<P>A decade of studies on middle cortex (MC) formation in the root endodermis of Arabidopsis (Arabidopsis thaliana) have revealed a complex regulatory network that is orchestrated by several GRAS family transcription factors, including SHORT-ROOT (SHR), SCARECROW (SCR), and SCARECROW-LIKE3 (SCL3). However, how their functions are regulated remains obscure. Here we show that mutations in the SEUSS (SEU) gene led to a higher frequency of MC formation. seu mutants had strongly reduced expression of SHR, SCR, and SCL3, suggesting that SEU positively regulates these genes. Our results further indicate that SEU physically associates with upstream regulatory sequences of SHR, SCR, and SCL3; and that SEU has distinct genetic interactions with these genes in the control of MC formation, with SCL3 being epistatic to SEU. Similar to SCL3, SEU was repressed by the phytohormone GA and induced by the GA biosynthesis inhibitor paclobutrazol, suggesting that SEU acts downstream of GA signaling to regulate MC formation. Consistently, we found that SEU mediates the regulation of SCL3 by GA signaling. Together, our study identifies SEU as a new critical player that integrates GA signaling with transcriptional inputs from the SHR-SCR-SCL3 module to regulate MC formation in the Arabidopsis root.</P>

Glehnia littoralis (GL) is widely used as an oriental medicine for cough, fever, stroke and other disease conditions. However, the anti-cancer properties of GL on MCF-7 human breast cancer cells have not been investigated. In order to elucidate anti-cancer properties and underlying cell death mechanisms, MCF-7cells ($5{\times}10^4/well$) were treated with Glehnia littoralis root extract at 0-400 ug/ml. A hot water extract of GL root inhibited the proliferation of MCF-7 cells in a dose-dependent manner. Analysis of the cell cycle after treatment of MCF-7 cells with increasing concentrations of GL root extract for 24 hours showed significant cell cycle arrest in the G1 phase. RT-PCR and Western blot analysis both revealed that GL root extract significantly increased the expression of p21 and p27 with an accompanying decrease in both CDK4 and cyclin D1. Our reuslts indicated that GL root extract arrested the proliferation of MCF-7 cells in G1 phase through inhibition of CDK4 and cyclin D1 via increased induction of p21 and p27. In summary, the current study showed that GL could serve as a potential source of chemotherapeutic or chemopreventative agents against human breast cancer.

The design and fabrication of a bilayer Ti6Al4V–Ti6Al4V/30Ta component were performed by using the powder metallurgyprocess and solid-state sintering as the consolidation step. Phase change and sintering densifcation of the component werestudied by dilatometry. The addition of 30 vol% of Ta to the Ti6Al4V matrix had a noticeable efect over the microstructuraland mechanical properties of the alloy, which showed decrements of up to 12.2 and 21.5% in nano-hardness and elastic modulus, respectively. The decrement of these properties strongly afected the wear and corrosion performance of the component. Special attention was focused on the intermediate zone between layers denoted by a transition zone, which presented betterwear response because of the properties and microstructure caused by the gradient difusion of Ta. Ti6Al4V/30Ta alloyshowed an improved corrosion behaviour compared to Ti6Al4V alloy, decreasing 2.4 times their susceptibility to corrosionand about two orders of magnitude their corrosion rate. The bilayer component in this study is proposed as an alternative todecrease the consumption of expensive materials with improved properties.
