The potential use of sorbents to manage emissions of ultrafine metal nuclei from residual oil combustion was investigated by using an 82-kW-rated laboratory-scale refractory-lined combustor. Without sorbent addition, baseline measurements of the fly ash particle size distribution (PSD) and chemical composition indicate that most of the metals contained in the residual oil form ultrafine particles (~0.1 mm diameter). These results are consistent with particle formation via mechanisms of ash vaporization and subsequent particle nucleation and growth. Equilibrium calculations predict metal vaporization at flame temperatures and were used to define regions above the dew point for the major metal constituents (iron [Fe], nickel [Ni], vanadium [V], and zinc [Zn]) where vapor-phase metal and solidphase sorbents could interact. The addition of dispersed kaolinite powder resulted in an approximate 35% reduction in the ultrafine nuclei as determined by changes to the PSDs as well as the size-dependent chemical compositon.

In-stent stenosis is a feared complication of flow diversion treatment for cerebral aneurysms. We present 2 cases of patients treated with pipeline flow diversion for unruptured cerebral aneurysms. Initial perioperative dual antiplatelet therapy (DAPT) consisted of standard aspirin plus clopidogrel. At 6-month follow-up cerebral angiography, the patients were noted to have developed significant in-stent stenosis (63% and 53%). The patients were treated with cilostazol and clopidogrel for at least 6 months. Subsequent angiography at 1-year post-treatment showed significant improvement of the in-stent stenosis from 63% to 34% and 53% to 21%. The role of cilostazol as treatment of intracranial in-stent stenosis has not been previously described. Cilostazol’s vasodilatory effect and suppression of vascular smooth muscle proliferation provides ideal benefits in this setting. Cilostazol plus clopidogrel may be a safe and effective alternative to standard DAPT for treatment of in-stent stenosis following flow diversion and warrants further consideration and investigation.

When recording single-unit responses from neural systems, a common problem is the accurate detection of spikes (action potentials) in the presence of competing unwanted (noise) signals. While some sources of noise can be readily dealt with through filtering or 'template subtraction' techniques, other sources present a more difficult problem. In particular, noise components introduced by power supplies, which contain harmonics of the power-line frequency, can be particularly troublesome in that they can mimic the shape of the desired spikes. Thus, standard 'template subtraction' techniques or notch-filtering approaches are not appropriate. In this study, we propose the use of a novel template-subtraction scheme that involves estimating the power-line noise waveform and using cross-correlation techniques to subtract them from the recordings. This technique requires two key steps: (1) cross-correlation analysis of each recorded waveform extracts a robust representation of the power-line noise waveform and (2) a second level of cross-correlation to successfully subtract that representation from each recorded waveform. This paper describes this algorithm and provides examples of its implementation using actual recorded waveforms that are contaminated with these noise signals. An improvement (reduction) in the noise level is reported, as are suggestions for future implementation of this strategy.

The inflammatory response requires coordinated activation of both transcription factors and chromatin to induce transcription for defense against pathogens and environmental insults. We sought to elucidate the connections between inflammatory signaling pathways and chromatin through genomic footprinting of kinase activity and unbiased identification of prominent histone phosphorylation events. We identified H3 serine 28 phosphorylation (H3S28ph) as the principal stimulation-dependent histone modification and observed its enrichment at induced genes in mouse macrophages stimulated with bacterial lipopolysaccharide. Using pharmacological and genetic approaches, we identified mitogen-and stress-activated protein kinases (MSKs) as primary mediators of H3S28ph in macrophages. Cell-free transcription assays demonstrated that H3S28ph directly promotes p300/CBP-dependent transcription. Further, MSKs can activate both signal-responsive transcription factors and the chromatin template with additive effects on transcription. Specific inhibition of MSKs in macrophages selectively reduced transcription of stimulation-induced genes. Our results suggest that MSKs incorporate upstream signaling inputs and control multiple downstream regulators of inducible transcription.
