        Silence of LncRNA GAS5Protects Cardiomyocytes H9c2 against Hypoxic Injury via Sponging miR-142-5p

        The regulatory role of long noncoding RNA (lncRNA) growth arrest-specific transcript 5 (GAS5) in both cancerous and noncancerous cells have been widely reported. This study aimed to evaluate the role of lncRNA GAS5 in heart failure caused by myocardial infarction. We reported that silence of lncRNA GAS5 attenuated hypoxia-triggered cell death, as cell viability was increased and apoptosis rate was decreased. This phenomenon was coupled with the down-regulated expression of p53, Bax and cleaved caspase-3, as well as the up-regulated expression of CyclinD1, CDK4 and Bcl-2. At the meantime, the expression of four heart failure-related miRNAs was altered when lncRNA GAS5 was silenced (miR-21 and miR-142-5p were up-regulated; miR-30b and miR-93 were down-regulated). RNA immunoprecipitation assay results showed that lncRNA GAS5 worked as a molecular sponge for miR-142-5p. More interestingly, the protective actions of lncRNA GAS5 silence on hypoxia-stimulated cells were attenuated by miR-142-5p suppression. Besides, TP53INP1 was a target gene for miR-142-5p. Silence of lncRNA GAS5 promoted the activation of PI3K/AKT and MEK/ERK signaling pathways in a miR-142-5p-dependent manner. Collectively, this study demonstrated that silence of lncRNA GAS5 protected H9c2 cells against hypoxia-induced injury possibly via sponging miR-142-5p, functionally releasing TP53INP1 mRNA transcripts that are normally targeted by miR-142-5p.

        From the phenomenon-driven perspective, on the basis of data from Thomson Reuters Web of ScienceTM database covering a period of 29 years (January 1990 to August 2019), this study employs bibliometric and content analyses to evaluate the research state of the internationalization and innovation relationship. This study is to assist internationalization scholars in positioning their research more explicitly and encourage them to deeply think about which approach and theoretical mechanisms best fit their research questions and designs.

        Dynamic process: international diversification and innovation performance from emerging economies

        Drawing upon the transaction cost economics and organizational learning theory, this study examines how international diversification affects firm's innovation performance. With a longitudinal data set obtained from a sample of 73 Chinese firms in information communication equipment manufacturing industry, home appliances manufacturing industry, and low voltage apparatus and cable manufacturing industry, we find a curvilinear inverted S-shaped relationship between international diversification and innovation performance. It implies that emerging enterprises might gain increase at the expansion stage, experience a hard time at the conflict stage, and then reap again at the convergence phase.

        펜데믹 이후의 홈트레이닝 발전추세 탐색 : 빅데이터 분석의 활용

        The purpose of this study was to propose the development trends of home training by using big data analysis, which has undergone market changes due to the post-pandemic. For this purpose, the social metrics program Textom and network analysis tool Ucinet were utilized, with the keyword ‘home training’ being used. Text mining, TF-IDF, degree centrality, N-gram, and semantic network analysis were conducted. The data analysis period was selected from September 26, 2022 to March 31, 2023. Analysis of word frequency, TF-IDF, degree centrality, and N-gram revealed that public interest in home training due to COVID-19 remains consistently high, and people recognize home training as a new exercise culture that raises interest in health. Semantic network analysis showed four groups, including ‘smart home training,’ ‘home training methods,’ ‘home training trends,’ and ‘home training market.’ The development of home training has also led to the marketing of home gyms and various fitness equipment. As the world emerges from isolation due to the post-pandemic, manufacturers of industries related to home training need to adjust by improving equipment quality and lowering prices to instill confidence in consumers that home training is a better choice than location-based gyms. Service industries related to home training should also provide safer and more convenient services by raising service standards that are closely linked with training content. Therefore, the analyzed results are used to provide practical data and propose strategies to promote the development of the home training market at a theoretical level.

        Novel Auto-regressive Measurement of Diamond Tool Wear in Ultra-precision Raster Milling

        In this paper, a new auto-regressive algorithm is proposed to reconstruct 3D topographic surface for diamond tool wear, using its in-process image. First, based on digital image processing technique, tool wear lands are separated from a tool wear image captured by a CCD camera under a 100X optical system; Second, a traverse search method of arc translation is put forward to eliminate feigned wear lands, and a least square polynomial method is adopted to fit inner- and outer-contours of the wear lands, self-adaptively filtering noises and connecting the discontinuous wear lands; Finally, the auto-regressive calibration method is developed to reconstruct its 3D topographic surface. The wear land is extracted self-adaptively, and the wear area, maximal wear width, average wear width and worn volume can be determined automatically by the algorithm. The reconstructed 3D topography of the tool wear land can be identified, based on the tool wear image captured by SEM. The result indicates that the method is capable of reconstructing 3D topography of the tool wear land and provides a possibility for in-process 3D-wear measurement in ultra-precision raster milling (UPRM) and the algorithm reliability is validated finally. And the influence of tool wear on surface roughness is discussed.

      • Network Analyses of Gene Expression following Fascin Knockdown in Esophageal Squamous Cell Carcinoma Cells

        Fascin-1 (FSCN1) is an actin-bundling protein that induces cell membrane protrusions, increases cell motility, and is overexpressed in various human epithelial cancers, including esophageal squamous cell carcinoma (ESCC). We analyzed various protein-protein interactions (PPI) of differentially-expressed genes (DEGs), in fascin knockdown ESCC cells, to explore the role of fascin overexpression. The node-degree distributions indicated these PPI sub-networks to be characterized as scale-free. Subcellular localization analysis revealed DEGs to interact with other proteins directly or indirectly, distributed in multiple layers of extracellular membrane-cytoskeleton/ cytoplasm-nucleus. The functional annotation map revealed hundreds of significant gene ontology (GO) terms, especially those associated with cytoskeleton organization of FSCN1. The Random Walk with Restart algorithm was applied to identify the prioritizations of these DEGs when considering their relationship with FSCN1. These analyses based on PPI network have greatly expanded our comprehension of the mRNA expression profile following fascin knockdown to future examine the roles and mechanisms of fascin action.

      • Blending Glowworm Swarm Optimization Algorithm with Thermal-Mechanical Coupling Finite Element Model for Optimization Method of Box Structure Welding Technology

        Welding sequence is the important influence factor of the temperature field and residual stress and deformation in welding structure, At present, determining the welding sequence is usually based on experience and test method. For complex structure it is very difficult to get the optimum welding sequence. This study is aiming to settle the problem of solving the optimum welding sequence of complex box welding structure. In our study, we take the welding deformation as objective function, and then, associate glowworm swarm optimization algorithm with thermal-mechanical coupling and nonlinear thermal elastoplastic finite element model to optimize numerical simulation, eventually the optimum welding sequence is determined. Optimization results demonstrate that the optimum welding sequence obtained by our method has small welding deformation and little change rate. At the same time, our method is faster, more accurate, more effective than the traditional experience or test method.

      • Characterization of a Late Gene, ORF60 from Bombyx mori Nucleopolyhedrovirus

        Open reading frame 60 of Bombyx mori nucleopolyhedrovirus (Bm60) is located between 56,673 and 57,479 bp in the BmNPV genome which encodes 268 amino acid residues with predicted molecular weight of 31.0 kDa. Bm60 and its homologues have been identified in 11 completely sequenced lepidopteran NPVs. The transcript of Bm60 was detected by RT-PCR at 18-72 h post-infection (p.i.), while the corresponding protein could be detected at 24-72 h p.i. in BmNPV-infected BmN cells by Western blot analysis using a polyclonal antibody against Bm60. The expression of Bm60 was inhibited in the presence of Ara-c, an inhibitor of viral DNA synthesis. These results together indicated that Bm60 was a late gene. The size of Bm60 product was found to be a 31 kDa in BmNPV-infected BmN cells, consistent with predicted molecular weight. Immuno-fluoresence analysis showed that the Bm60 product was first detected in the cytoplasm at 24 h p.i and also located in nucleus during later infection. In conclusion, the available data suggest that Bm60 is a functional ORF of BmNPV and encodes a 31kDa protein expressed in the later stage of infection cycle.
