        Analyses of alternative polyadenylation: from old school biochemistry to high-throughput technologies

        Alternations in usage of polyadenylation sites during transcription termination yield transcript isoforms from a gene. Recent findings of transcriptome-wide alternative polyadenylation (APA) as a molecular response to changes in biology position APA not only as a molecular event of early transcriptional termination but also as a cellular regulatory step affecting various biological pathways. With the development of high-throughput profiling technologies at a single nucleotide level and their applications targeted to the 3`-end of mRNAs, dynamics in the landscape of mRNA 3`-end is measureable at a global scale. In this review, methods and technologies that have been adopted to study APA events are discussed. In addition, various bioinformatics algorithms for APA isoform analysis using publicly available RNA-seq datasets are introduced. [BMB Reports 2017; 50(4): 201-207]

        mTOR-coordinated Post-Transcriptional Gene Regulations: from Fundamental to Pathogenic Insights

        Post-transcriptional regulations of mRNA transcripts such as alternative splicing and alternative polyadenylation can affect the expression of genes without changing the transcript levels. Recent studies have demonstrated that these post-transcriptional events can have significant physiological impacts on various biological systems and play important roles in the pathogenesis of a number of diseases, including cancers. Nevertheless, how cellular signaling pathways control these post-transcriptional processes in cells are not very well explored in the field yet. The mammalian target of rapamycin complex 1 (mTORC1) pathway plays a key role in sensing cellular nutrient and energy status and regulating the proliferation and growth of cells by controlling various anabolic and catabolic processes. Dysregulation of mTORC1 pathway can tip the metabolic balance of cells and is associated with a number of pathological conditions, including various types of cancers, diabetes, and cardiovascular diseases. Numerous reports have shown that mTORC1 controls its downstream pathways through translational and/or transcriptional regulation of the expression of key downstream effectors. And, recent studies have also shown that mTORC1 can control downstream pathways via post-transcriptional regulations. In this review, we will discuss the roles of post-transcriptional processes in gene expression regulations and how mTORC1-mediated post-transcriptional regulations contribute to cellular physiological changes. We highlight post-transcriptional regulation as an additional layer of gene expression control by mTORC1 to steer cellular biology. These emphasize the importance of studying post-transcriptional events in transcriptome datasets for gaining a fuller understanding of gene expression regulations in the biological systems of interest.

        Alternative Polyadenylation of mRNAs: 3'-Untranslated Region Matters in Gene Expression

        Almost all of eukaryotic mRNAs are subjected to polyadenylation during mRNA processing. Recent discoveries showed that many of these mRNAs contain more than one polyadenylation sites in their 3' untranslated regions (UTR) and that alternative polyadenylation (APA) is prevalent among these genes. Many biological processes such as differentiation, proliferation, and tumorigenesis have been correlated to global APA events in the 3' UTR of mRNAs, suggesting that these APA events are tightly regulated and may play important physiological roles. In this review, recent discoveries in the physiological roles of APA events, as well as the known and proposed mechanisms are summarized. Perspective for future directions is also discussed.

        대만 인플루언서 라이브방송에서 서비스품질 요인과 제품특성이 신규 브랜드 수용의도에 미치는 영향에 관한 연구

        현재 SNS 라이브방송이 활성화로 인해 인플루언서 생방송에 기반을 둔 온라인 서비스가 전세계 주요 소비 트렌드가 되고 있다. 본 연구는 인터넷 환경에서 인플루언서 방송을 통해 소비자들이 느끼는 인플루언서 방송에 대한 서비스품질, 고객만족, 제품특성과 수용의도 간의 관계들을 검증하고자 한다. 본 연구는 대만 SNS 라이브방송을 체험한 이용자들을 대상으로 2020년 6월 29일부터 8월 30일 까지 구글 설문지를 통하여 조사를 실시하였으며, 총 253부를 실증분석에 사용하였다. 수집된 자료는 SPSS 25.0을 통해 분석을 실시하였다. 실증분석 결과를 요약하면 다음과 같다. 첫째, 대만 인플루언서 라이브방송의 서비스품질 요인(신뢰성, 유형성, 응답성, 확신성, 공감성)은 생방송 만족도에 유의한 영향을 미치는 것으로 나타났다. 둘째, 대만 인플루언서 라이브방송의 제품특성이 제품 만족도에 유의한 영향을 미치는 것으로 나타났다. 셋째, 대만 인플루언서 라이브방송에서 생방송 만족도와 제품 만족도는 신규 브랜드 수용의도에 유의한 영향을 미치는 것으로 나타났다. 본 연구를 통해 대만 인플루언서 라이브 방송의 서비스품질 요인 및 제품특성을 파악함으로써 생방송 및 제품 만족도를 향상시키고 신규 브랜드 수용의도를 증가시키기 위한 효율적인 마케팅 전략 방안을 수립하는데 유용한 자료를 제공할 수 있을 것이다. Currently, due to the activation of SNS live broadcasting, online services based on influencer live broadcasting are becoming a major consumption trend around the world. This study aims to verify the relationship between service quality, customer satisfaction, product characteristics, and acceptance intention for influencer broadcasting based on nfluencer broadcasting experiences in an Internet environment. This study conducted a survey of users who experienced live broadcasting on social media in Taiwan from June 29 to August 30, 2020, and a total of 253 copies were used for empirical analysis. The collected data were analyzed through SPSS 25.0. The results of the empirical analysis are summarized as follows. First, it was found that the service quality factors (reliability, tangibility, responsiveness, certainty, and empathy) of Taiwan’s influencer live broadcast had a significant effect on live broadcast satisfaction. Second, it was found that the product characteristics of Taiwan’s influencer live broadcasting had a significant effect on product satisfaction. Third, it was found that live broadcasting satisfaction and product satisfaction had a significant effect on the acceptance intention of new brands in Taiwan’s influencer live broadcasting. This study will provide useful data for establishing efficient marketing strategies to improve live and product satisfaction and increase acceptance of new brands by identifying service quality factors and product characteristics of Taiwan’s influencer Live Broadcasting.

      • Synthesis and Characterization of 9 nm Pt–Ni Octahedra with a Record High Activity of 3.3 A/mg<sub>Pt</sub> for the Oxygen Reduction Reaction

        <P>Nanoscale Pt–Ni bimetallic octahedra with controlled sizes have been actively explored in recent years owning to their outstanding activity for the oxygen reduction reaction (ORR). Here we report the synthesis of uniform 9 nm Pt–Ni octahedra with the use of oleylamine and oleic acid as surfactants and W(CO)<SUB>6</SUB> as a source of CO that can promote the formation of {111} facets in the presence of Ni. Through the introduction of benzyl ether as a solvent, the coverage of both surfactants on the surface of resultant Pt–Ni octahedra was significantly reduced while the octahedral shape was still attained. By further removing the surfactants through acetic acid treatment, we observed a specific activity 51-fold higher than that of the state-of-the-art Pt/C catalyst for the ORR at 0.93 V, together with a record high mass activity of 3.3 A mg<SUB>Pt</SUB><SUP>–1</SUP> at 0.9 V (the highest mass activity reported in the literature was 1.45 A mg<SUB>Pt</SUB><SUP>–1</SUP>).         Investigations of δ -Doped In0.52Al0.48As/InxGa1-xAs/InP HEMTs with Different Channel Structures

        The device characteristics of -doped In0.52Al0.48As/InGaAs/InP high-electron-mobility transistors (HEMTs) with different channel designs, grown by using the low-pressure metal-organic chemical-vapor deposition (LP-MOCVD) technique, have been studied. The InxGa1.xAs channel structures include a step-graded channel (SGC) with x = 0.56/0.53/0.5, a lattice-matched channel (LMC) with x = 0.53, and an inverse linearly-graded channel (ILGC) with x = 0.5 ! 0.56. Improvements in the device’s threshold, extrinsic conductance, saturation current density, gate-voltage swing (GVS), output conductance, voltage gain, and high-frequency and high-temperature performances have been comprehensively investigated and compared with respect to the specific channel design.

        Experimental study and modeling of masonry-infilled concrete frames with and without CFRP jacketing

        Most existing concrete structures in Taiwan are considered nonductile due to insufficient transverse reinforcement and poor detailing of frame elements. Such features are fairly typical for buildings constructed prior to 1997, at which time the local building code was revised based on ACI 318-95. Among these structures, many contain perimeter or partition walls made of concrete or clay brick for architectural purposes. These walls, though treated as non-structural components in common design practice, could affect the structural behavior of the buildings during an earthquake. To study the behavior of such structures under seismic load, experiments were conducted on concrete frames of various configurations to show the force-deformation relationships, damage patterns, and other characteristics of the frames. For further interest, similar units with columns jacketed by carbon-fiber-reinforced-polymer (CFRP) were also tested to illustrate the effectiveness of this technique in the retrofit of concrete frames.
