      • A High Performance Soft-Switching AC/DC Converter

        This paper introduced a high performance single stage soft-switching AC/DC converter. The introduced soft-switching AC/DC converter uses four switches fullbridge circuit construct to achieve the input power factor correction and the output DC voltage regulation. The introduced soft-switching AC/DC converter adopts interleaved operation mode in ac input side to reduce the input current harmonic and increase the supply power level. Moreover, the introduced soft-switching AC/DC converter can get soft-switching function on its main power switches without soft-switching auxiliary circuit. Thus, the cost of circuit in low. A simple phase-shift control strategy is used to synthesize a suitable low harmonics sinusoidal waveform for the ac input current and achieve well dynamic regulation. A design example of 1000W high performance DC power supply system is examined to assess its performance.

        A Novel Esterase from Paenibacillus sp. PBS-2 Is a New Member of the β-Lactamase Belonging to the Family VIII Lipases/Esterases

        Screening of a gene library from Paenibacillus sp. PBS-2 generated in Escherichia coli led to the identification of a clone with lipolytic activity. Sequence analysis showed an open reading frame encoding a polypeptide of 378 amino acid residues with a predicted molecular mass of 42 kDa. The esterase displayed 69% and 42% identity with the putative β-lactamases from Paenibacillus sp. JDR-2 and Clostridium sp. BNL1100, respectively. The esterase contained a Serx- x-Lys motif that is conserved among all β-lactamases found to date. The protein PBS-2 was produced in both soluble and insoluble forms when E. coli cells harboring the gene were cultured at 18°C. The enzyme is a serine protein and was active against p-nitrophenyl esters of C2, C4, C8, and C10. The optimum pH and temperature for enzyme activity were pH 9.0 and 30°C, respectively. Relative activity of 55% remained at up to 5°C with an activation energy of 5.84 kcal/mol, which indicates that the enzyme is cold-adapted. Enzyme activity was inhibited by Cd2+, Cu2+, and Hg2+ ions. As expected for a serine esterase, activity was inhibited by phenylmethylsulfonyl fluoride. The enzyme was remarkably active and stable in the presence of commercial detergents and organic solvents. This cold-adapted esterase has potential as a biocatalyst and detergent additive for use at low temperatures.

        Determination of stay cable force based on effective vibration length accurately estimated from multiple measurements

        Due to its easy operation and wide applicability, the ambient vibration method is commonly adopted to determine the cable force by first identifying the cable frequencies from the vibration signals. With given vibration length and flexural rigidity, an analytical or empirical formula is then used with these cable frequencies to calculate the cable force. It is, however, usually difficult to decide the two required parameters, especially the vibration length due to uncertain boundary constraints. To tackle this problem, a new concept of combining the modal frequencies and mode shape ratios is fully explored in this study for developing an accurate method merely based on ambient vibration measurements. A simply supported beam model with an axial tension is adopted and the effective vibration length of cable is then independently determined based on the mode shape ratios identified from the synchronized measurements. With the effective vibration length obtained and the identified modal frequencies, the cable force and flexural rigidity can then be solved using simple linear regression techniques. The feasibility and accuracy of the proposed method is extensively verified with demonstrative numerical examples and actual applications to different cable-stayed bridges. Furthermore, several important issues in engineering practice such as the number of sensors and selection of modes are also thoroughly investigated.

        Elimination of environmental temperature effect from the variation of stay cable force based on simple temperature measurements

        Under the interference of the temperature effect, the alternation of cable force due to damages of a cable-stayed bridge could be difficult to distinguish. Considering the convenience and applicability in engineering practice, simple air or cable temperature measurements are adopted in the current study for the exclusion of temperature effect from the variation of cable force. Using the data collected from Ai-Lan Bridge located in central Taiwan, this work applies the ensemble empirical mode decomposition to process the time histories of cable force, air temperature, and cable temperature. It is evidently observed that the cable force and both types of temperature can all be categorized as the daily variation, long-term variation, and high-frequency noise in the order of decreasing weight. Moreover, the correlation analysis conducted for the decomposed variations of all these three quantities undoubtedly indicates that the daily and long-term variations with different time shifts have to be distinguished for accurately evaluating the temperature effect on the variation of cable force. Finally, consistent results in reducing the range of cable force variation after the elimination of temperature effect confirm the validity and stability of the developed method.

      • Hybrid Visual Servoing Design for a Continuum Robot under Visibility Constraint and Voice Commands

        This paper presents a method to combine voice control and hybrid visual servoing for a continuum robot. In this design, the continuum robot autonomously tracks the element of interest and maintains its position in the safe region based on image information. A method of hybrid visual servoing is proposed to integrate 2D image-based visual servoing (IBVS) to ensure the elements of interests under visibility constraints and 3D position-based visual servoing (PBVS) to adjust the robot to a desired pose in the constraint region. The robotic camera thus provides stable image while tracking the element of interest. In this design, the user can move the continuum robot to acquire the desired view by voice commands. In voice control, the system can recognize connecting words to control the robot. All software modules are integrated using Robot Operating System (ROS) for controlling the continuum robot. Experimental results obtained from a prototype continuum robot verify the effectiveness of the proposed method.

        Damage detection of a cable-stayed bridge based on the variation of stay cable forces eliminating environmental temperature effects

        This study aims to establish an effective methodology for the detection of instant damages occurred in cable-stayed bridges with the measurements of cable vibration and structural temperatures. A transfer coefficient for the daily temperature variation and another for the long-term temperature variation are firstly determined to eliminate the environmental temperature effects from the cable force variation. Several thresholds corresponding to different levels of exceedance probability are then obtained to decide four upper criteria and four lower criteria for damage detection. With these criteria, the monitoring data for three stay cables of Ai-Lan Bridge are analyzed and compared to verify the proposed damage detection methodology. The simulated results to consider various damage scenarios unambiguously indicate that the damages with cable force changes larger than +-1.5% can be detected within 12 hours. Even though not exhaustively reflecting the environmental temperature effects on the cable force variation, both the effective temperature and the air temperature can be considered as valid indices to eliminate these effects at high and low monitoring costs.

        Chrysophanol-induced Necrotic-like Cell Death through an Impaired Mitochondrial ATP Synthesis in Hep3B Human Liver Cancer Cells

        Liver cancer is the most common form of cancer in Taiwan and it usually responds to chemotherapy. However, patients often have side effects to the chemotherapeutic drugs. Thus new agents are urgently required to treat liver cancer. Chrysophanol, one of the anthraquinone derivatives, was reported to inhibit some human cancer cell growth which may be due to the induction of apoptosis similar to other anthraquinone derivatives though such actions have not been reported. In the present study, we reported that chrysophanol inhibits cell growth in Hep3B liver cancer cells based on the following observations: 1) induc cell morphological changes; 2) decreased percentage of viable cells; 3) induced S phase arrest of cell cycle progression; 4) induced DNA damage as measured by comet assay and DAPI staining. Chrysophanolinduced cell death however, seems to be related to necrotic processes rather than typical apoptosis. Chrysophanol induced reactive oxygen species and Ca2+ production and decreased mitochondrial membrane potential (ΔΨm) and ATP levels in Hep3B cells. No effects were observed on known protein regulators of apoptosis such as Bax and Bcl-2. Chrysophanolinduced cell death took place independently of caspase-8 and -9. Based on our findings, we propose that chrysophanol reduces cellular ATP levels causing a drop in energy resulting in necrotic-like cell death.

        Direct Purification of Burkholderia Pseudomallei Lipase from Fermentation Broth Using Aqueous Two-Phase Systems

        An aqueous two-phase purification process was employed for the recovery of Burkholderia pseudomallei from fermentation broth. The partition behavior of B.pseudmallei lipase was investigated with various parameters such as phase composition, tie-line length (TLL), volume ratio (VR), sample loading, system pH, and addition of neutral salts. Optimum conditions for the purification of lipase were obtained in polyethylene glycol (PEG) 6000-potassium phosphate system using TLL of 42.2% (w/w), with VR of 2.70, and 1% (w/w) NaCl addition at pH 7 for 20% (w/w) crude load. Based on this system, the purification factor of lipase was enhanced to 12.42 fold, with a high yield of 93%.
