        Selection of Potential Virulence Factors Contributing to Streptococcus suis Serotype 2 Penetration into the Blood-Brain Barrier in an In Vitro Co-Culture Model

        Meningitis caused by Streptococcus suis serotype 2 (S. suis 2) is a great threat to the pig industry and human health. Virulence factors associated with the pathogenesis of meningitis have yet to be clearly defined, even though many potential S. suis 2 virulence factors have been identified. This greatly hinders the progress of S. suis 2 meningitis pathogenesis research. In this study, a co-culture blood-brain barrier (BBB) model was established using primary porcine brain microvascular endothelial cells and astrocytes, and the whole genome library of S. suis 2 was constructed using phage display technology. Finally, a total of 14 potential virulence factors contributing to S. suis 2 adherence to and invasion of the BBB were selected by analyzing the interactions between the phage library and the co-culture model. Twelve of these factors have not been previously reported in meningitis-related research. The data provide valuable insight into the pathogenesis of S. suis 2 meningitis and potential targets for the development of drug therapies.

        Atractylodin induces apoptosis through downregulation of PI3Kγ-mediated PI3K/Akt/mTOR/p70S6K signalling in colon cancer cells and suppresses the tumour formation in xenograft mice model

        This study used both in vitro and in vivo models to evaluate the efficacy of atractylodin as an anticancer treatment for colorectal cancer. The cytotoxicity of atractylodin on colon cancer cells was assessed using the MTT assay, and atractylodininduced apoptosis was determined using flow cytometry. The expression of cleaved caspase 3 and other apoptotic proteins was examined using Western blotting to determine the mechanism underlying atractylodin's anticancer activity. In addition, the role of PI3K/Akt/mTOR/p70S6K signalling in atractylodin-induced apoptosis in colon cancer cells was analyzed. The study found that atractylodin caused dose-dependent ROS-mediated apoptosis and DNA damage in colon cancer cells and activated caspase 3. Furthermore, atractylodin inhibited the PI3K/Akt/mTOR/p70S6K signalling pathway by targeting PI3Kγ in colon cancer cells. Molecular docking analysis indicated that atractylodin binds to the Akt binding pocket of PI3Kγ. The study also evaluated the antitumour effects of atractylodin on a colon cancer tumour xenograft model and found that it significantly reduced tumour growth and volume by inducing apoptosis. These results suggest that atractylodin has potential as a candidate for the treatment of colorectal cancer, although further research is necessary.

        Preparation and Physicochemical Characterizations of Tanshinone IIA Solid Dispersion

        This investigation describes a novel approach to prepare solid dispersions of tanshinone IIA using a laboratory-scale planetary ball mill. Poloxamer 188 was employed as the surfactant carrier to improve the solubility and dissolution of the poorly soluble drug, tanshinone IIA. Solubility and dissolution were evaluated compared to the corresponding physical mixtures and pure drug. Furthermore, the physicochemical properties of the solid dispersions were investigated using scanning electron microscopy, powder X-ray diffraction, differential scanning calorimetry, Fourier transform infrared spectroscopy and ultraviolet spectrophotometry. The solid dispersion significantly enhanced drug solubility and dissolution compared with pure drug and the physical mixtures. Scanning electron microscopy, powder X-ray diffraction,differential scanning calorimetry and Fourier transform infrared spectroscopy analyses of tanshinone IIA/poloxamer 188 system confirmed that there were intermolecular interactions between tanshinone IIA and poloxamer 188 and no conversion to crystalline material. Tanshinone IIA existed in a microcrystalline form in the system. These results suggested that improvement of the dissolution rate could be correlated to the formation of a eutectic mixture between the drug and the carrier. After 60 days the solid dispersion samples were chemically and physically stable. The present studies indicated that the planetary ball mill technique could be considered as a novel and efficient method to prepare solid dispersion formulations.

        Does IoT service orchestration in public services enrich the citizens’ perceived value of digital society?

        The continuous growth of digital transformation aims to leverage the changes and opportunities for IoT technologies in public sector, which has created an impact on the sustainability of digital societies. The Internet of Things (IoT) contains significant challenges such as citizens’ perceived value towards intention to use in public services that could stand in a way of realizing its potential benefits. Our study aims to investigate the influencing factors of citizens’ usage intention concerning IoT service orchestration in public services along with the mediation influence of perceived value. A comprehensive public IoT Perceived Value Model (IoT-PVM) was enumerated to provide success factors for IoT service orchestration. This model was analytically tested by PLS-SEM approach. The results highlighted that perceived value has a mediation effect in IoT public service intentions to use with 59% perceived variance. The success of government IoT services is best measured by the perceptions of citizens, therefore, this study contributed to identifying the factors concerning public engagement with IoT service orchestration along with describing the positive role of digital society affinity. In terms of practical significance, a use case IoT model was presented with provides an intersectional perspective of citizens concerning IoT service orchestration in public services.

        Recent Progress of Gr/Si Schottky Photodetectors

        By combing the carrier mobility of graphene with the excellent light absorption properties of silicon, ultra-shallow Schottkyjunction can be obtained, and can exist stably for a long time. The photoelectric property of Schottky junction is determinednot only by graphene and silicon semiconductor layer, but also by the interface layer between the two. Through a series ofoptimizations, the performance of graphene/silicon Schottky junction photodetectors can be continuously improved. Asa result, graphene/silicon Schottky junctions more promising for the development of next generation photodetectors withits stability, ease of preparation and sensitivity. In this review, we firstly give a brief introduction to Gr Schottky junctionphotodetectors, and then present a comprehensive review on the recent progress of optimizing Gr/Si Schottky junction photodetectorsin the past few years, including light management engineering, band engineering and interfacial engineering. Finally, the current challenges are summarized and further perspectives are outlined.
