      • Expression of epithelial cell adhesion molecule and proliferating cell nuclear antigen in diethylnitrosamine-induced hepatocarcinogenesis in mice

        <P>To clarify the role of stem cells in hepatocarcinogenesis, the expression of epithelial cell adhesion molecule (EpCAM) and proliferating cell nuclear antigen (PCNA) was investigated in mouse hepatic tumors and embryonic cell lineages. Ten ICR mice were treated with diethylnitrosamine (DEN) at 14 days of age and sacrificed at 36 weeks subsequent to DEN treatment to obtain the hepatic tumors. Mouse embryonic stem cells, hepatic progenitor cells and hepatocyte-like cells, representing 0, 22 and 40 days of differentiation, respectively, were treated <I>in vitro</I> with DEN at four doses (0, 1, 5 and 15 mM; G1, G2, G3 and G4, respectively) for 24 h and RNA was isolated. A total of 71 hepatic tumors were obtained from the DEN-treated mice. EpCAM expression was increased mainly in hepatic tumor cells, although it was also detected in the surrounding visually normal cells. Double staining showed that EpCAM and PCNA were co-expressed in numerous tumor cells. <I>In vitro</I>, EpCAM expression was significantly different for G4 at day 0 (P<0.01) and for G2, G3 and G4 at day 40 (P<0.01) compared with the control (G1) at the corresponding time-point. PCNA expression was significantly different for G3 and G4 at day 0 (P<0.01), for G2, G3 and G4 at day 22 (P<0.01) and for G2 at day 40 (P<0.01) compared with G1 at the corresponding time-point. In summary, the expression of EpCAM and PCNA was increased in DEN-induced tumors and the expression of EpCAM and PCNA was altered by DEN treatment in cultured cells. This suggests that EpCAM expression may be modulated in the progeny of adult liver stem cells during their differentiation toward hepatocytes and may be increased during DEN-induced hepatocarcinogenesis.</P>

        Insulin-Secreting Cells from Human Eyelid-Derived Stem Cells Alleviate Type I Diabetes in Immunocompetent Mice

        <P>Various attempts have been made to develop stem cell-based therapy to alleviate type I diabetes using animal models. However, it has been a question whether human insulin produced from explanted cells is solely responsible for the normoglycemia of diabetic animals. In this study, we isolated neural crest-like stem cells from the human eyelid fat and examined their therapeutic potentials for diabetes. The human eyelid adipose-derived stem cells (HEACs) displayed characteristics of neural crest cells. Using a two-step culture condition combined with nicotinamide, activin, and/or GLP-1, we differentiated HEACs into insulin-secreting cells and examined in vivo effects of differentiated cells by transplantation experiments. Following differentiation in vitro, HEACs released insulin and c-peptide in a glucose-dependent manner. Upon their transplantation under kidney capsules of streptozotocin-treated immunocompetent mice, we observed normalization of hyperglycemia in 10 of 20 recipient mice until sacrifice after 2 months. Only the human, but not the mouse, insulin and c-peptide were detected in the blood of recipient mice. Removal of the kidneys transplanted with HEACs resulted in a sharp increase of blood glucose level. Removed kidney tissues showed distinct expression of various human genes including insulin, and colocalization of the human insulin and the human nuclear protein in many cells. However, they showed diminished or null expression of some immune-related genes. In conclusion, human insulin alone produced from eyelid-derived stem cells following differentiation into insulin-secreting cells and transplantation could normalize type I diabetes in mice.</P>

      • gsGator: an integrated web platform for cross-species gene set analysis

        <P><B>Background</B></P><P>Gene set analysis (GSA) is useful in deducing biological significance of gene lists using a priori defined gene sets such as gene ontology (GO) or pathways. Phenotypic annotation is sparse for human genes, but is far more abundant for other model organisms such as mouse, fly, and worm. Often, GSA needs to be done highly interactively by combining or modifying gene lists or inspecting gene-gene interactions in a molecular network.</P><P><B>Description</B></P><P>We developed <I>gsGator</I>, a web-based platform for functional interpretation of gene sets with useful features such as cross-species GSA, simultaneous analysis of multiple gene sets, and a fully integrated network viewer for visualizing both GSA results and molecular networks. An extensive set of gene annotation information is amassed including GO & pathways, genomic annotations, protein-protein interaction, transcription factor-target (TF-target), miRNA targeting, and phenotype information for various model organisms. By combining the functionalities of <I>Set Creator, Set Operator and Network Navigator</I>, user can perform highly flexible and interactive GSA by creating a new gene list by any combination of existing gene sets (intersection, union and difference) or expanding genes interactively along the molecular networks such as protein-protein interaction and TF-target. We also demonstrate the utility of our interactive and cross-species GSA implemented in gsGator by several usage examples for interpreting genome-wide association study (GWAS) results. gsGator is freely available at http://gsGator.ewha.ac.kr.</P><P><B>Conclusions</B></P><P>Interactive and cross-species GSA in gsGator greatly extends the scope and utility of GSA, leading to novel insights via conserved functional gene modules across different species.</P>

        한국과 독일의 문화민주주의와 문화센터 : 한국 생활문화센터와 독일 사회문화센터의 비교분석

        본 논문은 문화민주주의를 실천하고 있는 한국의 생활문화센터와 독일의 사회문화센터의 비교연구이다. 세계적으로 문화민주주의 이념이 크게 확대되면서 한국과 독일도 문화민주주의 실천을 위하여 생활문화센터와 사회문화센터를 도입하여 적극적으로 노력하고 있다. 문화민주주의 이념이 거시적인 명분에 그치지 않고 구체적으로 각 국가에서 실제 어떻게 이루어지고 있는가를 분석하기 위해, 한국과 독일의 생활문화와 사회문화의 기본개념, 생활문화센터와 사회문화센터의 설립․운영 주체, 주민주도 운영방식을 비교하였다. 첫째, 한국 생활문화와 독일 사회문화는 모두 확장된 문화개념으로 접근하며 삶의 총체로서의 문화를 중시한다. 이것은 문화의 민주화에서의 문화가 고급예술과 엘리트예술로 한정되었다면 문화민주주의에서의 문화는 보다확장된 개념인 삶 중심으로 접근하고 있음을 보여준다. 둘째, 한국 생활문화센터와 독일 사회문화센터는 설립․운영주체의 차이가 있다. 한국은 정부의 문화정책으로 인하여 생활문화센터가 조성되었고, 운영주체는 지자체 직영, 민간위탁, 주민자율형의 3가지 구조로 나뉜다. 반면 독일 사회문화센터는 민간에 의해 설립되고 운영되는 구조로 되어 있다. 한국과 독일 모두 생활문화센터와 사회문화센터를 통해 문화민주주의 이념에 따라 주민이 주체적으로 시설운영에 참여하는 방식을 가지고 있지만 한국은 주민에 의한 의견수렴과정에 참여하면서 소극적 문화민주주의를보여주고 독일은 주민이 설립과 운영을 직접적으로 주도하면서 적극적 문화민주주의가 나타난다. 셋째, 한국 생활문화센터와 독일 사회문화센터는 모두 주민주도 운영방식을 지향하고 있으나 주민 주도성에 대한 차이가 나타난다. 한국 생활문화센터의 다양한 운영구조로 인해 주민주도 운영방식이 각각 다르게 나타나고 있다. 독일 사회문화센터는 주민의 의사결정구조와 조직구조에 동참함으로써 주민이 재정, 운영, 프로그램 등에 참여하여 적극적인형태를 보여준다. 한국과 독일의 문화민주주의에 대한 실천은 모두 같은 지향을 가지고 있지만 각국의 실천과정에서 다른 양상을보여주는데 한국은 정부로부터 시작했기에 다양한 운영주체를 통해 지역적 상황에 맞는 다양한 방식을 취하면서제도적으로 주민에 의한 민주적 결정구조가 정착되지 못하고 있는 반면 독일은 정부지원 없이 민간에서 시작된 만큼 운영과 재정의 문제는 오롯이 주민의 공동책임이 된다. 그로인해 사회문화센터를 운영함에 있어서 주민들의 동의와 합의가 전제되고 주민에 의한 의사결정구조를 제도적으로 보장하여 지속성을 담보해내고 있다. This study compares the living cultural centers in Korea and the sociocultural centers in Germany, which practice cultural democracy. With the significant expansion of cultural democracy ideology worldwide, Korea and Germany are actively striving to practice cultural democracy by introducing living cultural centers and sociocultural centers. Specifically, to analyze how each country’s efforts are being made, this study compares the basic concepts of living culture and social culture in Korea and Germany, the subject of establishment and operation of living cultural centers and sociocultural centers, and resident-led operations respectively. First, both Korean living culture and German social culture approach with expanded cultural concepts, emphasizing culture as a whole of life. Second, there is a difference between the subjects of establishment and operation of the Korean living cultural centers and the German sociocultural centers. Third, although both the Korean living cultural centers and the German sociocultural centers are oriented toward resident-led operation, there is a difference in resident initiative. The practice of cultural democracy in Korea and Germany both have the same orientation, but show different aspects in the practice process of each country. In Korea, starting from the government, the democratic decision structure by the residents has not been institutionally established although various methods are adopted according to local circumstances through various operating subjects. On the other hand, in Germany, since it was started by the private sector without government support, operation and financial issues are the sole responsibility of the residents. Therefore, in the operation of the sociocultural centers, the consent and agreement of the residents is premised, and the decision-making structure by the residents is systematically guaranteed to ensure continuity.

      • Rapid fecal cytokeratin-19 test and fecal occult blood test in screening for gastrointestinal diseases.

        <P>To evaluate the screening power of the fecal cytokeratin-19 test (CK-19) and the fecal occult blood test (FOBT), we performed rapid fecal CK-19 and FOBT tests on 515 stool samples from patients with various GI diseases and 814 stool samples from control patients. The rapid fecal CK-19 test (developed by DiNonA Research Institute, Seoul, Korea) is based on gold immunochromatography and has a sensitivity of 1 ng/ml. The positive rate of the FOBT was 2.1% in controls, 14.0% in GI cancer patients, 3.5% in GI inflammation patients, 11.7% in bone marrow transplant (BMT) patients, and 6.0% in childhood diarrhea patients. Except for the GI inflammation patients, the patients' positive rates for FOBT were all higher than the controls (p <0.05). The positive rate of the fecal CK-19 test was 8.2% in controls, 42.1% in GI cancer patients, 66.0% in GI inflammation patients, 84.8% in BMT patients, and 19.9% in childhood diarrhea patients. In all of the patient groups, positive rates for the CK-19 test were higher than in the controls (p <0.05). The fecal CK-19 test was more frequently positive (42.1%) in GI cancer patients than the FOBT; if both tests were used, the sensitivity was 49.1%. The fecal CK-19 test (but not the FOBT) gave a higher positive rate in GI inflammation patients than the controls, suggesting that the CK-19 test could serve as a screening test for GI inflammation. The highest positive rate of the fecal CK-19 test was found in the BMT group, indicating that significant GI epithelial desquamation had occurred. Although the positive rate of the fecal CK-19 test in childhood diarrhea patients was higher than in the controls, it was much lower than in adults with GI inflammatory disease. Evidently, children with GI inflammation do not desquamate as much intestinal epithelium as adult patients with GI inflammation. This study shows that the rapid fecal GK-19 test, used in conjunction with the FOBT, may be a valuable screening technique for GI diseases and can assist physicians in the differential diagnosis of GI diseases.</P>

        ESG정보와 공매도의 관련성에 대한 실증 연구

        In Korean capital market, the short selling has increased continuously and the impact of short selling also has increased. The literature on short selling involves conflict results that it can increase market efficiency by reflecting negative information of the company into the market, while reducing efficiency by inducing excessive stock price decline. This study investigates whether short sellers, as informed investors, consider corporate’s ESG score in making investment decision. We examine whether the companies with high ESG score have lower weight on short selling. Meanwhile, to consider the impact of chaebol firms on capital market, we analyze the chaebol and non-chaebol firms separately. Empirical result are as follows. For the total sample, the ESG score and Short Selling shows positive relations but not significant. On the other hand, the chaebol firms showed a significantly positive correlations, while the non-chaebol firms showed a negative correlation. As far as we know, this study is the first empirical research investigating the impact of ESG score on short selling in Korea. Especially, we analyzed the impact for chaebol and non-chaebol seperately, widen the scope of academic research. 최근 자본시장에서 공매도거래가 증가하면서 공매도가 시장에 미치는 영향이 커지고 있다. 공매도에 대한 연구들은 기업의 부정적 정보를 시장에 반영시켜 시장효율성을 증가시킬 수 있는 반면 과도한 주가 하락을 유발시켜 효율성을 감소시킨다는 상반된 결과가 혼재되어 있으며 최근에도 다양한 연구가 이루어지고 있다. 본 연구에서는 공매도가 기업의 지속가능경영지표 정보들을 시장에 반영시켜 정보비대칭을 완화시키는지 알아보고자 한다. 공매도자들은 지속가능경영지표의 측정치인 ESG점수가 높은 기업의 경우 회계투명성이 높고 정보비대칭이 완화될 것으로 보아 공매도가 적게 발생할 것으로 예측하였다. 한편, 본연구에서는 우리 자본시장의 특징인 재벌기업의 영향을 고려하여 재벌·비재벌기업을 별도로 구분하여 분석하였다. 2014년에서 2016년 까지 한국증권선물거래소의 코스닥시장과 유가증권시장에 상장된 기업 중 ESG 점수가 있는 기업을 대상으로 분석한 결과, 전체표본의 경우 ESG지수와 공매도 간에 양의 관계를 보였으나 유의하지 않다. 그러나 재벌기업을 별도로 분리하여 분석한 결과 재벌기업의 경우 ESG 점수와 공매도는 유의한 양(+)의 상관관계를 보인 반면 비재벌기업의 경우 유의한 음(-)의 상관관계를 보였다. 본연구는 공매도거래가 ESG 점수와 공매도에 미치는 영향을 재벌, 비재벌기업으로 나누어 살펴봄으로써 ESG와 재벌기업에 대한 기존 연구를 확장시켰다.

      • High Performance of Low Band Gap Polymer-Based Ambipolar Transistor Using Single-Layer Graphene Electrodes

        <P>Bottom-contact bottom-gate organic field-effect transistors (OFETs) are fabricated using a low band gap pDTTDPP-DT polymer as a channel material and single-layer graphene (SLG) or Au source/drain electrodes. The SLG-based ambipolar OFETs significantly outperform the Au-based ambipolar OFETs, and thermal annealing effectively improves the carrier mobilities of the pDTTDPP-DT films. The difference is attributed to the following facts: (i) the thermally annealed pDTTDPP-DT chains on the SLG assume more crystalline features with an edge-on orientation as compared to the polymer chains on the Au, (ii) the morphological features of the thermally annealed pDTTDPP-DT films on the SLG electrodes are closer to the features of those on the gate dielectric layer, and (iii) the SLG electrode provides a flatter, more hydrophobic surface that is favorable for the polymer crystallization than the Au. In addition, the preferred carrier transport in each electrode-based OFET is associated with the HOMO/LUMO alignment relative to the Fermi level of the employed electrode. All of these experimental results consistently explain why the carrier mobilities of the SLG-based OFET are more than 10 times higher than those of the Au-based OTFT.
