J Periodont Res 2011; 46: 193–203. © 2010 John Wiley & Sons A/S</I> </P><P><B>Background and Objective: </B> Recombinant human bone morphogenetic protein‐2 (rhBMP‐2) is a potent inducer for the regeneration of mineralized tissue, but has a limited effect on the regeneration of cementum and periodontal ligament (PDL). The aim of the present study was to determine the effects of rhBMP‐2 on the <I>in vitro</I> and <I>in vivo</I> biologic activity of well‐characterized human PDL stem cells (hPDLSCs) and to elucidate the underlying mechanism of minimal periodontal regeneration by rhBMP‐2.</P><P><B>Material and Methods: </B> hPDLSCs were isolated and cultured, and then transplanted into an ectopic subcutaneous mouse model using a carrier treated either with or without rhBMP‐2. Comprehensive histologic, histometric and immunohistochemical analyses were performed after an 8‐wk healing period. The effects of rhBMP‐2 on the adipogenic and osteogenic/cementogenic differentiation of hPDLSCs were also evaluated. The effect of rhBMP‐2 on both soluble and insoluble collagen synthesis was analyzed, and the expression of mRNA and protein for collagen types I, II, III and V was assessed.</P><P><B>Results: </B> In the present study, rhBMP‐2 promoted both adipogenic and osteogenic/cementogenic differentiation of hPDLSCs <I>in vitro</I>, and the <I>in vivo</I> potential of hPDLSCs to form mineralized cementum and organized PDL tissue was down‐regulated following treatment with rhBMP‐2. Collagen synthesis, which plays a crucial role in the regeneration of cementum and the periodontal attachment, was significantly reduced, with associated modification of the relevant mRNA and protein expression profiles.</P><P><B>Conclusion: </B> In summary, the findings of the present study suggest that enhanced adipogenic differentiation and inhibition of collagen synthesis by hPDLSCs appear to be partly responsible for the minimal effect of rhBMP‐2 on cementum and PDL tissue regeneration by hPDLSCs.</P>

      • Diagnostic usefulness of a T cell-based assay for latent tuberculosis infection in kidney transplant candidates before transplantation

All rights reserved</P><P>Background</P><P>The presence of latent tuberculosis (TB) infection (LTBI) should be evaluated before kidney transplantation. Although a new T cell-based assay for diagnosing LTBI gave promising results, this assay has not yet been compared with the tuberculin skin test (TST) for diagnosing LTBI in renal transplant candidates before transplantation.</P><P>Patients and methods</P><P>All adult patients admitted to a single institute for renal transplantation over a 1-year period were prospectively enrolled. A clinically predictive risk of LTBI was defined as: (i) recent close contact with a person with pulmonary TB; (ii) abnormal chest radiography; (iii) a history of untreated or inadequately treated TB; or (iv) a new infection (i.e., a recent conversion of TST).</P><P>Results</P><P>Of 209 renal recipients, 47 (22%) had a positive TST≥5 mm, 21 (10%) had a positive TST≥10 mm, 65 (30%) had a positive T-SPOT.<I>TB</I> test, and 25 (12%) had an indeterminate T-SPOT.<I>TB</I> test. The induration size of TST was significantly associated with a high positivity rate on T-SPOT.<I>TB</I> (<I>P</I><0.001). Agreement between T-SPOT.<I>TB</I> test and TST≥10 mm was fair (<I>k</I>=0.24, 95% confidence interval 0.11–0.36). However, neither univariate nor multivariate analysis showed any association between the clinical risk for LTBI and positivity on T-SPOT.<I>TB</I> or TST.</P><P>Conclusion</P><P>T-SPOT.<I>TB</I> test was more frequently positive than TST in renal transplant candidates. However, further longitudinal studies are awaited to determine whether the ability of T-SPOT.<I>TB</I> assay to detect LTBI in renal transplant recipients can better predict the development of TB than can TST after transplantation.</P>

        Potential roles of D-serine and serine racemase in experimental temporal lobe epilepsy

        <P>To confirm the roles of D-serinergic gliotransmission in epilepsy, we investigated the relationship between spatiotemporally specific glial responses and the D-serine/serine racemase system in mesial temporal structures following status epilepticus (SE). In control animals, D-serine and serine racemase immunoreactivities were detected mainly in astrocytes. After SE, D-serine and serine racemase immunoreactivities were increased in astrocytes. Double-immunofluorescence study revealed that up-regulation of serine racemase immunoreactivity was relevant not to D-serine immunoreactivity but to nestin or vimentin immunoreactivity. Neither D-serine nor serine racemase was found in naïve or reactive microglia. In addition, phosphorylated N-methyl-D-aspartate (NMDA) receptor subunit 1 (pNR1-Ser896) immunoreactivity in the hippocampus was increased compared with controls. Increased D-serine immunoreactivity showed direct correlation with the phosphorylation of Ser896 of NR1. Given the findings of our previous study, these findings suggest that D-serine and serine racemase in astrocytes may play roles in neuronal hyperexcitability via a cooperative activation of NMDA receptors. Furthermore, serine racemase may be involved in migration and differentiation of immature astrocytes, which is relevant to reactive astrogliosis. © 2010 Wiley-Liss, Inc.</P>

      • d-pinitol regulates Th1/Th2 balance via suppressing Th2 immune response in ovalbumin-induced asthma

        d-pinitol has been demonstrated to exert insulin-like and anti-inflammatory activities. However, its anti-allergic effect in the Th1/Th2 immune response is poorly understood. Recently, it was shown that T-bet and GATA-3 are master Th1 and Th2 regulatory transcription factors. In this study, we have attempted to determine whether d-pinitol regulates Th1/Th2 cytokine production, T-bet and GATA-3 gene expression in OVA-induced asthma model mice. We also examined to ascertain whether d-pinitol could influence eosinophil peroxidase (EPO) activity. After being sensitized and challenged with ovalbumin (OVA) showed typical asthmatic reactions. These reactions included an increase in the number of eosinophils in bronchoalveolar lavage (BAL) fluid, an increase in inflammatory cell infiltration into the lung tissue around blood vessels and airways, airway luminal narrowing, and the development of airway hyper-responsiveness (AHR). The administration of d-pinitol before the last airway OVA challenge resulted in a significant inhibition of all asthmatic reactions. Accordingly, this study may provide evidence that d-pinitol plays a critical role in the amelioration of the pathogenetic process of asthma in mice. These findings provide new insight into the immunopharmacological role of d-pinitol in terms of its effects in a murine model of asthma, and also broaden current perspectives in our understanding of the immunopharmacological functions of d-pinitol.

        D-π-A organic dyes with various bulky amine-typed donor moieties for dye-sensitized solar cells employing the cobalt electrolyte

        SGT dyes containing various amine-typed donors as triphenylamine, bis-fluorenylamine and bis-phenothiazinylamine as the electron donor and a cyanoacrylic acid moiety as electron acceptor in D-π-A system, were developed to use in dye-sensitized solar cells (DSSCs). The SGT-102 dye containing bis-fluorenylamine had a better prevented charge recombination than other SGT dyes; leading to improvement in V<SUB>oc</SUB>. As a result, the conversion efficiency of 7.22% was achieved with a J<SUB>sc</SUB> of 12.1mAcm<SUP>-2</SUP>, V<SUB>oc</SUB> of 865mV and a FF of 69.1 for the DSSC employing a dye containing the bulky bis-fluorenylamine donor unit, while the DSSC based on a dye containing the bulky bis-phenothiazinylamine donor unit showed a lower J<SUB>sc</SUB> and V<SUB>oc</SUB>, leading to a lower efficiency of 5.16%, due to slow charge recombination associated with differently geometric structure orientations.

        Decreased biliary excretion of tributylmethyl ammonium in cholestyramine pretreated rats due to reduced formation of ion-pair complexes with hepatic bile salts

        <P>The hypothesis that higher molecular weight (MW) quaternary ammoniums (QAs) form lipophilic ion-pair complexes with bile salts in the liver, and are subsequently excreted into bile via a canalicular transporter, P-gp, was re-examined in the present study for its validity. The biliary excretion of tributylmethyl ammonium (TBuMA), a QA with a MW of 200, in bile salt-depleted rats was determined. Depletion was induced by a daily oral administration of a resin, cholestyramine, at a dose of 0.5 g/kg for 2 consecutive weeks, which decreased the concentration of total bile salts in the liver by 38%. When TBuMA was administered intravenously (12 µmol/kg) to these rats, the plasma level, area under the plasma concentration-time curve (AUC), systemic clearance (CL) and volume of distribution (V<SUB>ss</SUB>) of the compound remained unchanged, whereas bile flow (23.03 vs 16.94 µl/min, p<0.05) and biliary clearance (CL<SUB>bile</SUB>, 12.75 vs 5.34 ml/min/kg, p<0.01) were decreased significantly. These results implied the biliary clearance of TBuMA in rats with bile salt depletion was significantly decreased as a result of decreased ion-pair complexation of TBuMA. The above results are consistent with our hypothesis and the existence of a MW threshold (i.e. 200±50 for rats) for the biliary excretion of QAs. Copyright © 2007 John Wiley & Sons, Ltd.</P>

        The high speed deformation behavior of TRIP steels and HSLA steels has been investigated at strain rates ranging from 10^(-2) to 10³/sec. The effects of metallurgical factors such as prestrain (2%) and bake hardening condition (170℃, 20 min) on tensile properties at various strain rates are being evaluated. Ultimate tensile strength (UTS), total elongation, strain rate sensitivity, and absorbed energy are reported. In general, UTS increases with increasing strain rate, that is, it shows positive strain rate sensitivity. And strain rate sensitivity increases sharply over 100/sec. Strength increases with increasing the amount of prestrain. The bake hardening treatment slightly influences UTS values. The absorbed energy decreases with increasing prestrain but it is not affected by bake hardening treatment. Received February 14, 2005)

      • A variety of local structure patterns and their hybridization for accurate eye detection

        In this paper, existing local structure patterns (LSPs) are reviewed and categorized into two types: intensity-based LSPs (I-LSPs) and gradient-based LSPs (G-LSPs). I-LSPs include local binary pattern (LBP), modified census transform (MCT) and generalized binary pattern (GBP) methods that compare the intensities of differently selected neighboring pixels within a 3x3 neighborhood with differently-formed reference intensities, encoding 256, 511, and 19,162 binary patterns respectively. G-LSPs include local gradient pattern (LGP), modified gradient pattern (MGP), and generalized gradient pattern (GGP) methods that compare the gradient magnitudes of differently selected neighboring pixels within a 3x3 neighborhood with differently-formed reference gradients, encoding 256, 511, and 19,162 binary patterns respectively. We extend all these LSPs to multi-scale block LSPs (MB-LSPs) that concatenate multiple block-based LSPs. Finally, we propose several hybrid LSPs that combine I-LSPs and G-LSPs by means of the AdaBoost feature selection. In experiments using AR564, BioID, ColorFERET and LFW databases to evaluate the eye detection accuracy of the proposed LSPs, the I-LSPs were good for detecting the eyes in low quality images, the G-LSPs were good for detecting the eyes in high quality images, and the hybrid LSPs achieve state-of-the-art eye detection accuracy across image qualities.

        Carrier transport of inverted quantum dot LED with PEIE polymer

        An inverted-type quantum-dot light-emitting-diode (QD LED), employing low-work function organic material polyethylenimine ethoxylated (PEIE) as electron injection layer, was fabricated by all solution processing method, excluding anode electrode. From transmission electron microscopy (TEM) and scanning electron microscopy (SEM) studies, it was confirmed that CdSe@ZnS QDs with 7nm size were uniformly distributed as a monolayer on PEIE layer. In this inverted QD LED, two kinds of hybrid organic materials, [poly (9,9-di-n-octyl-fluorene-alt-benzothiadiazolo)(F8BT)+poly(N,N'-bis(4-butylphenyl)-N,N'-bis(phenyl)benzidine (poly-TPD)] and [4,4'-N,N'-dicarbazole-biphenyl (CBP)+poly-TPD], were adopted as hole transport layer having high highest occupied molecular orbital (HOMO) level for improving hole transport ability. At a low-operating voltage of 8V, the device emits orange and red spectral radiation with high brightness up to 2450 and 1420cd/m<SUP>2</SUP>, and luminance efficacy of 1.4cd/A and 0.89cd/A, respectively, at 7V applied bias. Also, the carrier transport mechanisms for the QD LEDs are described by using several models to fit the experimental I-V data.
