      • Induction of heme oxygenase-1 protects against podocyte apoptosis under diabetic conditions

        Heme oxygenase-1 (HO-1) is an anti-oxidant enzyme normally upregulated in response to oxidant injury. Here we determined the role of HO-1 in podocyte apoptosis in glomeruli of streptozotocin-treated rats and in immortalized mouse podocytes cultured in media containing normal or high glucose. HO-1 expression, its activity, the ratio of Bax/Bcl-2 protein, and active caspase-3 fragments were all significantly higher in isolated glomeruli of diabetic rats and in high glucose–treated podocytes. These increases were inhibited by zinc protoporphyrin treatment of the rats or by HO-1 siRNA treatment of the podocytes in culture. The number of apoptotic cells was also significantly increased in the glomeruli of diabetic rats and in high glucose–treated podocytes. Inhibition of HO-1 accentuated the increase in apoptotic cells both in vivo and in vitro. Our findings suggest that HO-1 expression protects against podocyte apoptosis under diabetic conditions.

        도시공원 조성에 따른 미기후환경의 변화 분석

        본 연구에서는 대구광역시 중구의 국채보상운동기념공원과 2.28기념중앙공원을 대상으로 도시공원의 조성 후 열섬 영향 정도와 미기후환경 변화를 정량적으로 분석하여 과거대비 미기후 측면의 효율성 검증과 도시내 공원의 중요성 및 필요성에 대해 살펴보고 이를 바탕으로 미기후개선을 위한 친환경적 도시공원 계획 및 도시계획 방안 제시를 목적으로 한다. 본 연구에서는 대구광역시 중구에 위치하는 2.28기념중앙공원과 국채보상운동기념공원을 대상으로 3차원 미기후분석프로그램인 ENVI-met을 이용하여 미기후환경(온도, 습도, 바람)의 변화를 분석ㆍ고찰하였다. 2.28공원 및 국채보상기념공원 조성으로 인한 미기후환경 변화를 요약하면, 지점별로 차이가 있지만 기존대비 각각 2.27%, 1.50%의 온도저감효과가 있는 것으로 나타났다. 습도는 기존대비 각각 5.52%, 2.83%의 상승효과가 나타났다. 본 연구는 과거의 미기후환경을 예측하여 공원 조성을 통한 미기후환경의 변화를 정량적으로 분석하였다는데 가장 큰 의의가 있다. 향후 도시계획 이전에 미기후의 변화를 모의하여 계획에 반영한다면 보다 쾌적한 도시조성에 효과를 줄 것이라 판단된다. This study examined the effect of Gukchae-bosang Memorial Park(GBM park) and 2.28 Jung-ang Memorial Park(JAM park) located at Jung-gu, Daegu on heat island phenomenon. The changes in micro-climate environment after the development of urban parks were analyzed quantitatively to evaluate the efficiency of the parks in terms of micro-climate control. The importance and necessity of urban parks are reviewed based on the analysis, and the guidelines for eco-friendly park and town planning are suggested. In this study, the microclimate environment factors of the urban parks, such as temperature, humidity, and wind, were analyzed and examined by ENVI-met -- a three dimensional micro-climate analysis program. The micro-climate environmental changes caused by the urban parks are as follows. The temperature decreasing effects by the parks were 2.27%(JAM park) and 1.50%(GBM park), and the humidity increasing effects were 5.52%(JAM park) and 2.83%(GBM park) respectively. The significance of this study is that the changes of micro-climate environment by urban parks were analyzed quantitatively on the assumption of the past micro-climate environment. It is expected that the simulation of micro-climate changes from the results of this study helps planners develop a better plan to create more pleasant urban environment.

      • <i>Prunella vulgaris</i> Suppresses HG-Induced Vascular Inflammation via Nrf2/HO-1/eNOS Activation

        <P>Vascular inflammation is an important factor which can promote diabetic complications. In this study, the inhibitory effects of aqueous extract from <I>Prunella vulgaris</I> (APV) on high glucose (HG)-induced expression of cell adhesion molecules in human umbilical vein endothelial cells (HUVEC) are reported. APV decreased HG-induced expression of intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin. APV also dose-dependently inhibited HG-induced adhesion of HL-60 monocytic cells. APV suppressed p65 NF-κB activation in HG-treated cells. APV significantly inhibited the formation of intracellular reactive oxygen species (ROS). HG-stimulated HUVEC secreted gelatinases, however, APV inhibited it. APV induced Akt phosphorylation as well as activation of heme oxygenase-1 (HO-1), eNOS, and nuclear factor E2-related factor 2 (Nrf2), which may protect vascular inflammation caused by HG. In conclusion, APV exerts anti-inflammatory effect via inhibition of ROS/NF-κB pathway by inducing HO-1 and eNOS expression mediated by Nrf2, thereby suggesting that <I>Prunella vulgaris</I> may be a possible therapeutic approach to the inhibition of diabetic vascular diseases.</P>

        도시열섬현상 완화를 위한 녹지네트워크 및 바람길 구축

        현대의 도시는 살아있는 생명체처럼 매일 변화를 거듭하고 있다. 이전과 달리 집중된 공간에서 짧은 시간동안 일어난 공간의 변화는 도시환경을 악화시켜 환경문제를 유발하였다. 특히 지표면 및 토지이용의 변화 등으로 발생된 도시열섬현상은 도시민의 건강과 경제적인 손실 등 사회문제로 대두되고 있다. 이러한 도시열섬현상을 해결하기 위한 도시계획적 접근방법의 하나가 녹지네트워크의 도입이다. 본 연구에서는 대구광역시 중구와 남구를 사례대상지로 선정하여 도시열섬현상 분석, 토지피복분류, 녹지네트워크 연계가능성 분석, 바람길 분석을 통하여 녹지네트워크 구축을 제안하였다. 연구결과 남쪽의 앞산 및 산지지역을 녹지네트워크의 핵, 두류공원 및 달성공원 등 남구 2개 지역과 중구의 6개 지역의 근린공원을 거점으로 하고 학교부지와 공원시설 등을 포함하는 지역을 거점 가능지역으로 설정하여 대상지역의 광역 및 세부 그린네트워크 구축을 제안하였다. 특히 앞산과 연결되는 캠프워커는 핵과 연결되고 있는 거점지역으로서 향후 미군부대 이전 후에 반드시 보전되어야 할 지역으로서 판단된다. 또한 녹지네트워크를 통해 바람길을 확보하기 위하여 중구의 수성교 인근과 남구의 캠프워커지역을 바람통로확보지구를 설정하는 것이 바람직할 것으로 판단된다. Like other organic beings, modern cities change every day. Unlike in the past, changes of space which have been taking place in concentrated spaces during short periods of time have worsened the environment of cities creating environmental problems. In particular, the urban heat-island phenomenon, which is caused by changes in using the earth`s surface, has emerged as a key social issue along with health problems and economic difficulties. The Green Network has been introduced as an urban planning approach to solve the urban heat-island phenomenon. This study suggests constructing a Green Network through analyzing the urban heat island phenomenon, land cover classification, and linkage possibility with the Green Network and wind corridors by researching Jung-Gu, and Nam-Gu in Daegu Metropolitan City. In conclusion, this study presented the construction of a broad and detailed Green Network of object areas by assigning Apsan to the southern area and mountain area as the core, two areas of Nam-Gu and six areas of Jung-Gu such as Duryu Park and, Dalsung Park as footholds and other school land and park facility areas as footholds-to-be areas. In particular, Camp Walker, which is connected with Apsan, is not only a foothold but connected to the core, is necessary to preserve after being transferred from the U.S. Army. In addition, assigning the Susung Bridge area of Jung-Gu and the Camp Walker area of Nam-Gu as the wind corridor areas is recommended.

      • Vascular Protective Role of Samul-Tang in HUVECs: Involvement of Nrf2/HO-1 and NO

        <P>Samul-Tang (Si-Wu-Tang, SMT), composed of four medicinal herbs, is a well-known herbal formula treating hematological disorder or gynecologic disease. However, vascular protective effects of SMT and its molecular mechanisms on the vascular endothelium, known as the central spot of vascular inflammatory process, are not reported. The aim of this study was to investigate vascular protective effects of SMT water extract in human umbilical vein endothelial cells (HUVECs). Water extract of SMT was prepared and identified by HPLC-PDA analysis. Expression of cell adhesion molecules (CAMs) and heme oxygenase-1 (HO-1) and translocation of nuclear factor-kappa B (NF-<I>κ</I>B) and nuclear factor-erythroid 2-related factor 2 (Nrf2) were determined by western blot. Nuclear localization of NF-<I>κ</I>B and Nrf2 was visualized by immunofluorescence and DNA binding activity of NF-<I>κ</I>B was measured. ROS production, HL-60 monocyte adhesion, and intracellular nitric oxide (NO) were also measured using a fluorescent indicator. SMT suppressed NF-<I>κ</I>B translocation and activation as well as expression of CAMs, monocyte adhesion, and ROS production induced by TNF-<I>α</I> in HUVECs. SMT treated HUVECs showed upregulation of HO-1 and NO which are responsible for vascular protective action. Our study suggests that SMT, a traditionally used herbal formula, protects the vascular endothelium from inflammation and might be used as a promising vascular protective drug.</P>

        8년간(1992~1999) 산업안전보건연구원에 의뢰된 직업병 심의 사례 분석

        Objectives : Pneumoconisis and noise-induced hearing loss (NIHL) have been reported as main occupational diseases by the Special Health Examination. The Industrial Accident Compensation Insurance has reported various work-related diseases, however, these two diseases occupied almost a half of compensated cases. Therefore, it was not well known about the status of occupational diseases other than pneumoconiosis, NIHL, and cardio-cerebrovascular accident (CVA). This study was conducted to analyze claimed cases as an occupational disease, that was requested to the Korea Industrial Safety and Health Agency (KOSHA). Methods : The local office of the Korea Labor Welfare Corporation (KLWC) has asked the KOSHA for confirmation of claimed cases as an occupational disease. We analyzed 379 cases requested from KLWC, the Ministry of Labor, employers, unions and occupational health agencies from 1992 to 1999. Results : Male was 80.7 % of the requested cases. Their mean age was 42 years old and 75.5 % of them were more than 35 years old. The requested cases were increased rapidly from 25 cases in 1992 to 108 cases in 1999 and the accept rate was 50.7 %. The majority of requested cases were respiratory diseases(22.4%), cancers(18.5%), Neuropsychiatric problems(14.5%), and musculoskeletal problems(13.5%). The accept rate was high in reproductive, respiratory, musculoskeletal and digestive disorders and low in neuropsychiatric, renal and otologic problems and occupational cancers. 73.6% of them were caused by chemical agents, especially 28.5 % were by organic solvents. 67% of them were clinically confirmed at university hospitals. A half of the cases were from KyongIn area, even the request came from the whole country. Conclusions : A claim was common in workers whose age was over 35 years old and exposure history was over 10 years. The respiratory diseases and neuropsychiatric disorders were still main problems in occupational health and occupational cancers was increasing even though its accept rate was not high yet.

      • Anti-Proliferative Effect of an Aqueous Extract of <i>Prunella vulgaris</i> in Vascular Smooth Muscle Cells

        <P>The abnormal proliferation of vascular smooth muscle cells (VSMCs) in arterial walls is an important pathogenic factor of vascular disorders such as diabetic atherosclerosis. We have reported the anti-inflammatory effect of an aqueous extract from <I>Prunella vulgaris</I> (APV) in vascular endothelial cell. In the present study, APV exhibited inhibitory effects on high glucose-stimulated VSMC proliferation, migration, and invasion activities, inducing G<SUB>1</SUB> cell cycle arrest with downregulation of cyclins and CDKs and upregulation of the CKIs, p21<SUP>waf1/cip1</SUP> and p27<SUP>kip1</SUP>. Furthermore, APV dose dependently suppressed the high glucose-induced matrix metalloproteinase activity. High glucose-induced phosphorylation of ERK, p38 MAPK, was decreased by the pretreatment of APV. NF-<I><I>κ</I></I>B activation by high glucose was attenuated by APV, as an antioxidant. APV attenuated the high glucose-induced decrease of nuclear factor E2-related factor-2 (Nrf2) translocation and heme oxygenase-1 (HO-1) expression. Intracellular cGMP level was also increased by APV treatment. These results demonstrate that APV may inhibit VSMC proliferation via downregulating ROS/NF-<I><I>κ</I></I>B /ERK/p38 MAPK pathways. In addition, APV has a beneficial effect by the interaction of Nrf2-mediated NO/cGMP with HO-1, suggesting that <I>Prunella vulgaris</I> may be useful in preventing diabetic atherosclerosis.</P>

        불화물의 첨가가 아연화유지놀의 압축강도, 이차 우식에 대한 저항성, 변연누출에 미치는 영향에 관한 실험적 연구

        The purpose of this study was to investigate the effect of addition of fluoride to conventional zinc oxide eugenol on compressive strength, resistance of enamel to secondary caries and marginal leakage. Groups were divided by adding materials and their content. Group 1 was control group that did not contain fluoride, Group 2 was 10% sodium fluoride group, Group 3 was 20% sodium fluoride group, Group 4 was 10% stannous fluoride group and Group 5 was 20% stannous fluoride group. Each of four experimental groups was compared with control group. The results were as follows : 1. In compressive strength after 3 days, 10% stannous fluoride group was highest and then 20% sodium fluoride group, 10% stannous fluoride group, 10% sodium fluoride group, and non-fluoride group were in order. Compressive strength after 1 day water soakong after 3 days was stastically not significant(p>0.05). 2. In resistance of enamel to secondary caries, hardness reduction rate of experimental groups was lower than control group. There was no significant difference among groups (p>0.05). 3. Sodium fluoride group showed less marginal leakage than stannous fluoride group(p<0.05). 4. The amount of fluoride release was largest in 20% sodium fluoride group, and then 10% sodium fluoride group, 20% stannous fluoride, 10% stannous fluoride group were in order.

      • 얼굴인식 시스템을 위한 효과적인 인공해마 알고리즘 구현

        In this paper, we propose the development of Artificial Hippocampus Algorithm(AHA) which remodels a principle of brain of hippocampus. Hippocampus takes charge auto-associative memory and controlling functions of long-term or short-term memory strengthening. We organize auto-associative memory based 4 steps system (EC, DG, CA3, and CA1) and improve speed of learning by addition of modulator to long-term memory learning. In hippocampus system, according to the 3 steps order, information applies statistical deviation on Dentate Gyrus region and is labeled to responsive pattern by adjustment of a good impression. In CA3 region, pattern is reorganized by auto-associative memory. In CA1 region, convergence of connection weight which is used long-term memory is learned fast a by neural network which is applied modulator. To measure performance of Artificial Hippocampus Algorithm, PCA(Principal Component Analysis) and LDA(Linear Discriminants Analysis) are applied to face images which are classified by pose, expression and picture quality. Next, we calculate feature vectors and learn by AHA. Finally, we confirm cognitive rate. The results of experiments, we can compare a proposed method of other methods, and we can confirm that the proposed method is superior to the existing method.
