      • The prevalence of BRCA mutations among familial breast cancer patients in Korea: results of the Korean Hereditary Breast Cancer study.

        <P>The primary aim of this study was to estimate the prevalence of BRCA1/2 mutations among familial breast cancer (BC) patients in Korea. We analyzed 775 familial BC patients who were enrolled in the Korean Hereditary Breast Cancer (KOHBRA) study and treated at 36 institutions between May 2007 and May 2010. Patients with familial BC were defined as BC patients with family histories of BC or ovarian cancer (OC) in any relatives. All probands received genetic counseling and BRCA genetic testing was performed after obtaining informed consent. The mean age of BC diagnosis was 43.6 years. The numbers of probands with family histories of BC only and OC only were 682 and 93, respectively. The overall prevalence of the BRCA mutation among familial BC patients was 21.7 % (BRCA1 9.3 % and BRCA2 12.4 %). Subgroup analyses observed prevalences of the BRCA mutation as follows: 19.6 % among patients with BC family history only (BRCA1 7.6 % and BRCA2 12.0 %) and 36.6 % among patients with OC family history only (BRCA1 21.5 % and BRCA2 15.1 %). Most of the subgroups satisfied the 10 % probability criteria to undergo BRCA testing. However, the prevalence of the BRCA mutations among subgroups that had 2 BC patients in a family with both age at diagnosis of more than 50 years old did not reach the 10 % criteria (4.1 %). Korean familial BC patients are good candidates for BRCA testing even when they have family histories of single breast cancers. However, proband age at diagnosis should be carefully considered when selecting patients for testing.</P>

        The Application of Delphi-AHP Method in the Priority of Policies for Expanding the Use of Artificial Intelligence

        Governments around the world are actively establishing strategies and initiatives to spread the use of artificial intelligence (AI), for AI is not a mere new technology, but is an innovative technology that brings about extensive changes in industrial and social structures and is a core engine that will lead the 4th Industrial Revolution. The South Korean government has also been paying attention to AI as a technology and tool for innovative growth, but its application to the industries is still rather sluggish. The government has prepared multifarious AI-related policies with the aim of constructing South Korea as an AI powerhouse, but there is no clear strategy on which detailed policies to implement first and which industries to apply AI preferentially. With these limitations of South Korea's AI policies in mind, this paper analyzed the priorities of industries in AI adoption and the priorities of AI-related national policies, using Delphi-AHP method for 30 top-level AI experts in South Korea. The results of analysis show that AI application is urgent and necessary in the fields of medical/healthcare, public and safety, and manufacturing, which seems to reflect the peak of the COVID-19 crisis in the second half of 2020 at the time of the investigation. And it turns out that policies related to AI talent cultivation, data, and R&D investment are important and urgent above all in order for organizations to apply AI. This suggests that strategies are required to focus limited national resources on these industries and policies first.

        <P>A Disintegrin And Metalloprotease (ADAM) family members expressed in male reproductive tissues are divided phylogenetically into three major groups. In the present study, we analyzed six ADAMs in one of the groups (ADAMs 4, 6, 24, 26, 29, and 30) of which function is largely unknown. Our results showed that most of the ADAMs undergo unique processing during sperm maturation and are located at the surface of sperm head. We found that the levels of ADAM4 and ADAM6 are dramatically reduced in Adam2 and Adam3 knockout sperm defective in various fertilization processes. We observed premature processing of ADAM4 in the Adam3-null mice. Furthermore, we obtained a result showing complex formation of ADAM6 with ADAM2 and ADAM3 in testis. Taken together, these results disclose involvement of ADAM4 and ADAM6 in a reproductive ADAM system that functions in fertilization.</P>

      • Segmental hair analysis and estimation of methamphetamine use pattern.

        <P>The aim of this study was to investigate whether the results of segmental hair analysis can be used to estimate patterns of methamphetamine (MA) use. Segmental hair analysis for MA and amphetamine (AP) was performed. Hair was cut into the hair root, consecutive 1?cm length segments and 1-4?cm length segments. Whole hair was also analyzed. The hair samples were incubated for 20?h in 1?mL methanol containing 1?% hydrochloric acid after washing the hair samples. Hair extracts were evaporated and derivatization was performed using trifluoroacetic anhydride in ethylacetate at 65?C for 30?min. Derivatized extract was analyzed by gas chromatography/mass spectrometry. The 15 subjects consisted of 13 males and two females and their ages ranged from 25 to 42 (mean, 32). MA and AP concentrations in the whole hair ranged from 3.00 to 105.10?ng/mg (mean, 34.53) and from 0.05 to 4.76?ng/mg (mean, 2.42), respectively. Based on the analysis of the 1?cm length segmental hair, the results were interpreted in a way to distinguish between continuous use of MA (n?=?10), no recent but previous use of MA (n?=?3), and recent but no previous use of MA (n?=?2). Furthermore, the individuals were interpreted as light, moderate, and heavy users based on concentration ranges previously published.</P>

      • Segmental hair analysis for 11-nor-δ?-tetrahydrocannabinol-9-carboxylic acid and the patterns of cannabis use.

        <P>Cannabis is the most widely abused drug in the world. The purpose of this study is to detect 11-nor-9-carboxy-δ?-tetrahydrocannabinol (THCCOOH) in segmental hair and to evaluate the patterns of cannabis use. We investigated the relationship between the concentrations of THCCOOH in hair and the self-reported use data and the route of administration. For this purpose, the hair samples were washed, digested with 1 mL of 1 M NaOH at 85C for 30 min along with the internal standard, THCCOOH-d3 (2.5 pg/mg) and extracted in 2 mL of n-hexane-ethyl acetate (9:1) twice after adding 1 mL of 0.1N sodium acetate buffer (pH = 4.5) and 200 ?L of acetic acid. The organic extract was transferred and evaporated and the mixture was derivatized with 50 ?L of pentafluoropropionic anhydride and 25 ?L of pentafluoropropanol for 30 min at 70C. Reconstituted final extract was injected into the gas chromatography-tandem mass spectrometer operating in the negative chemical ionization mode. In segmental hair analysis, the concentrations of THCCOOH decreased from the proximal to distal segments. The concentrations of THCCOOH in hair and the self-reported dose and frequency of administration from cannabis users were not well correlated because of the low accuracy and reliability of the self-reported data. However, this study provides preliminary information on the dose and frequency of administration among cannabis users in our country.</P>

        청소년의 비행성향이 진로성숙에 미치는 영향

        학교는 청소년들이 가장 많은 시간을 보내는 곳으로 청소년들의 자아형성 및 미래의 진로 결정에 중요한 역할을 한다. 본 연구는 청소년의 비행성향이 진로성숙에 미치는 영향과 이 관계에서 학교환경에 대한 인식의 매개효과 검증을 목적으로 하였다. 이를 위해 한국복지패널조사 4차년도 아동용부가조사의 중학교 1∼3학년에 재학 중인 605명을 분석대상으로 하였다. 기술분석과 상관분석으로 응답자 및 주요 변수들의 특성을 파악하였으며, 다중회귀분석과 sobel test를 통해 연구모형 및 학교환경에 대한 인식의 매개효과를 검증하였다. 주요 연구결과로 자아존중감, 친구애착, 비행성향이 진로성숙에 유의미한 영향을 주는 것으로 나타났으며, 학교환경에 대한 인식은 비행성향과 진로성숙의 관계를 완전 매개하는 것으로 나타났다. 즉, 비행성향이 진로성숙에 직접적인 영향을 미치기보다는 학교환경에 대한 인식을 통해 영향을 주는 것을 확인하였다. 이러한 연구결과를 바탕으로 한 본 연구의 의의는 다음과 같다. 첫째, 기존 선행연구에서 많이 다루지 못했던 진로성숙과 비행성향의 인과적 관계를 밝혀내었다. 둘째, 비행과 진로성숙의 관계에 학교환경에 대한 인식의 매개효과를 검증함으로써 청소년들의 진로성숙을 높이기 위해 학생들에 대한 개별적 접근뿐 아니라 학교환경의 개선 및 청소년들의 학교생활에 적응할 수 있도록 하는 개입의 중요성을 밝혀내었다. It is at school that adolescents spend most of their time, and this plays a significant role as they develop their confidence and make decisions about their future careers. The purpose of this study is to examine the effect of tendencies towards juvenile delinquency on career maturity, and the mediating effect of fitness in school between juvenile delinquency and career maturity. The subjects were 605 middle school students (1st-3rd grade) who participated in a 4th year of additional research for adolescents undertaken by the Korea Welfare Panel. Descriptive analysis and correlations analysis were used to investigate variables and the characteristics of the respondents. Multiple regression analysis and the Sobel test were utilized for creating the research model and measuring the mediating effects of the fitness on school. According to these results, the fitness on school fully mediated the relationship between juvenile delinquency and career maturity. That is, the tendency towards juvenile delinquency influenced career maturity through the fitness on school rather than directly influencing it. As a result, based on these findings, it is suggested that there is a correlation between career maturity and juvenile delinquency, which is something which has not been examined so far in previous research. These results also reveal the importance of individual characteristics and the school approach to adolescents as ways to increase overall career maturity by examining the mediating effects of school adaptation.

        온라인 커뮤니티 교환구조, 결속력, 교환혜택에 관한 연구

        온라인 커뮤니티에서 교환구조가 커뮤니티 결속력에 미치는 영향에 관한 연구는 매우 드물다. 더욱이 이 연구들은 서로 상충된 연구결과를 보고하고 있다. 본 연구는 이러한 연구결과를 재조명하기 위해 사회적 교환이론에 근거하여 커뮤니티 내 교환구조를 직접적 및 일반적 교환구조로 분류하고, 일반적 교환구조의 결정요인으로 지식의 분산도를 포함하였다. 그리고 상충된 연구결과를 설명하기 위해 교환혜택을 조절변수로 도입하였다. 본 연구는 네이버에서 활동하는 380개의 온라인 커뮤니티에서 데이터를 수집하여 분석하였다. 그 결과 지식의 분산도는 일반적 교환구조를 촉진하는 것으로 나타났다. 그리고 직접적, 일반적 교환구조 모두 커뮤니티 결속력에 긍정적 영향을 미치나, 이 관계는 교환혜택의 수준에 따라 조절되는 것으로 나타났다. 즉, 교환혜택이 높은 그룹에서는 일반적 교환구조가 직접적 교환구조보다 커뮤니티 결속력에 미치는 영향이 더 큰 반면, 교환혜택이 낮은 그룹에서는 두 교환구조의 영향력 사이에 통계적으로 유의미한 차이가 없는 것으로 나타났다. Only a few studies investigated the relationship between exchange structure and solidarity in online communities and even reported conflicting results. This study attempts to shed lights on this relationship by introducing a moderating variable, i.e., exchange benefits, based on the social exchange theory. Also, distributed knowledge is considered as a determinant of exchange structure. The results of a field study from 380 online communities show that both direct and generalized exchange structures have positive effects on community solidarity, but their effects vary depending on the level of exchange benefits. When exchange benefits are high, generalized exchange influences community solidarity more than direct exchange does, whereas at a low level of benefits there are no significant differences between the two exchange structures. Also distributed knowledge turns out to be a significant influencer of generalized exchange.

        <P>To determine the mechanisms of spermatogenesis, it is essential to identify and characterize germ cell-specific genes. Here we describe a protein encoded by a novel germ cell-specific gene, Mm.290718/ZFP541, identified from the mouse spermatocyte UniGene library. The protein contains specific motifs and domains potentially involved in DNA binding and chromatin reorganization. An antibody against Mm.290718/ZFP541 revealed the existence of the protein in testicular spermatogenic cells (159 kDa) but not testicular and mature sperm. Immunostaining analysis of cells at various stages of spermatogenesis consistently showed that the protein is present in spermatocytes and round spermatids only. Transfection assays and immunofluorescence studies indicate that the protein is localized specifically in the nucleus. Proteomic analyses performed to explore the functional characteristics of Mm.290718/ZFP541 showed that the protein forms a unique complex. Other major components of the complex included histone deacetylase 1 (HDAC1) and heat-shock protein A2. Disappearance of Mm.290718/ZFP541 was highly correlated with hyperacetylation in spermatids during spermatogenesis, and specific domains of the protein were involved in the regulation of interactions and nuclear localization of HDAC1. Furthermore, we found that premature hyperacetylation, induced by an HDAC inhibitor, is associated with an alteration in the integrity of Mm.290718/ZFP541 in spermatogenic cells. Our results collectively suggest that the Mm.290718/ZFP541 complex is implicated in chromatin remodeling during spermatogenesis, and we provide further information on the previously unknown molecular mechanism. Consequently, we re-designate Mm.290718/ZFP541 as 'SHIP1' representing spermatogenic cell HDAC-interacting protein 1.</P>
