Molecular insights into the role of genetic determinants of congenital hypothyroidism

In our previous studies, we have demonstrated the association of certain variants of the thyroid-stimulating hormone receptor (TSHR), thyroid peroxidase (TPO), and thyroglobulin (TG) genes with congenital hypothyroidism. Herein, we explored the mechanistic basis for this association using different in silico tools. The mRNA 3'-untranslated region (3'-UTR) plays key roles in gene expression at the post-transcriptional level. In TSHR variants (rs2268477, rs7144481, and rs17630128), the binding affinity of microRNAs (miRs) (hsa-miR-154-5p, hsa-miR-376a-2-5p, hsa-miR-3935, hsa-miR-4280, and hsa-miR-6858-3p) to the 3'-UTR is disrupted, affecting post-transcriptional gene regulation. TPO and TG are the two key proteins necessary for the biosynthesis of thyroid hormones in the presence of iodide and H<sub>2</sub>O<sub>2</sub>. Reduced stability of these proteins leads to aberrant biosynthesis of thyroid hormones. Compared to the wild-type TPO protein, the p.S398T variant was found to exhibit less stability and significant rearrangements of intra-atomic bonds affecting the stoichiometry and substrate binding (binding energies, ΔG of wild-type vs. mutant: -15 vs. -13.8 kcal/mol; and dissociation constant, K<sub>d</sub> of wild-type vs. mutant: 7.2E<sup>-12</sup> vs. 7.0E<sup>-11</sup> M). The missense mutations p.G653D and p.R1999W on the TG protein showed altered ΔG(0.24 kcal/mol and 0.79 kcal/mol, respectively). In conclusion, an in silico analysis of TSHR genetic variants in the 3'-UTR showed that they alter the binding affinities of different miRs. The TPO protein structure and mutant protein complex (p.S398T) are less stable, with potentially deleterious effects. A structural and energy analysis showed that TG mutations (p.G653D and p.R1999W) reduce the stability of the TG protein and affect its structure-functional relationship.

Mononuclear cells isolated from human umbilical cord blood were differentiated in to spindle shaped fibrous mesenchymal stem cells (MSCs) in DMEM with 10% fetal bovine serum. These progenitor cells were further examined for their ability to differentiate into osteoblasts by culturing them in osteogenic differentiation media (Stem Pro(R)). Calcium mineralization assay, alizarin red S, von kossa staining confirmed that MSCs from cord blood were capable of mineralization when they were cultured in osteogenic medium. The adhesion studies shown that MSCs have greater affinity towards fibronectin (FN) coated dishes, compared to albumin (Alb) coated dishes due to CD29and CD44 surface receptors.

Apart from the traditional risk factors thatcontribute to this devastating condition, in the past fewdecades, much attention has been focused on plasma totalhomocysteine mainly because of its strong association withhomocysteine induces oxidative stress and hence the presentwork was undertaken to assess the total homocysteinestatus and plasma total antioxidant capacity in the acutemyocardial infarction (AMI) patients among Tamil population.The study subjects included only the Tamil population. Bloodsamples were collected from 100 AMI patients and 100controls. Plasma was separated and the total antioxidantstatus was assessed as a measure of ferric reducing powerof antioxidants using spectrophotometric method. Plasmatotal homocysteine concentrations were assessed byantioxidant status was significantly decreased, the plasmahomocysteine concentrations were elevated in AMI patientscompared to the controls. However, there was no correlationbetwen the homocysteine levels and total antioxidantstatus. The findings of this study may have therapeuticimplications, including food sources rich in antioxidants forall AMI patients to minimize the effect of free radicals formedduring oxidative stress among Tamil population.

Plasma was separated and the total antioxidant status was assessed as a measure of ferric reducing power of antioxidants using spectrophotometric method. Plasma total homocysteine concentrations were assessed by automated chemiluminescence method. While Total antioxidant status was significantly decreased, the plasma homocysteine concentrations were elevated in AMI patients compared to the controls. However, there was no correlation between the homocysteine levels and total antioxidant status. The findings of this study may have therapeutic implications, including food sources rich in antioxidants for all AMI patients to minimize the effect of free radicals formed during oxidative stress among Tamil population.

This research article aims at reporting the influence of magnesium by studying the structural, electrical impedance and modulus properties of the Mg substituted Li 4 Ti 5 O 12 . These studies are useful for the electrochemical properties. The XRD reveals that the structure of all the Mg substituted materials belongs to the cubic spinel group having Fd-3m space symmetry. SEM images display the structural, morphological properties with the average size of grains falling in the vicinity of 1 μm. The electrical impedance of Li 4− x Mg x Ti 5 O 12 materials was analyzed at frequencies between 20 Hz and 1 MHz and in the 30–120 °C range of temperature by employing the complex impedance spectroscopy (CIS) method. The modulus formalism is also a suitable tool to understand the dynamical characteristics of electrical transport phenomena. The complex electric modulus spectrum signifi es quantifying the allocation of ion energies or confi gurations in the lattice. Also, it portrays the electrical relaxation of ion-conducting lattices as a feature of materials at a minuscule level. The obtained results of substitution of Mg in Li 4 Ti 5 O 12 anode materials improve the potential applications of conductivity and charge/discharge performance.
