        Gum arabic-encapsulated gold nanoparticles modulate hypoxamiRs expression in tongue squamous cell carcinoma

        Background Oral tongue squamous cell carcinoma (OTSCC) is a popular aggressive malignancy of the oral cavity. Despite advances in OTSCC therapy, the overall 5-year survival rate is low. The tumor microenvironment resistance factors lead to chemotherapy failure, especially intratumoral hypoxia. HIF-1α, the main protein in hypoxia pathway, influences cell survival and angiogenesis. Hypoxia/HIF-1α system is a potential strategic target in cancer therapeutics. The expression of hypoxia-regulating miRNAs (hypoxamiRs; miR-210 and miR-21), regulators of HIF-1α, is high in OTSCC. Gum Arabic-encapsulated gold nanoparticles (GA-AuNPs) have been reported as promising modality in cancer treatment. Objective This study aimed to investigate the influence of GA-AuNPs on the hypoxia regulators in OTSCC (CAL-127 cells). GA-AuNPs cytotoxicity assessed by MTT assay; cell death mode was detected by dual DNA staining; monitoring of cellular hypoxia was followed by pimonidazole; miR-210 and miR-21 expression was assessed by qPCR; and their targets (HIF-1α and c-Myc) assayed by immunocytofluorescence and ELISA, respectively. Results GA-AuNPs (75–80 nm; λmax of ~ 540 nm) reduced cell viability with IC50 of 392.3 and 247.3 µg/ml after 24 h and 48 h, respectively. Cell death was mainly due to apoptosis. CAL-27 cells exhibited high hypoxia and the treatment with GA-AuNPs inhibited this hypoxia in a dose-dependent manner, as detected by pimonidazole. GA-AuNPs (30% IC50) significantly reduced miR-210 and miR-21 expression. HIF-1α and c-Myc were inhibited by GA-AuNPs (30% IC50, for 48 h). Conclusion The study findings may suggest GA-AuNPs as a promising carrier for chemotherapies to diminish intratumoral hypoxia-stimulated resistance.       • Tethers tension force effect in the response of a squared tension leg platform subjected to ocean waves

        The tension leg platform (TLP) is one of the compliant structures which are generally used for deep water oil exploration. With respect to the horizontal degrees of freedom, it behaves like a floating structure moored by vertical tethers which are pretension due to the excess buoyancy of the platform, whereas with respect to the vertical degrees of freedom, it is stiff and resembles a fixed structure and is not allowed to float freely. In the current study, a numerical study for square TLP using modified Morison equation was carried out in the time domain with water particle kinematics using Airy's linear wave theory to investigate the effect of changing the tether tension force on the stiffness matrix of TLP's, the dynamic behavior of TLP's; and on the fatigue stresses in the cables. The effect was investigated for different parameters of the hydrodynamic forces such as wave periods, and wave heights. The numerical study takes into consideration the effect of coupling between various degrees of freedom. The stiffness of the TLP was derived from a combination of hydrostatic restoring forces and restoring forces due to cables. Nonlinear equation was solved using Newmark's beta integration method. Only uni-directional waves in the surge direction was considered in the analysis. It was found that for short wave periods (i.e., 10 sec.), the surge response consisted of small amplitude oscillations about a displaced position that is significantly dependent on tether tension force, wave height; whereas for longer wave periods, the surge response showed high amplitude oscillations that is significantly dependent on wave height, and that special attention should be given to tethers fatigue because of their high tensile static and dynamic stress.

        Spinal versus general anesthesia for Cesarean section in patients with sickle cell anemia

        Background: Sickle cell anemia (SCA) increases the rate of maternal and fetal complications. This pilot study was designed to compare the maternal and fetal outcomes of spinal versus general anesthesia (GA) for parturients with SCA undergoing cesarean delivery. Methods: Forty parturients with known SCA scheduled for elective Cesarean delivery were randomized into spinal anesthesia (n = 20) and GA groups (n = 20). Perioperative hemodynamic parameters were recorded. Postpartum complications were followed up. Opioid consumption was calculated. Blood loss during surgery and the number of patients who received intraoperative or postpartum blood transfusion were recorded. Patient satisfaction with the type of anesthesia was assessed. The Apgar score at 1 and 5 min, neonatal admission to the intensive care unit, and mortality were also recorded. Results: Blood loss was significantly higher in the GA than spinal group (P = 0.01). However, the number of patients who received an intraoperative or postpartum blood transfusion was statistically insignificant. Significantly more patients developed intraoperative hypotension and bradycardia in the spinal than GA group. Opioid use during the first 24 h was significantly higher in the GA than spinal group (P < 0.0001). More patients had vaso-occlusive crisis in the GA than spinal group without statistical significance (P = 0.4). There was one case of acute chest syndrome in the GA group. No significant differences were observed in postoperative nausea and/or vomiting, patient satisfaction, or hospital length of stay. Neonatal Apgar scores were significantly better in the spinal than GA group at 1 and 5 min (P = 0.006 and P = 0.009, respectively). Neonatal intensive care admission was not significantly different between the two groups, and there was no neonatal mortality. Conclusions: Spinal anesthesia may have advantages over GA in parturients with SCA undergoing Cesarean delivery.

      • Experimental shear strengthening of GFRC beams without stirrups using innovative techniques

        Eighteen (18) (120×300×2200 mm) beams were prepared and tested to evaluate the shear strength of Glass Fiber Reinforced Concrete (GFRC) beams with no shear reinforcement, and evaluate the effectiveness of various innovative strengthening systems to increase the shear capacity of the GFRC beams. The test variables are the amount of discrete glass fiber (0.0, 0.6, and 1.2% by volume of concrete) and the type of longitudinal reinforcement bars (steel or GFRP), the strengthening systems (externally bonded (EB) sheet, side near-surface mounted (SNSM) bars, or the two together), strengthening material (GFRP or steel) links, different configurations of NSM GFRP bars (side bonded links, full wrapped stirrups, side C-shaped stirrups, and side bent bars), link spacing, link inclination angle, and the number of bent bars. The experimental results showed that adding the discrete glass fiber to the concrete by 0.6%, and 1.2% enhanced the shear strength by 18.5% and 28%, respectively in addition to enhancing the ductility. The results testified the efficiency of different strengthening systems, where it is enhanced the shear capacity by a ratio of 28.4% to 120%, and that is a significant improvement. Providing SNSM bent bars with strips as a new strengthening technique exhibited better shear performance in terms of crack propagation, and improved shear capacity and ductility compared to other strengthening techniques. Based on the experimental shear behavior, an analytical study, which allows the estimation of the shear capacity of the strengthened beams, was proposed, the results of the experimental and analytical study were comparable by a ratio of 0.91 to 1.15.
