
Lee, Sanghee,Ahn, Jungil,Jo, Kanghyun Korea Multimedia Society 2015 The journal of multimedia information system Vol.2 No.1
Many overlay texts are artificially superimposed on the broadcasting videos by humans. These texts provide additional information to the audiovisual content. Especially, the overlay text in news videos contains concise and direct description of the content. Therefore, it is most reliable clue for constructing a news video indexing system. To make the automatic person indexing of interview video in the TV news program, this paper proposes the method to only detect the name text line among the whole overlay texts in one frame. The experimental results on Korean television news videos show that the proposed framework efficiently detects the overlaid name text line.
