        CXCL12/CXCR4 Axis is Involved in the Recruitment of NK Cells by HMGB1 Contributing to Persistent Airway Inflammation and AHR During the Late Stage of RSV Infection

        We previously showed that both high-mobility group box-1 (HMGB1) and natural killer (NK) cells contribute to respiratory syncytial virus (RSV)-induced persistent airway inflammation and airway hyperresponsiveness (AHR). Meanwhile, Chemokine (C-X-C motif) ligand 12 (CXCL12) and its specific receptor (chemokine receptor 4, CXCR4) play important roles in recruitment of immune cells. CXCL12 has been reported to form a complex with HMGB1 that binds to CXCR4 and increases inflammatory cell migration. The relationship between HMGB1, NK cells and chemokines in RSV-infected model remains unclear. An anti-HMGB1 neutralizing antibody and inhibitor of CXCR4 (AMD3100) was administered to observe changes of NK cells and airway disorders in nude mice and BALB/c mice. Results showed that the mRNA expression and protein levels of HMGB1 were elevated in late stage of RSV infection and persistent airway inflammation and AHR were diminished after administration of anti-HMGB1 antibodies, with an associated significant decrease in CXCR4+ NK cells. In addition, CXCL12 and CXCR4 were reduced after HMGB1 blockade. Treatment with AMD3100 significantly suppressed the recruitment of NK cells and alleviated the airway disorders. Thus, CXCL12/CXCR4 axis is involved in the recruitment of NK cells by HMGB1, contributing to persistent airway inflammation and AHR during the late stage of RSV infection.

        The Effect of Transformational Leadership on Job Performance and Employee Happiness: The Mediating Role of Organizational Commitment–Focus on China’s Financial Institutions

        Amid the escalating tensions between the United States and China, alongside growing geopolitical uncertainties, the global economy and financial sectors face increased risks of fragmentation. In response to these challenges, China has focused on enhancing the management quality and capabilities within its financial sector, emphasizing the gradual but steady opening up of its financial industry to global markets. However, there is still a significant gap in the current literature concerning the positive influence of transformational leadership on employee happiness within the financial sector. Addressing this gap, our study introduces and evaluates a pioneering research model aimed at exploring the mediating role of organizational commitment in the relationship between transformational leadership and key outcomes such as job performance and employee happiness. To conduct this comprehensive analysis, data were meticulously collected from 334 employees working across a diverse array of financial institutions in China,including a comparative analysis between banks and other financial institutions using AMOS 24. Utilizing SPSS 26.0 for correlation and regression analysis,our findings substantiate the proposed hypotheses. Key outcomes include: transformational leadership significantly boosts organizational commitment; enhances job performance; elevates employee happiness, notably within the banking sector compared to other financial areas; and organizational commitment serves as a crucial intermediary in the relationship between transformational leadership, job performance, and employee happiness. Furthermore, our research highlights the nuanced differences in the impact of transformational leadership across various types of financial institutions, underscoring the unique challenges and opportunities presented within the banking sector compared to other financial domains. This research underscores the urgent need for adaptive leadership strategies in the financial sector to navigate contemporary challenges, offering fresh insights into fostering a resilient and thriving organizational culture amidst global uncertainties.

        Long non-coding RNA T-cell leukemia/lymphoma 6 serves as a sponge for miR-21 modulating the cell proliferation of retinoblastoma through PTEN

        Retinoblastoma (Rb) is one of the most common eye malignancies occur in childhood. The crucial roles of non-coding RNAs, particularly long non-coding RNAs (lncRNAs) and microRNAs (miRNAs), have been widely reported in Rb progression. In the present study, we found the expression of lncRNA T-cell leukemia/lymphoma 6 (TCL6) was significantly downregulated in Rb tissues and cell lines. Knockdown of lncRNA TCL6 promoted cell proliferation while reduced cell apoptosis in Rb cells. Moreover, lncRNA TCL6 serves as a sponge for miR-21, a previously-reported oncogenic miRNA in Rb, by direct targeting to negatively regulated miR-21 expression, therefore modulating Rb proliferation through miR-21. TCL6 overexpression inhibited Rb cell proliferation while miR-21 overexpression exerted an opposing effect; the effect of TCL6 overexpression was partially attenuated by miR-21 overexpression. PTEN/PI3K/AKT signaling pathway was involved in lncRNA TCL6/miR-21 axis modulating Rb cell proliferation. Taken together, lncRNA TCL6 serves as a tumor suppressor by acting as a sponge for miR-21 to counteract miR-21-mediated PTEN repression.

        Crushing Characteristics on Square Tubes Under Progressive Buckling

        This paper presented the research on crushing characteristics for progressive bucking square tubes under axial loading using the dimensional analysis method. According to the mechanical properties of mild steel and aluminium alloy, the finite element models of progressive buckling square tubes made up of different materials were established. Then dimensional analysis was developed to obtain the influence of inherent parameters on energy absorption characteristics of progressive buckling square tubes, and a theoretical prediction model was proposed to predict the energy absorption characteristics of progressive buckling square tubes subjected to axial loading. The theoretical prediction results were found to match well with finite element analysis and dynamic test results. Furthermore, the equivalence methods of square tubes made up of different materials and geometric parameters were discussed. The results showed that the equivalence of deformation displacement and equivalence of mean crushing force are better equivalence methods for lightweight design, and keeping a constant length and width to obtain an equivalent thickness for progressive buckling square tubes is a more stable equivalence method.

        Serum Anti-Fumarate Hydratase Autoantibody as a Biomarker for Predicting Prognosis of Acute-on-Chronic Liver Failure

        Background/Aims: To investigate the autoantibody against fumarate hydratase (FH), which is a specific liver failure-associated antigen (LFAA) and determine whether it can be used as a biomarker to evaluate the prognosis of acute-on-chronic liver failure (ACLF). Methods: An immunoproteomic approach was applied to screen specific LFAAs related to differential prognosis of ACLF (n=60). Enzyme-linked immunosorbent assay (ELISA) technology was employed for the validation of the frequency and titer of autoantibodies against FH in ACLF patients with different prognoses (n=82). Moreover, we clarified the expression of autoantibodies against FH in patients with chronic hepatitis B (n=60) and hepatitis B virus-related liver cirrhosis (n=60). The dynamic changes in the titers of autoantibodies against FH were analyzed by sample collection at multiple time points during the clinical course of eight ACLF patients with different prognoses. Results: Ultimately, 15 LFAAs were screened and identified by the immunoproteomic approach. Based on ELISA-based verification, anti-FH/Fumarate hydratase protein autoantibody was chosen to verify its expression in ACLF patients. ACLF patients had a much higher anti-FH autoantibody frequency (76.8%) than patients with liver cirrhosis (10%, p=0.000), patients with chronic hepatitis B (6.7%, p=0.022), and normal humans (0%, p=0.000). More importantly, the frequency and titer of anti-FH protein autoantibodies in the serum of ACLF patients with a good prognosis were much higher than that of patients with a poor prognosis (83.9% vs 61.5%, p=0.019; 1.41±0.85 vs 0.94±0.56, p=0.017, respectively). The titer of anti-FH autoantibodies showed dynamic changes in the clinical course of ACLF. Conclusions: The anti-FH autoantibody in serum may be a potential biomarker for predicting the prognosis of ACLF.
