        Dynamic network biomarker identifies cdkn1a-mediated bone mineralization in the triggering phase of osteoporosis

        The identification of predictive markers to determine the triggering phase prior to the onset of osteoporosis is essential to mitigate further irrevocable deterioration. To determine the early warning signs before osteoporosis, we used the dynamic network biomarker (DNB) approach to analyze time-series gene expression data in a zebrafish osteoporosis model, which revealed that cyclin-dependent kinase inhibitor 1 A (cdkn1a) is a core DNB. We found that cdkn1a negatively regulates osteogenesis, as evidenced by loss-of-function and gain-of-function studies. Specifically, CRISPR/Cas9-mediated cdkn1a knockout in zebrafish significantly altered skeletal development and increased bone mineralization, whereas inducible cdkn1a expression significantly contributed to osteoclast differentiation. We also found several mechanistic clues that cdkn1a participates in osteoclast differentiation by regulating its upstream signaling cascades. To summarize, in this study, we provided new insights into the dynamic nature of osteoporosis and identified cdkn1a as an early-warning signal of osteoporosis onset.

      • Lack of Association between the COMT rs4680 Polymorphism and Ovarian Cancer Risk: Evidence from a Meta-analysis of 3,940 Individuals

        Catechol-O-methyltransferase (COMT) is involved in estrogen metabolism and is vital to estrogen-induced carcinogenesis, including that of ovarian cancer. Although many recent epidemiologic studies have investigated associations between the COMT rs4680 polymorphism and ovarian cancer risk, the results remain inconclusive. We therefore performed a meta-analysis to derive a more precise estimate of associations. Systematic searches of the PubMed, Embase, Web of Science, Cochrane Library, Wanfang, China National Knowledge Infrastructure, and Chinese Biomedicine databases were undertaken to retrieve eligible studies. Odds ratios (ORs) with their corresponding 95% confidence intervals (CIs) were pooled to assess the strength of the association. In total, 8 case-control studies involving 1,293 cases and 2,647 controls were included in the meta-analysis. Overall, the results showed no evidence of significant association between the COMT rs4680 polymorphism and ovarian cancer risk in any of the assessed genetic models. Subgroup analyses by ethnicity also did not reveal any significant association in any genetic model (p>0.05). In conclusion, our findings suggest that the COMT rs4680 polymorphism may not contribute to the risk of ovarian cancer.

        Over-expression of GmHAL3 modulates salt stresses tolerance in transgenic arabidopsis

        The halotolerance protein HAL3, also known as SIS2, is a yeast protein that regulates the cell cycle and tolerance to salt stress through inhibition of the Ppz1 type 1 protein phosphatase. Although the roles of HAL3 have been demonstrated during the growth, development, and stress adaptation of Arabidopsis thaliana and Nicotiana tabacum, the function of HAL3 in other plant species, including soybean (Glycine max), has not been elucidated. In this study, GmHAL3a and GmHAL3b were isolated from Glycine max, and their roles were analyzed. GmHAL3a and GmHAL3b transcripts were detected in the roots, stems, leaves and seeds, with higher levels in the roots, and were induced by sodium chloride (NaCl), lithium chloride (LiCl), sorbitol, cold and ABA treatment. Overexpression of GmHAL3a or GmHAL3b in Arabidopsis accelerated the onset of flowering and resulted in more vigorous seed germination and increased tolerance to NaCl, LiCl, and sorbitol stress in seedlings, compared with wild type (WT) and empty vector control (VC) plants. Transgenic Arabidopsis plants accumulated proline and eliminated superoxide radical (O2 −) in response to the stress. In addition, transcription levels of the stress-related genes RD22 and P5CS1 were substantially higher in transgenic Arabidopsis than in WT and VC plants. Taken together, the data indicate that GmHAL functions as a positive regulator of the response to salt, lithium cations and sorbitol stress.

      • Lack of Associations of the COMT Val158Met Polymorphism with Risk of Endometrial and Ovarian Cancer: a Pooled Analysis of Case-control Studies

        This meta-analysis was conducted to examine whether the genotype status of Val158Met polymorphism in catechol-O-methyltransferase (COMT) is associated with endometrial and ovarian cancer risk. Eligible studies were identified by searching several databases for relevant reports published before January 1, 2014. Pooled odds ratios (ORs) were appropriately derived from fixed-effects or random-effects models. In total, 15 studies (1,293 cases and 2,647 controls for ovarian cancer and 2,174 cases and 2,699 controls for endometrial cancer) were included in the present meta-analysis. When all studies were pooled into the meta-analysis, there was no evidence for significant association between COMT Val158Met polymorphism and ovarian cancer risk (Val/Met versus Val/Val: OR=0.91, 95% CI=0.76-1.08; Met/Met versus Val/Val: OR=0.90, 95% CI=0.73-1.10; dominant model: OR=0.90, 95% CI=0.77-1.06; recessive model: OR=0.95, 95% CI=0.80-1.13). Similarly, no associations were found in all comparisons for endometrial cancer (Val/Met versus Val/Val: OR 0.97, 95% CI=0.77-1.21; Met/Met versus Val/Val: OR=1.02, 95% CI=0.73-1.42; dominant model: OR=0.98, 95% CI=0.77-1.25; recessive model: OR=1.02, 95% CI=0.87-1.20). In the subgroup analyses by source of control and ethnicity, no significant associations were found in any subgroup of population. This meta-analysis strongly suggests that COMT Val158Met polymorphism is not associated with increased endometrial and ovarian cancer risk.

        In the present drilling materials supplies management informationization level is low, which lead to low efficiency of management in the Chinese oil enterprises. This paper presents an effective solution combined with RFID and web technology for oil enterprise information service platform system, and studies several key technologies in the design. The system has taken the advantages of RFID and web services in the practical application process, the enterprises can use this RFID solution to enhance the logistics management efficiency in the oil-drilling materials supplies. At the end of the paper we proposed technical barriers is one of the reasons for RFID popularization.

      • PEALD 법에 의한 TiO₂박막의 상형성 및 전기적 특성

        플라즈마를 이용한 원자층 증착법 (PEALD) 으로 TDMA T 와 산소 플라즈마를 사용하여 균일하고 등방성한 TiO₂박막을 제조하였다. Cycle 당 증착율은 0.36 A 으로 공정 cycle 의 증가에 상관없이 일정하게 나타났으며, 표면 자기제어 반응에 의해 우수한 step coverage 특성 올 얻을 수 있었다. 적절한 유전성과 좋은 누설전류 특성을 연구하기 위해 PEALD 법으로 증착한 박막을 급속열처리 하였다. 열처리 온도가 증가함에 따라 TiO₂박막의 누설전류 특성은 향상되었고, 반면에 유전율은 약간 감소하는 것으로 나타났다 . 결론적으로 PEALD 법으로 제조된 TiO₂박막은 다양한 온도에서의 열처리를 통해 우수한 전기적 특성을 가진 유전체 박막임올 알 수 있었다.

      • Lectin from Agrocybe aegerita as a Glycophenotype Probe for Evaluation of Progression and Survival in Colorectal Cancer

        Background: Agrocybe aegerita Lectin (AAL) has been identified to have high affinity for sulfated and ${\alpha}2$-3-linked sialic acid glycoconjugates, especially the sulfated and sialyl TF (Thomsen-Friedenreich) disaccharide. This study was conducted to investigate the clinicopathological and prognostic value of AAL in identifying aberrant glycosylation in colorectal cancer (CRC). Materials and Methods: Glycoconjugate expression in 59 CRC tissues were detected using AAL-histochemistry. Clinicopathological associates of expression were analyzed with chisquare test or Fisher's exact test. Relationships between expression and the various clinicopathological parameters was estimated using Kaplan-Meier analysis and Cox regression models. Results: AAL specific glycoconjugate expression was significantly higher in tumor than corresponding normal tissues (66.1% and 46.1%, respectively, p=0.037), correlating with depth of invasion (p=0.015) and TNM stage (p=0.024). Patients with lower expression levels had a significantly higher survival rate than those with higher expression (p=0.046 by log rank test and p=0.047 by Breslow test for overall survival; p=0.054 by log rank test and P=0.038 by Breslow test for progress free survival). A marginally significant association was found between AAL specific glycoconjugate expression and overall survival by univariate Cox regression analysis (p=0.059). Conclusions: Lower AAL specific glycoconjugate expression is a significant favorable prognostic factor for overall and progress free survival in CRC. This is the first report about the employment of AAL for histochemical analysis of cancer tissues. The binding characteristics of AAL means it has potential to become a powerful tool for the glycan investigation and clinical application.

        Metabolic Engineering of Probiotic <i>Saccharomyces boulardii</i>

        <P>Saccharomyces boulardii is a probiotic yeast that has been used for promoting gut health as well as preventing diarrheal diseases. This yeast not only exhibits beneficial phenotypes for gut health but also can stay longer in the gut than Saccharomyces cerevisiae. Therefore, S. boulardii is an attractive host for metabolic engineering to produce biomolecules of interest in the gut. However, the lack of auxotrophic strains with defined genetic backgrounds has hampered the use of this strain for metabolic engineering. Here, we report the development of well-defined auxotrophic mutants (leu2, ura3, his3, and trp1) through clustered regularly interspaced short palindromic repeat (CRISPR)-Cas9-based genome editing. The resulting auxotrophic mutants can be used as a host for introducing various genetic perturbations, such as overexpression or deletion of a target gene, using existing genetic tools for S. cerevisiae. We demonstrated the overexpression of a heterologous gene (lacZ), the correct localization of a target protein (red fluorescent protein) into mitochondria by using a protein localization signal, and the introduction of a heterologous metabolic pathway (xylose-assimilating pathway) in the genome of S. boulardii. We further demonstrated that human lysozyme, which is beneficial for human gut health, could be secreted by S. boulardii. Our results suggest that more sophisticated genetic perturbations to improve S. boulardii can be performed without using a drug resistance marker, which is a prerequisite for in vivo applications using engineered S. boulardii.</P>

        Experimental research and molecular dynamics simulation on thermal properties of capric acid/ethylene-vinyl/graphene composite phase change materials

        In this paper, a new series of phase change materials (PCMs) composed of capric acid/ethylene-vinylacetate/graphene (CA/EVA/GR) were prepared and thermal properties were investigated using moleculardynamics simulation. The composite PCMs were characterized by Fourier transform infraredspectroscopy (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM), differential scanningcalorimetry (DSC) and thermal conductivity measurement. FT-IR, XRD and SEM results manifest that CAcan be successfully wrapped by EVA and GR additives, and there is no chemical reaction between CA, EVAand GR. DSC results indicate that adding GR into CA/EVA can result in composite PCMs maintain the highlatent heat, while too much GR will cause a significant reduction in latent heat. Thermal conductivityobtained from experimental tests reveal that GR can gradually enhance the thermal conductivity of CA/EVA with increasing dosage of GR. The experimental results of thermal conductivity fall close to that ofmolecular dynamics (MD) simulation at GR dosages below 1.8 wt%, but the experimental results present atrend which is contrary to MD simulation at higher GR dosages. The mean square displacement (MSD)results manifest that composite PCMs containing 1.8 wt% GR has the highest diffusion coefficient, whilehigher GR dosage will reduce the diffusion coefficient gradually.

        Effects of Cu (II)-exchanged Montmorillonite on Growth Performance, Intestinal Microflora, Bacterial Enzyme Activities and Morphology of Broilers

        Two hundred forty 1-d-old Arbor Acres broiler chicks were used to investigate the effects of Cu (II)-exchanged montmorillonite (CEM) or montmorillonite on the growth performance, intestinal microflora, bacterial enzyme activities and morphology of broilers. The chicks were assigned randomly into three groups with 80 chicks per treatment. The three dietary treatments were basal diet only (control group), basal diet +1 g $kg^{-1}$ montmorillonite, and basal diet +1 g $kg^{-1}$ CEM. The results showed that the addition of CEM to the diet increased significantly the body weight and feed efficiency, but a similarly significant increase was not found in broilers fed the diet containing montmorillonite. Supplementing the CEM in the diet of broilers also decreased the numbers of Clostridium perfringens and Escherichia coli in the small intestine and cecum. The addition of either CEM or montmorillonite to the diet depressed the activities of $\beta$-glucosidase and $\beta$-glucuronidase in the small intestinal and cecal contents. Data of villus height and crypt depth for duodenum, jejunum and ileum indicated that dietary addition of CEM or montmorillonite improved the small intestinal mucosal morphology.
