Extensive research supported the therapeutic potential of curcumin, a naturally occurring compound, as a promising cytokinesuppressive anti-inflammatory drug. This study aimed to investigate the synergistic anti-inflammatory and anti-cytokine activities by combining 6-shogaol and 10-shogaol to curcumin, and associated mechanisms in modulating lipopolysaccharides and interferon-ɣ-induced proinflammatory signaling pathways. Our results showed that the combination of 6-shogaol-10-shogaolcurcumin synergistically reduced the production of nitric oxide, inducible nitric oxide synthase, tumor necrosis factor and interlukin- 6 in lipopolysaccharides and interferon-γ-induced RAW 264.7 and THP-1 cells assessed by the combination index model. 6-shogaol-10-shogaol-curcumin also showed greater inhibition of cytokine profiling compared to that of 6-shogaol-10-shogaol or curcumin alone. The synergistic anti-inflammatory activity was associated with supressed NFκB translocation and downregulated TLR4-TRAF6-MAPK signaling pathway. In addition, SC also inhibited microRNA-155 expression which may be relevant to the inhibited NFκB translocation. Although 6-shogaol-10-shogaol-curcumin synergistically increased Nrf2 activity, the anti-inflammatory mechanism appeared to be independent from the induction of Nrf2. 6-shogaol-10-shogaol-curcumin provides a more potent therapeutic agent than curcumin alone in synergistically inhibiting lipopolysaccharides and interferon-γ induced proinflammatory mediators and cytokine array in macrophages. The action was mediated by the downregulation of TLR4/TRAF6/MAPK pathway and NFκB translocation.
Adaptive compensation method for real-time hybrid simulation of train-bridge coupling system

Real-time hybrid simulation (RTHS) was applied to investigate the train-bridge interaction of a high-speed railway system, where the railway bridge was selected as the numerical substructure, and the train was physically tested. The interaction between the two substructures was reproduced by a servo-hydraulic shaking table. To accurately reproduce the high-frequency interaction responses ranging from 10-25Hz using the hydraulic shaking table with an inherent delay of 6-50ms, an adaptive time series (ATS) compensation algorithm combined with the linear quadratic Gaussian (LQG) was proposed and implemented in the RTHS. Testing cases considering different train speeds, track irregularities, bridge girder cross-sections, and track settlements featuring a wide range of frequency contents were conducted. The performance of the proposed ATS+LQG delay compensation method was compared to the ATS method and RTHS without any compensation in terms of residual time delays and root mean square errors between commands and responses. The effectiveness of the ATS+LQG method to compensate time delay in RTHS with high-frequency responses was demonstrated and the proposed ATS+LQG method outperformed the ATS method in yielding more accurate responses with less residual time delays.

In a wind tunnel experiment employing a reduced scale model, Reynolds number (Re) can hardly be respected. Its effects on the aerodynamics of closed-box bridge decks have been the subject of research in recent years. Stonecutters Bridge in Hong Kong is a cable-stayed bridge having an unprecedented central span of 1018m. The issue of Re sensitivity was raised early in the design phase of the deck of Stonecutters Bridge. The objective of this study is to summarise the results of various wind tunnel experiments in order to demonstrate the effect of Re on the steady state aerodynamic force coefficients. The results may provide an insight on the choice of scale for section model experiments in bridge design projects. Computational Fluid Dynamics (CFD) analysis of forces on bridge deck section was also carried out to see how CFD results are compared with experimental results.
