        톱밥 발효사료 급여 및 분말어유의 첨가사료가 돈육의 이화학적 성질에 미치는 영향

        This study was conducted to investigate the influence of pork quality pigs which were fed with diets supplemented with fermented sawdust and sardin powder oil. The pigs were randomly assigned to one of the four treatments: a), control (commercial feed); b), T1 (30% fermented sawdust); c), T2 (10% sardin powder oil) d), T3 (30% fermented sawdust, l0% sardin powder oil and 30% limited amino acid) from 30 kg to 110 kg B. Wt. and slaughtered. The samples were stored at 0±1 C. The physico-chemical properties were determined at the dust of 2, 4, 6, 8 and 15. For the loin the pH's of all the treatments on the 15th day were higher than those of the 2nd day, and that of TI was higher than those of the others. The total moisture contents of all treatments decreased with the storage (P$lt;0.05), and there were no significant differences among treatments on the 15th day. The water soluble proteins of all treatments increased with time and were highest on the 8th day and decreased gianibicanth thereater. T1 had significantly higher water soluble protein than others(P$lt;0.05). The salt soluble proteins of all treatments increased with lure the days(P$lt;0.05). T1 had higher salt soluble protein than others during the storage period(P$lt;0.05). The brittleness, hardness, cohesiveness, gumminess and chewiness of all the treatments were not significantly different among during the period, but elasticity of them increased after from the 2nd day. The brittleness of control was higher than that of the others on the 15th day, and the hardness and chewiness of control were higher than those of the others on the 6th day(P$lt;0.05). Forthebelly the pH's of all the treatments on the 15th day were higher than those of the at 2nd day(P$lt;0.05). T1 showed higher pH than the others during the period(P$lt;0.05). The total moisture contents of control and T1 were significantly decreased with time and there were no significant differences between T2 and T3. T1 showed the highest value. The water soluble proteins of all the treatments decreased with time and was lower on the 15th day(P$lt;0.05). That of control and T1 was significantly higher than that of others on the 2nd day, but there were no significant differences among the treatments on the 15th day. The salt soluble proteins of all treatments increased with time (P$lt;0.05), and they were highest on the 15th day. T1 had significantly higher salt soluble protein than the others on the 8th and 15th days.

      • Adenovirus-mediated gene transfer of interleukin-23 shows prophylactic but not therapeutic antitumor effects

        A novel cytokine interleukin (IL)-23 bears a structural and functional resemblance to IL-12. A recombinant adenovirus expressing IL-23N220L (recombinant replication-defective adenovirus (rAd)/IL-23N220L) that selectively secrets IL-23 was constructed and compared with rAd/IL-12N220L in terms of immunological and antitumor effects. In a prophylactic setting, vaccination with rAd/ovalbumin (OVA) and rAd/IL-23N220L enhanced OVA-specific CD8<SUP>+</SUP> T-cell responses that were closely associated with complete protection against the subsequent challenge of OVA-expressing E.G7 thymoma. However, in a therapeutic setting, the intratumoral injection of rAd/IL-23N220L showed only marginal antitumor activity against several established tumors such as E.G7, CT26 and B16F10. Interestingly, whereas IL-23 still induced tumor-specific CD8<SUP>+</SUP> T-cell responses, it could not activate natural killer (NK) cells in vitro and in vivo. In addition, the adoptive transfer of activated NK cells partially restored the therapeutic antitumor effect of IL-23, indicating that NK cells are one of the crucial factors responsible for the regression of established tumors.         한약찌꺼기 급여가 비육돈의 생산형질 및 도체품질에 미치는 영향

        This study was conducted to investigate the effect of the medicinal herb residue on growth performance, carcass quality and production cost employing 120 Landrace-Yorkshire x Duroc cross-bred gilts and boars. The medicinal herb residue was added to a commercial finishing diet(CONT) at a 3% level(TRT) and the animals were fed for 30 days either CONT or TRT diet beginning from either 58.50∼61.47㎏(finishing phase I) or 83.58∼89.94㎏(finishing phase II), 10 heads in each sex per treatment, were slaughtered. For meat quality evaluation. loin eye sections were removed for determination of cholesterol content and fatty acid compositions. 1. ADG was not affected by the dietary treatment in either gills or boars during phase I. During phase II, ADG was lesser in TRT compared with CONT, but the overall ADG for phases I plus II was not different between CONT and TRT. Feed/gain was reduced by the TRT feeding during phase I but was increased during phase Il, which resulted in a decrease in overall feed efficiency. 2. Backfat thickness was not different between the two dietary groups or sexes. Preliminary carcass grade was better in CONT than in TRT, but final grade was not different between CONT and TRT. Dressing percentage was increased by the dietary TRT treatment; among the four dietary treatment x sex groups, TRT gills showed the greatest percentage. 3. Cholesterol content in the blood was decreased by the TRT feeding. HDL and AI. which were not different between CONT and TRT. CONT gilts had the highest cholesterol and HDL content and AI value. 4. Blood leukocytes(WBC) and hemoglobin(Hb) were increased by the dietary TRT, but the reverse was true for eosinophils(EOS). TRT gilts had the lowest erythrocytes(RBC), WBC, Hb and hematocrit and the greatest EOS. 5. Cholesterol content in the loin eye section was decreased by the dietary treatment. During 45 days of storage it decreased progressively in both CONT and TRT. UFA/ Total, MUFA / Total and UFA / SFA, which were not different between CONT and TRT, increased progressively during storage in CONT, but this trend was not apparent in TRT. The above results indicate that the medicinal herb supplementation can increase weight gain during early finishing phase. However, with extended usage of it up to marketing, it can adversely affect weight gain although it improves both carcass and meat quality.

      • Cryptotanshinone, a lipophilic compound of Salvia miltiorrriza root, inhibits TNF-α-induced expression of adhesion molecules in HUVEC and attenuates rat myocardial ischemia/reperfusion injury in vivo

        The aim of the present study was to evaluate the protective effect of cryptotanshinone (CTS), one of active ingredients of Salvia miltiorrhiza root, on myocardial ischemia-reperfusion injury in rat due to inhibition of some inflammatory events that occur by NF-kB-activation during ischemia and reperfusion. Myocardial ischemia and reperfusion injury was induced by occluding the left anterior descending coronary artery for 30 min followed by either 2 h (biochemical analysis) or 24 h (myocardial function and infarct size measurement) reperfusion. CTS injected (i.v.) 10 min before ischemia and reperfusion insult. CTS significantly reduced the infarct size and improved ischemia and reperfusion-induced myocardial contractile dysfunction. Furthermore, CTS inhibited NF-kB translocation, expression of pro-inflammatory cytokines (TNF-α, IL-1β, IL-6), neutrophil infiltration and MPO activity in ischemic myocardial tissues. CTS also significantly reduced plasma levels of TNF-α, IL-1β due to ischemia and reperfusion. Interestingly, H<SUB>2</SUB>O<SUB>2</SUB>-stimulated NF-kB-luciferase activity and TNF-α-induced expression of vascular cell adhesion molecule-1 (VCAM-1) and intracellular adhesion molecule-1 (ICAM-1) expressions in human umbilical vein endothelial cells (HUVEC) were significantly inhibited by CTS. Taken together, it is concluded that CTS may attenuate ischemia and reperfusion-induced microcirculatory disturbances by inhibition of proinflammatory cytokine production, reduction of neutrophil infiltration and possibly inhibition of adhesion molecules through inhibition of NF-kB-activation during ischemia and reperfusion.

        방사선 조사 및 포장방법이 생계육 및 조리계육의 콜레스테롤 산화에 미치는 영향

        Chicken thigh from a retail market were used as experimental samples. Some chicken samples of raw state were packaged with PVDC at an aerobic and vacuum condition. The other samples were cooked until core temperature arrived at 70℃ and then packaged immediately in the same way of raw samples. After samples were irradiated by electron beam at 6k㏉, they were stored in a refrigerator. Identification and quantity of cholesterol oxides were made at 0 and 7 days of storage, respectively. During the early stage of storage, 7β-hydroxycholesterol, α, β-epoxide, cholestanetriol and 7-ketocholesterol were produced from the raw meat samples, and the production of these chemicals were significantly higher(P<0.05) from the samples with aerobic packaging than those with vacuum packaging. With storage time, 7α-hydroxycholesterol, 6-ketocholesterol and some other chemicals, which were not found during the early stage of storage, were found. Also, the formation of these chemicals were significantly increased(P<0.05) with storage time. Cholesterol and lipid oxidation products of cooked meat after irradiation and irradiated meat after cooking were significantly increased(P<0.05) with storage time for all treatments, and vacuum packaging results in showed significantly lower value(p<0.05) than aerobic packaging. Summarizing the aforementioned results, it was found that the formation of cholesterol and lipid oxides and lipid oxidation was more easily affected by packaging condition than irradiation.

        저장기간에 따른 한약찌꺼기 급여 돈육의 이화학적 특성변화

        This study was conducted to investigate the effect of feeding used oriental medicine refuse(CMR) on physicochemical properties of porks were investigated. Twenty LY × D boars weighing approximately 90 ± 5㎏ were randomly assigned to each of control and treatment groups. The control animals received a regular fattening type diet ad libitum for 30 d and the other ones were fed a mixture of the fattening diet and OMR at the rate of 77 to 3 on as fad basis. On the 30th d of dietary treatment, the animals were slaughtered and their loins and bellies were collected from each treatment. The samples were stored at 4 ± 1 ℃. The physico-chemical properties was determined on the days of 2, 4, 7, 12 and 15. The pH of loin and belly were significantly lower in the OMR compared to the treatment(P$lt;0.05). The shear forces of loin and belly were not different statistically between the two groups. The water soluble proteins of loin and belly were significantly lower in OMR than those in the treatment. The proteins of loin increased until 7 days of storage and they tended thereafter. The salt soluble proteins of loin and belly tended to higher in the control than those in the treatment and they increased with the storage time(P$lt;0.05). The Hunt L values of loin appeared to be higher in OMR compared to the treatment. On the other hand the OMR was significantly higher than the other in the Hunt L values(P$lt;0.05). Hunt a values of loin was significantly higher in OMR(P$lt;0.05), than those in the other, but those of belly were significantly higher in the control group on the 2nd day(P$lt;0.05). Hunt b values of loin and belly were significantly higher in the OMR than those in the other on the 15th days(P$lt;0.05).

      • 압연 중 핀홀 결함의 변형 이력의 추적

        In this paper, two-dimensional finite element approach of prediction the plastic deformation of tiny pinhole defects on the initial bloom surface, which were formed during continuous casting process of the base material of bloom. The initial pinhole shapes of which plastic deformations are to be traced are assumed V-shaped and U-shaped. Two different kinds of pass schedule of consecutive rolling stages are assumed to investigate the defect closing or lapping behaviors, i.e., one pass schedule of two forward rolling stages and another pass schedule of reverse rolling stage after forward rolling stage. The dimensions of the processes and the sizes of pinholes are also assumed to reflect the actual three-dimensional rolling processes. It has been shown that most plastic deformation occurs at the entry side and lapped or closed defects slightly open when they pass through the exit side. It is noteworthy that the reverse rolling decreases the lapped defect formed in the previous pass, implying that there exists an optimized pass schedule design to minimize the surface defect.

      • Activation of Akt as a mechanism for tumor immune evasion.

        <P>Immune evasion is an important reason why the immune system cannot control tumor growth. To elucidate the mechanism for tumor immune evasion, we generated an immune-resistant human papillomavirus type 16 (HPV-16) E7-expressing tumor cell line by subjecting a susceptible tumor cell line to multiple rounds of in vivo immune selection with an E7-specific vaccine. Comparison of parental and immune-resistant tumors revealed that Akt is highly activated in the immune-resistant tumors. Retroviral transfer of a constitutively active form of Akt into the parental tumor significantly increased its resistance against E7-specific CD8(+) T-cell mediated apoptosis. The observed resistance against apoptosis was found to be associated with the upregulation of antiapoptotic molecules. We also observed that intratumoral injection of an Akt inhibitor enhanced the therapeutic efficacy of E7-specific vaccine or E7-specific CD8(+) T-cell adoptive transfer against the immune-resistant tumors. Thus, our data indicate that the activation of PI3K/Akt pathway represents a new mechanism of immune escape and has important implications for the development of a novel strategy in cancer immunotherapy against immune-resistant tumor cells.</P>
