        Intravenous Administration of Substance P Attenuates Mechanical Allodynia Following Nerve Injury by Regulating Neuropathic Pain-Related Factors

        This study aimed to investigate the analgesic effect of substance P (SP) in an animal model of neuropathic pain. An experimental model of neuropathic pain, the chronic constriction injury (CCI) model, was established using ICR mice. An intravenous (i.v.) injection of SP (1 nmole/kg) was administered to the mice to examine the analgesic effects of systemic SP on neuropathic pain. Behavioral testing and immunostaining was performed following treatment of the CCI model with SP. SP attenuated mechanical allodynia in a time-dependent manner, beginning at 1 h following administration, peaking at 1 day post-injection, and decaying by 3 days post-injection. The second injection of SP also increased the threshold of mechanical allodynia, with the effects peaking on day 1 and decaying by day 3. A reduction in phospho-ERK and glial fibrillary acidic protein (GFAP) accompanied the attenuation of mechanical allodynia. We have shown for the first time that i.v. administration of substance P attenuated mechanical allodynia in the maintenance phase of neuropathic pain using von Frey's test, and simultaneously reduced levels of phospho-ERK and GFAP, which are representative biochemical markers of neuropathic pain. Importantly, glial cells in the dorsal horn of the spinal cord (L4-L5) of SP-treated CCI mice, expressed the anti-inflammatory cytokine, IL-10, which was not seen in vehicle saline-treated mice. Thus, i.v. administration of substance P may be beneficial for improving the treatment of patients with neuropathic pain, since it decreases the activity of nociceptive factors and increases the expression of anti-nociceptive factors.

        Characterization of sphere-forming HCT116 clones by whole RNA sequencing

        Purpose: To determine CD133<SUP>+</SUP> cells defined as cancer stem cells (CSCs) in colon cancer, we examined whether CD133<SUP>+</SUP> clones in HCT116 demonstrate known features of CSCs like sphere-forming ability, chemodrug-resistance, and metastatic potential. Methods: Magnetic cell isolation and cell separation demonstrated that <1% of HCT116 cells expressed CD133, with the remaining cells being CD133<SUP>-</SUP> clones. In colon cancer cells, radioresistance is also considered a CSC characteristic. We performed clonogenic assay using 0–4 Gy g-irradiation. Results: Interestingly, there were no differences between HCT116 parental and HCT116 CD133<SUP>+</SUP> clones when the cells comprised 0.5% of the total cells, and CD133<SUP>-</SUP> clone demonstrated radiosensitive changes compared with parental and CD133<SUP>+</SUP> clones. Comparing gene expression profiles between sphere-forming and nonforming culture conditions of HCT116 subclones by whole RNA sequencing failed to obtain specific genes expressed in CD133<SUP>+</SUP> clones. Conclusion: Despite no differences of gene expression profiles in monolayer attached culture conditions of each clone, sphere-forming conditions of whole HCT116 subclones, parental, CD133<SUP>+</SUP>, and CD133<SUP>-</SUP> increased 1,761 coding genes and downregulated 1,384 genes related to CSCs self-renewal and survival. Thus, spheroid cultures of HCT116 cells could be useful to expand colorectal CSCs rather than clonal expansion depending on CD133 expressions.

        베이비 부머의 고용불안정이 건강에 미치는 영향: 통제감의 조절·매개효과를 중심으로

        본 연구의 목적은 베이비 부머가 경험하는 고용불안정이 건강에 미치는 영향을 조사하고, 이러한 영향이 통제감에 의해 조절 및 매개되는지를 검증하는 것이다. 이를 위해 한국 베이비부머 패널의 1차년도 및 2차년도 자료를 활용하여, 1차시의 고용불 안정 정도 및 2년 사이 고용불안정 정도의 변화가 조사 참여자들의 건강상태(신체증 상)에 어떤 영향을 미치는지, 이러한 관계를 통제감이 조절하는지, 그리고 2년 사이 통제감 수준의 변화가 고용불안정과 건강 간의 관계를 매개하는지를 살펴보았다. 분석 결과 고용불안정 정도가 높을수록, 또한 2년간 고용불안감이 증가할수록 신체증 상을 경험하는 빈도가 높아졌다. 통제감은 고용불안정 정도가 신체증상에 미치는 영향을 조절하지는 못했지만 고용불안정 변화가 신체증상에 미치는 영향을 조절 하였다. 한편 고용불안정 정도 및 그 변화가 신체증상에 미치는 영향은 2년간의 통제감 변화에 의해 부분적으로 매개되었다. 이러한 결과는 베이비 부머에게 있어 고용불안 정이 건강상의 문제를 초래하는 주요한 스트레스원이며, 개인의 심리적 자원으로서 통제감을 증진하는 것뿐만 아니라 고용안정성을 보장하기 위한 거시적 차원의 접근이 함께 이루어져야 함을 시사한다. This study examines the effects of job insecurity on baby boomers` health and the extent to which these effects are moderated and mediated by a sense of mastery. Analyses are based on data from the 2010 (Time 1; T1) and 2012 (Time 2; T2) waves of the Korean Baby Boomers Panel Study. The results show that higher levels of job insecurity at T1 and changes in the level of job insecurity from T1 to T2 are both associated with more physical symptoms at T2. A sense of mastery moderates the relationship between a change in the level of job insecurity and physical symptoms, but it does not moderate the effect of job insecurity at T1. Also, higher levels of job insecurity at T1 and changes in the level of job insecurity lead to a decrease in mastery, which in turn increases physical symptoms. The findings suggest that job insecurity is a major stressor which leads to adverse health outcomes. Moreover, the findings underscore the need to develop social policies which ensure employment stability as well as interventions to increase psychological resources.

        Linear Dependence of Energy Band-gap on the Composition of MgxZn1-xTe Single Crystals Studied by Photocurrent Measurements

        Single crystals structures of MgxZn1−xTe (0 < x< 0.48) were grown for various Mg mole compositions by using a vertical Bridgman method. The optical properties of the grown crystal were characterized using photocurrent (PC) and persistent photoconductivity (PPC) measurements. As a result of the PC spectra, single crystalline MgxZn1−xTe (x = 0) showed an energy band gap of 2.380 and 2.260 eV at 4.2 and 294 K, respectively. The PC peak blue-shifts with increasing Mg mole fraction and shows linear dependence of energy band gap, Eg(x) = b + (0.8)x. The extrapolation shows the energy band gaps of MgTe as 3.18 and 3.06 eV at temperatures of 4.2 and 294 K, respectively. Furthermore, the PC peak red-shifts with increasing temperature, and the temperature coefficient is given by a value of dEg/dT = – (5.6 6.1) × 10−4 eV/K for the temperature range above 100 K. In the PPC measurements, the decay times of the PPC spectra strongly depended on the Mg content. These results clearly indicate a change in crystal structure with increasing Mg mole fraction.

        이미지 검색 실패에 나타난 이미지 요구와 맥락에 관한 분석

        이용자의 이미지요구 규명은 이미지 정보 서비스와 시스템 효율성 증진을 위해 필수적인 요소이다. 최근에는 이미지요구 분석에 있어서 다양한 맥락 요소를 접목하여 다층적인 관점에서 이미지요구를 규명하고자 하는 시도들이 있다. 본 연구는 이러한 최근 연구의 흐름과 맥을 같이 하며, 이용자의 이미지요구를 규명하기 위해서 총 70건의 이미지 검색 실패 사례를 분석하였다. 그 결과 이미지 검색 실패 사례에 나타난 이미지요구는 구체적, 일반적/지칭 가능한 요구가 주를 이루는 것으로 나타났으며, 이러한 결과는 선행 연구와 유사한 것으로써 검색 실패한 이미지요구의 특성으로 볼 수 있다. 또한 이미지요구에 배태된 맥락요소에 관한 분석결과를 살펴보면, 업무/과제의 목표와 이용목적에 관련된 맥락요소가 주를 이루는 것으로 나타났다. 이용목적의 맥락을 보다 상세하게 살펴보면, 일러스트레이션 이용목적이 높은 비중으로 나타났다. 이미지 검색 질의 특성은 고유어와 정련어의 쌍으로 분석할 때 고유어/정련어, 비고유어/정련어, 고유어/비정련어, 고유어/정련어 쌍 순서로 비중을 차지하는 것으로 나타났다. 검색 실패 이미지요구의 특성으로써 구체적이거나 일반적/지칭 가능한 요구이며 이는 비고유어/비정련어 쌍을 특징적으로 살펴볼 수 있다. 맥락의 관점에서는 목표 중심이며 일러스트레이션 이용목적이 주된 맥락 요소임을 알 수 있다. 이러한 결과는 이미지 검색 시스템이나 서비스 개선을 위한 시사점이 될 수 있다. As a way of identifying users' image needs for improved effectiveness of image search, there have been recent research approaches to examine contextual factors in image needs with multiple perspectives. In this line of research, this study examined a total of 70 unsuccessful image searches for the purpose of investigating users' image needs. In order to achieve the purpose of this study, in particular, the characteristics of image needs, contextual factors on image needs, and image queries were investigated. The findings of this study demonstrated that information needs from the failed image searches are categorized primarily into specific and general/nameable categories. More importantly, these information needs are embedded with multiple contextual factors, primarily, task purpose and use purpose. With an analysis of detailed use purposes for image, illustration use was found most in this data set. For query analysis, the type of unique/refined image query was revealed primarily. As the results of this study were found similar to the findings of previous studies, it is possible to characterize the image needs from the failed image searches. In addition, the findings of this study are expected to be useful to the design and service of image retrieval.
