        Candida fermentati SI의 exo-β-(1,3)-glucanase 유전자의 클로닝 및 그 특성

        본 연구에서는 C. fermentati SI가 생산하는 isoflavone 배당체 가수분해 효소를 클로닝하여 염기 서열을 밝힌 뒤 P. pastoris X-33에 형질전환하여 재조합 효소의 과발현을 시켰고, 또한 재조합 isoflavone 가수분해 효소의 효소학적 특성을 조사하였다. 재조합 isoflavone 가수분해 효소의 분자량은 약 50.4 kDa이었으며, Meyerozyma guilliermondii ATCC 6260의 exo-1,3-β-glucanase와 96%로 가장 높은 homology 를 나타내었다. exo-1,3-β-glucanase의 ORF는 pPICZA 벡터로 클로닝 후 P. pastoris X-33으로 형질전환을 하였으며, His6-tag을 이용하여 효소를 정제하였다. 정제된 효소는 citrate phosphate buffer pH 4.5에서 최적 활성을 나타내었으며, 효소의 최적 활성 온도는 40℃로 나타났다. 40℃이상에서는 효소의 활성이 급격하게 감소함을 확인 하였으며, pH안정성을 조사한 결과 비교적 넓은 범위인 4-8 사이에서 80%이상의 활성을 유지하였다. 따라서, 재조합 효소의 과발현을 통해 isoflavone aglycone의 효율적인 생산에 이용할 수 있을 것으로 사료된다. An isoflavone glucosidase that catalyzes the hydrolysis of isoflavone glucosides into glucose and corresponding aglycones was purified from Candida fermentati SI. The N-terminal sequence was determined to be GLNCDYCN. We designed degenerate primers on the basis of these amino acid sequences and successfully cloned the full structural gene sequence of the isoflavone glucosidase using inverse PCR. The exo β- (1,3)-glucanase gene consists of 1227 base-pair nucleotides, encoding a 408-amino-acid sequence that shares 41-96% amino acid homology with other yeast exo-β-(1,3)-glucanases belonging to glycoside hydrolase family 5. The recombinant exo-β-(1,3)-glucanase was expressed in Pichia pastoris X-33, using a pPICZA vector system, and further characterized. The molecular mass of the purified exo-β-(1,3)-glucanase was estimated by SDS-PAGE to be 47 kDa. The optimal pH and temperature were pH 4.5 and 40°C, respectively. The Km values of the purified exo-β-(1,3)-glucanase for daidzin and genistin were 0.12 mM and 0.14 mM, respectively. The Vmax values of the purified isoflavone glucosidase were 945.03 U/mg for daidzin and 835.92 U/mg and for genistin.

        Inhibitory Effect of Scopoletin Isolated from Sorbus commixta on TNF-α-Induced Inflammation in Human Vascular Endothelial EA.hy926 Cells through NF-κB Signaling Pathway Suppression

        마가목은 한방에서 기침, 천식, 기관지 질환 등의 치료에 이용되고 있다. 본 연구에서는 마가목(sorbus commixta) 수피의 chloroform 분획물로부터 3종의 triterpenoid 화합물과 1종의 coumarin 화합물을 분리하였다. 분리된 화합물의 구조는 MS와 1D-, 2D-NMR분석에 의해 확인하였으며, 이들은 lupeol, β-sitosterol, ursolic acid와 scopoletin으로 구조동정되었다. 분리된 화합물 중 scopoletin은 마가목에서 처음으로 분리된 화합물이다. Scopoletin은 식물에 널리 분포하고 있는 물질로써 항염증 활성을 가진 기능성 화합물이다. 분리된 화합물들의 혈관 염증 억제에 대한 효과를 평가하기 위해 in vitro에서 LDL 산화 억제능을 평가한 결과, 분리된 화합물 중 scopoletin (IC50=10.2 μM)이 강한 억제 활성을 나타내었다. TNF-α로 활성화된 인체혈관내피세포(EA.hy926)를 이용한 실험에서 scopoletin은 세포부착인자인 ICAM-1, VCAM-1, E-selectin의 발현을 저해하였고, THP-1 단핵구와 EA.hy926 혈관내피세포 간의 부착력도 약화시켰다. 뿐만 아니라, scopoletin은 TNF-α로 유도된 NF-κB 전사인자의 핵내 이동 및 IκBα의 인산화도 저해하였다. 따라서 마가목 추출물로부터 분리된 scopoletin은 NF-κB 신호전달의 억제를 통해 세포부착인자의 발현을 감소시키고, 단핵구의 혈관내피세포로의 부착을 억제시켜 혈관내 항염증 활성을 나타내었다. 이러한 실험결과, scopoletin은 혈관염증 반응으로부터 유도되는 죽상동맥경화증 치료를 위한 후보소재로서 이용될 가능성이 있다고 사료된다. Sorbus commixta Hedl. has traditionally been used as a remedy for cough, asthma, and other bronchial disorders. In this study, three major triterpenoids—lupeol, β-sitosterol, and ursolic acid and a coumarin, scopoletin, were isolated from a CHCl3-soluble fragment of the bark of S. commixta. Their structures were identified by spectroscopic analyses, including mass spectrometry (MS), 1D-, and 2D- nuclear magnetic resonance spectroscopy (NMR), as well as by comparing the data with data reported in the literature. Scopoletin was isolated from this plant for the first time. It is a nutraceutical compound contained in many plants that has been reported to exert diverse biological activities, including anti-inflammatory effects. This study examined the inhibitory effect of scopoletin on TNF-α-induced vascular endothelial inflammation. Unlike the marginal impact of other compounds against low-density lipoprotein (LDL) oxidation and vascular endothelial inflammation, scopoletin showed remarkable activity on LDL oxidation (IC50 = 10.2 μM) and exerted vascular anti-inflammatory effects in EA.hy926 human endothelial cells activated by TNF-α. It suppressed the expression of adhesion molecules, such as ICAM-1, VCAM-1, and E-selectin, and blocked the adhesion between THP-1 monocytes and EA. hy926 endothelial cells. It also inhibited TNF-α-induced NF-κB translocation from the cytosol to the nucleus. Moreover, IκBα phosphorylation, which was increased by TNF-α treatment, was reduced after treatment with scopoletin. Thus, scopoletin inhibited TNF-α-induced vascular inflammation in endothelial cells by suppressing the NF-κB signaling pathway. These results demonstrate that owing to its anti-inflammatory activity in the vascular endothelium, scopoletin has the potential to inhibit atherosclerosis development.

        For the evaluation of the d.f., D.F. teeth and the recurrent caries frequency of the filled teeth in Korean children, the author performed oral examination of 1,648 children of L primary school on June 1975. The results obtained were as follows: 1. The percentage of d.f., D.F. person was 93.93 and average number of d.f., D.F. teeth per child was 5.36. 2. The d.f., D.F. teeth ratio was 41.5, where d.f. was 39.83 and D.F. was 45.34. 3. The d.D. teeth ratio was 35.86, where d. was 34.82 and D. was 38.26. 4. The f.F. teeth ratio was 5. 64, where f. was 5.01 and F. was 7.08. 5, The percentage of f.F. teeth to d.f., D.F. teeth was 13.58, and the percentage of f.F. teeth to total filled teeth was 36.05. 6 In the total filled teeth, the percentage of amalgam filled teeth was 74.08, gold inlay filled teeth was 18.48 and metal crown weared teeth was 7.49. 7 In the total filled teeth, 84.42 percent of amalgam filled teeth was in deciduous teeth, 95.27 percent of gold inlay filled teeth was in permanent teeth and 92.77 percent of metal crown weared teeth was in deciduous teeth. 8. The percentage of f.F. teeth in amalgam filled teeth was 26.79, where f. was 24.04 and F. was 41.67. 9. The percentage of f. F. teeth in gold inlay filled teeth was 47.31, where f . was 34.38 and F. was 47.95.
