        Anti-allergic effects of a nonameric peptide isolated from the intestine gastrointestinal digests of abalone (Haliotis discus hannai) in activated HMC-1 human mast cells

        <P>The aim of the present study was to examine whether the intestine gastrointestinal (GI) digests of abalone [Halioti s discus hannai (H. discus hannai)] modulate inflammatory responses and to elucidate the mechanisms involved. The GI digests of the abalone intestines were fractionated into fractions I (>10 kDa), 11 (5-10 kDa) and III (<5 kDa). Of the abalone intestine GI digests (AIGIDs), fraction III inhibited the passive cutaneous anaphylaxis (PCA) reaction in mice. Subsequently, a bioactive peptide [abalone intestine GI digest peptide (AIGIDP)] isolated from fraction III was determined to be 1175.2 Da, and the amino acid sequence was found to be PFNQGTFAS. We noted that the purified nonameric peptide (AIGIDP) attenuated the phorbol-12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-induced histamine release and the production of pro-inflammatory cytokines, such as tumor necrosis factor-alpha (TNF-alpha), interleukin (IL)-1 beta and IL-6 in human mast cells (HMC-1 cells). In addition, we also noted that AIGIDP inhibited the PMACI-induced activation of nuclear factor-kappa B (NF-kappa B) by suppressing I kappa B alpha phosphorylation and that it suppressed the production of cytokines by decreasing the phosphorylation of JNK. The findings of our study indicate that AIGIDP exerts a modulatory, anti-allergic effect on mast cell-mediated inflammatory diseases.</P>

        Anti-inflammatory Effect of Gyulpidaehwangbakcho-tang (Jupidahuangpoxiao-tang) in the Collagen-induced Arthritis Mouse Model

        Objectives: To investigate anti-inflammatory and anti-arthritic effects of Gyulpidaehwangbakcho-tang (GDBT) extract in a murine model of rheumatoid arthritis. Methods: The mice received $100{\mu}g$ of bovine type II collagen in Freund's complete adjuvant by intradermal injection at the base of the tail on day 0 and a booster injection on day 21. The mice were orally administered with GDBT (200 or 50mg/kg dissolved in distilled water) daily from day 1 to day 21 after arthritis incidence, and monitored for disease incidence and the severity of arthritis up to day 21. In order to evaluate the effect of GDBT on disease progression, we examined pro-inflammatory cytokines including IL-$1{\beta}$, IL-6, TNF-${\alpha}$, COX-2 and NOS-II. Results: GDBT produced a significant and dose dependent inhibition of arthritis and inflammation during the entire duration of the study. This action was characterized by the decreased production of IL-$1{\beta}$, IL-6, TNF-${\alpha}$, COX-2, and NOS-II in vivo. Conclusion: We believe that the anti-arthritic activity of GDBT is due to its modulatory effect on the expression of pro-inflammatory cytokine in the synovium. Our results contribute towards validation of the traditional use of GDBT in the treatment of RA and other inflammatory joint disorders.

        방기복령탕이 papain으로 유도된 생쥐의 골관절염에 미치는 영향

        Objectives:This study was carried out to know the anti-osteroarthritic effects of Bangkibokryeong-tang(Fanjifuling-tang(BBT)) on the papain-induced osteoarthritis C57BL/10 mouse.Methods:Osteoarthritis was induced by injection of papain(6 μl) into knee joint of mouse. Osteoarthritic mice were divided into 4 groups(normal, control, joinsⓡ BBT).The injection did not fit the normal group.A week later, after the injection of papain, control group was taken normal saline 200 μl, positive control group was taken joinsⓡ(100 mg/kg), treated group was taken extract of Bangkibokryeong-tang(Fanjifuling-tang(BBT))(400 mg/kg).After then, we examined hepatotoxicity, nephrotoxicity, inflammation cytokines, expression of inflammation factor mRNA, hemotology, histology through the micro CT-arthrography, and etc.Results:1.Hepatotoxicity and nephrotoxicity have not expressed.2.The levels of IL-1β, TNF-α, IL-6, MCP-1, Thromboxane B2, Leukotriene B4, Prostaglandin E2 in serum were significantly decreased.3.In hematology, the levels of neutrophils and monocytes were significantly decreased.4.The expression of inflammation factor mRNA like TNF-α and IL-6, COX-2, iNOS-Ⅱ were significantly inhibited.5.In micro CT-arthrography, cartilage volume was less decreased.6.The degree of osteoarthritis induced damage of joint of BBT group is low in histopathologic observation(hematoxylin&eosin(H&E), Safranin-O).Conclusions:According to this study, BBT has effect of anti-osteoarthritis.Further clinical research for the cartilage protective effect is necessary.

        관절3호방이 인간 섬유아세포양 활막세포조직에 미치는 연구

        Objectives : Gwanjul-bang(Guanjie-fang)-3 (GJB-3) is a prescription used clinically to treat rheumatoid arthritis. The purpose of this study is to investigate the immunity responses of GJB-3 to the human fibroblast-like synoviocytes isolated from patients with rheumatoid arthritis. Methods : Experimental groups were divided into 6 groups ; The hFLS were stimulated with hrTNF-α(100 ng/ml) and hrIL-1β(10 U/ml). Afterwards, They were treated with medium (CT), methotrexate (10㎍/ml, MTX) and GJB-3 (1, 10, 100㎍/ml) for 6 hours. The cells were harvested following the further incubation with [3H]-thymidine reagent for 4 hours. The WT group was cultured without cytokine stimulation. Results : 1. The proliferation of hFLSs was significantly reduced in 100㎍/ml dose. 2. The mRNA expression of TNF-α and the production of ROS was dose-dependently reduced in hFLS. 3. The mRNA expression of IL-1β, IL-6, IL-8, ICAM-1, NOS-Ⅱ, MMP-3 were reduced in high concentration. 4. The production of NO was significantly reduced in over 10㎍/ml dose. 5. The mRNA expression of TIMP-1 was dose-dependently increased. Conclusions : We found out that GJB-3 is considered to be effective in treating rheumatoid arthritis.

        The effects of BRL-50481 on ovalbumin-induced asthmatic lung infl ammation exacerbated by co-exposure to Asian sand dust in the murine model

        Asian sand dust (ASD), which mainly originatesin China and Mongolia in the spring and blows into Korea,can exacerbate respiratory and immunological diseases. This study aims to observe eff ects of co-exposure to ASD onovalbumin (OVA)-induced asthmatic lung infl ammation andof treatment with a phosphodiesterase 7 (PDE7) inhibitor ina mouse model. The challenge with OVA increased airwayhyperresponsiveness (AHR) and infl ammatory cell infi ltrationinto the lung tissue. Interleukin (IL)-13, tumor necrosisfactor-alpha, monocyte-protein-1, mucin, and antigen-specific IgE and IgG1 production increased in mouse serum. The co-exposure of ASD signifi cantly exacerbated theseeff ects in this asthma model. Notably, the administration ofa PDE7 inhibitor, BRL-50481 (BRL), signifi cantly reducedAHR, infi ltration of infl ammatory cells into the lungs, andthe levels of type 2 T helper cell-related cytokines, antigenspecific immunoglobulins, and mucin. Thus, the administrationof BRL ameliorated OVA-induced allergic asthmaticresponses exacerbated by co-exposure to ASD. This studysuggests that PDE7 inhibition can be a therapeutic strategyfor infl ammatory lung diseases and asthma via the regulation of T lymphocytes and reduction of IL-13, and, consequently,mucin production.

        관절1호방이 Collagen Ⅱ로 유발된 관절염 억제에 관한 연구

        Objectives : The aim of this study was to know the immunity responses of Gwanjulbang-1(Quanjiefang-1)(hereinafter referred to GJB-1) to on Rheumatoid Arthritis in Collagen-induced Arthritis(CIA) Mice Methods : For this purpose, experiments were performed to measure the cytotoxicity against mLFC, hFLSs and the production of proinflammatory cytokines IL-1β, IL-6, TNF-α, IFN-γ in hFLSs and the production of pro-inflammatory cytokines IL-4, IL-10 and the value of CD3+(T), CD19+(B), CD3+/CD69+, CD4, γδ T cells, CD4+/CD25+ Results : 1. The cytotoxicity against mLFC, hFLSs were not measured. 2. The production of pro-inflammatory cytokines IFN-γ were reduced in hFLSs. 3. The production of pro-inflammatory cytokines IL-4, IL-10 were increased. 4. Statistically significant value of CD3+(T), CD19+(B), CD3+/CD69+, CD4, γδ T cells, CD4+/CD25+ compared with control group, wild type, MTX. Conclusions : Comparison of the results for this study showed that GJB-1 had immunomodulatory effects of suppressing or enhancing. So we expect that GJB-1 should be used as a effective drugs for not only rheumatoid arthritis but also another auto-immune disease. Therefore we have to survey continuously in looking for the effective substance and mechanism in the future.

        Recently, many a multimedia software is introduce named CD-title due to equipment of multimedia environment. This paper describes design and implement of multimedia program for guidance of cultural assets and a way of use possessed in a national Chonju museum. Our system use ToolBook as authoring tools, designs system for convenience of user to the highest degree considered. Different point from others provides image enlargement and image processing ability for more delicate observance about surface of cultural assets. Image enlargement is composed of five steps, provides image processing such as bright control and edge enhancement and implement interface through combination of C-routine and ToolBook.

      • 유도전동기 속도제어를 위한 비선형 비례적분 제어기 설계

        This paper presents a robust speed control method of induction motors(IM) using a Non-linear PI controller(NPI). NPI is high gain controller in region of small error, and low gain controller in region of large error. So in steady state, system will be robust against variation of load torque. The simulation and experiment results confirm the validity of proposed control scheme.

      • 우편물 영상에서 우편번호 숫자의 자동 추출

        Recently, the postal automation becomes essential as the amount of mails to be delivered is large and increases rapidly. In this paper, we propose a method to extract the 6 ZIP code digits from the image of Korean regular letter envelope. The method first identifies the ZIP code area by searching for the 12 corners of 6 boxes enclosing the digits. Then we extract 6 digit images by eliminating the boxes. The experiment was performed with lots of actual letters delivered to our Department. The experimental result shows that the proposed method produces the digit images which could be successfully recognized by a digit recognizer.
