Prognostic Model Built on Blood-based Biomarkers in Patients with Metastatic Colorectal Cancer

Background: We had previously showed that the neutrophil lymphocyte ratio (NLR), ${\gamma}$-glutamyl transpeptidase (GGT) and carcinoembryonic antigen (CEA) are prognostic factors for metastatic colorectal cancer (mCRC) patients. In this study we developed a prognostic model based on these three indices. Materials and Methods: A total of 243 patients who were initially diagnosed as mCRC between 2005 and 2010 in the Sun Yat-sen University Cancer Center were studied. The endpoint was overall survival (OS). Results: NLR>3, elevated GGT and elevated CEA were confirmed as independent risk factors which could predict poor prognosis. Patients could be divided into three groups according to the number of risk factors they had. Those with two or three were defined as the high risk group, individuals with one risk factor as the modest risk group and patients without risk factor as the low risk group. The OS values for these three groups were 16.2 months (2.80~68.8), 24.2 months (4.07~79.0), and 37.2 months (12.6~87.8), respectively (p<0.001). Conclusions: We developed a simple but useful model based on NLR, GGT and CEA to provide prognostic information to clinical practice in highly selected mCRC patients. Further prospective and multi-center studies are warranted to test our model.

Previous work reveals that the adult Chinese citrus fly, Bactrocera minax (Enderlein), emerges from the ground in citrus orchards and presumably disperses to alternate food sources in neighboring shrubs and trees, where it stays and becomes reproductively mature, and then later returns to the orchard to lay eggs. We investigated the trajectories of early emerged adult Chinese citrus flies, which were tracked with portable harmonic radar in 2013, 2014, 2015, and 2016 in four different habitats in Hubei province of China that presumably provided food sources for adults. This study we investigated whether early emerged adult Chinese citrus flies migrate into forests. A total of 47 adults were tracked following release in a citrus orchard. Our results showed that a total of 33 adults released in a citrus orchard flew into the forest, and 14 adults that disappeared during the tracking process in the citrus orchard. A total of 10 adults (in 2014) that were tracked and released in the forest flew to the forest. Of the adult flies investigated, 54.5% flew directly into the forest, while the remaining 45.5% finally arrived in the forest after repeatedly adjusting their trajectories. When there were Castanea mollissima Blume (Fagaceae) in the surrounding forest, the proportion of frequency of adult landing on the plant represented 58.3% and 88.5% of landing on all species in 2014 and in 2015, respectively. We conclude that early emerged adult Chinese citrus flies in the mosaic-type citrus orchard of the hilly terrain landform dispersed into adjacent forest.

Objective: To compare the efficacy of taxane-based regimens in the first line setting retrospectively in Chinese patients with recurrent and/or metastatic esophageal cancer. Methods: We analyzed 102 recurrent and/or metastatic esophageal cancer patients who received taxanes-based regimens in a first-line setting from January 2009 to December 2013. Sixteen (15.7%) patients were administered Nab-PTX based chemotherapy and 86 patients (84.3%) received paclitaxel (PTX) or docetaxel (DTX) based chemotherapy. Patients in the PTX/DTX group could be further divided into TP (71 patients) and TPF (15 patients) groups. Results: The objective response rate (ORR) of all patients was 20.6%, and the disease control rate (DCR) was 67.6%. The median overall survival (OS) was 10.5 months (95% CI 10.1-16.4) and the median progression-free survival (PFS) was 6.04 months (95% CI 5.09-7.91). The DCR was higher in the TPF group than the TP group (93.3% vs. 59.1%; p = 0.015 ). There were no significant differences in ORR, OS, and PFS among Nab-PTX, TPF and TP groups. Conclusions: The three regimens of Nab-PTX based, TP and TPF proved active in a first line setting of Chinese patients with recurrent and/or metastatic esophageal cancer, and should thus be regarded as alternative treatments.

Carboxylesterases (CXEs) belong to a family of metabolic enzymes that are widely distributed in insects and other organisms and can rapidly degrade the components of sex pheromones and plant volatiles with an acetate functional group. The common cutworm, Spodoptera litura, is an important agricultural pest around the world, causing vast economic losses every year. The female sex pheromones of S. litura comprise four acetates, Z9, E11-14:OAc; Z9, E12-14:OAc; Z9-14:OAc; and E11-14:OAc, but the degradation mechanisms of these components are not well understood. By analysing previously obtained transcriptomic data of the sex pheromone glands,we identified a total of 24 putative CXE genes in S. litura. Gene expression patterns and phylogenetic analysis revealed 5 genes with antennae-specific or biased expression, and clusteredwith genes showed involvement in the degradation of sex pheromones or other detoxification in other insects. SlitCXE10was expressed specifically in the antennae of both sexes, and SlitCXE14, 17, 19, and 21 had high antenna biased expression. Interestingly, RT-PCR and qPCR tests indicated that SlitCXE24 had significantly higher expression in PG than in other tissue, and that it could be a potential candidate gene for sex pheromone degradation in PG. This study is the first to provide solid background information for the further elucidation of sex pheromone degradation, and ultimately provides potential targets for the disruption of sexual communication in S. litura for new pest management.

The antioxidant and antiproliferation effects of fractions from methanol extract of Sparganiun stoloniferum were investigated in this study. The ethyl acetate fraction of the methanol extract showed significant antioxidant activity against α,?α-diphenyl-picrylhydrazyl (DPPH) and hydroxyl ( · OH) radical scavenging, and it showed greater reducing power than the n-hexane, dichloromethane, n-butanol and water fractions. Qualitative HPLC analysis was performed on the extractions. The high content of phenolics in ethyl acetate fractions were believed to be responsible for its antioxidant activity, which was the strongest in all five fractions. Moreover, the ethyl acetate fraction showed a dose-dependent inhibition of cell proliferation with an EC?? of 139.21 ± 0.54 μg · mL?¹ against the human colon cell, HT-29. The anti-proliferative effects of the ethyl acetate fraction were associated with apoptosis. Semi-quantitative RT-PCR analysis indicates that, after treatment with the ethyl acetate fraction, the expression of caspase-3, p53, and Bax in HT-29 cells was significantly up-regulated and c-myc expression was induced. These findings demonstrate that the ethyl acetate fraction of S. stoloniferum has potent antioxidant and antiproliferation effects and induces apoptosis. Thus, the extract of S. stoloniferum may be considered as a potential functional food resource and complementary medicinal supplement.
