<P>The nature and importance of the DNA repair system in the chloroplasts of higher plants under oxidative stress or UV radiation‐induced genotoxicity was investigated via gain‐of‐functional approaches exploiting bacterial RecAs. For this purpose, transgenic tobacco (<I>Nicotiana tabacum</I>) plants and cell suspensions overexpressing <I>Escherichia coli</I> or <I>Pseudomonas aeruginosa</I> RecA fused to a chloroplast‐targeting transit peptide were first produced. The transgenic tobacco plants maintained higher amounts of chloroplast DNA compared with wild‐type (WT) upon treatments with methyl viologen (MV), a herbicide that generates reactive oxygen species (ROS) in chloroplasts. Consistent with these results, the transgenic tobacco leaves showed less bleaching than WT following MV exposure. Similarly, the MV‐treated transgenic Arabidopsis plants overexpressing the chloroplast RecA homologue RECA1 showed weak bleaching, while the <I>recA1</I> mutant showed opposite results upon MV treatment. In addition, when exposed to UV‐C radiation, the dark‐grown <I>E. coli</I> RecA‐overexpressing transgenic tobacco cell suspensions, but not their WT counterparts, resumed growth and greening after the recovery period under light conditions. Measurements of UV radiation‐induced chloroplast DNA damage using <I>Dra</I>I assays (Harlow et al. 1994) with the chloroplast <I>rbcL</I> DNA probe and quantitative PCR analyses showed that the transgenic cell suspensions better repaired their UV‐C radiation‐induced chloroplast DNA lesions compared with WT. Taken all together, it was concluded that RecA‐overexpressing transgenic plants are endowed with an increased chloroplast DNA maintenance capacity and enhanced repair activities, and consequently have a higher survival tolerance to genotoxic stresses. These observations are made possible by the functional compatibility of the bacterial RecAs in chloroplasts.</P>

The On-machine measurement (OMM), which measures a workpiece during or after the machining process in the machining center, has the advantage of measuring the workpiece directly within the work space without moving it. However, the path generation procedure used to determine the measuring sequence and variables for the complex features of a target workpiece has the limitation of requiring time-consuming tasks to generate the measuring points and mostly relies on the proficiency of the on-site engineer. In this study, we propose a touch-probe path generation method using similarity analysis between the feature vectors of three-dimensional (3-D) shapes for the OMM. For the similarity analysis between a new 3-D model and existing 3-D models, we extracted the feature vectors from models that can describe the characteristics of a geometric shape model; then, we applied those feature vectors to a geometric histogram that displays a probability distribution obtained by the similarity analysis algorithm. In addition, we developed a computer-aided inspection planning system that corrects non-applied measuring points that are caused by minute geometry differences between the two models and generates the final touch-probe path.

Recently, there has been a gradual increase in the number of people who are interested in cycling, leading to an increasing number of cycling equipment consumers. However, many bicyclists get hurt because of their lack of knowledge about the right size of bicycle for their body. Although it is necessary for a rider to fit their bicycle to prevent injury, they reject a fitting service because of the long hours and high cost. In this study, we propose a bicycle fitting system that uses a depth camera to improve the limitations of existing manual fitting systems. With the defined formula, the system calculates the size of the bicycle using body image information extracted by a depth camera and visualizes a customized bicycle for a specific consumer. This system will not only save the customer time and money, but will prevent injury from the use of a bicycle that does not fit.

Compound 7b provides a possible lead for further structural modification towards potential β-amyloid aggregation inhibitors to treat Alzheimer's disease.

Intracellular retrograde transport in eukaryotic cells relies exclusively on the molecular motor cytoplasmic dynein 1. Unlike its counterpart, kinesin, dynein has a single isoform, which raises questions about its cargo specificity and regulatory mechanisms. The precision of dynein-mediated cargo transport is governed by a multitude of factors, including temperature, phosphorylation, the microtubule track, and interactions with a family of activating adaptor proteins. Activating adaptors are of particular importance because they not only activate the unidirectional motility of the motor but also connect a diverse array of cargoes with the dynein motor. Therefore, it is unsurprising that dysregulation of the dynein-activating adaptor transport machinery can lead to diseases such as spinal muscular atrophy, lower extremity, and dominant. Here, we discuss dynein motor motility within cells and in in vitro, and we present several methodologies employed to track the motion of the motor. We highlight several newly identified activating adaptors and their roles in regulating dynein. Finally, we explore the potential therapeutic applications of manipulating dynein transport to address diseases linked to dynein malfunction.
