        고지방식이를 섭취시킨 흰쥐의 혈청 지질성분에 도라지가 미치는 영향

        Objects : This study was carried out to investigate the effects of Platycodi Radix powder on improvement of lipid composition in high fat-fed rats. Methods : Male sprague-Dawley rats of 7 weeks old, weighing 318.43±3.09g, were divided into two groups; the control group(C group: high fat(30%, Lard)) and platycodi Radix group(P group: 20% Platycodi Radix powder), they were fed experimental diets for 8 weeks. Results : Food intake and body weight gains had differences between the groups. Serum cholesterol levels were significantly lower in P group than C group. The level of serum HDL- Cholesterol was higher in P group than C group. The level of Serum LDL-C was significantly lower in P group than in C group. The ratio of HDC-Cholesterol to total Cholesterol were higher in P group than in the C group, too. The Value of atherogenic index(AI) was determined to be low in P group.

      • 마늘成分이 Lard와 Alcohol을 攝取한 흰쥐 血淸中의 酵素活性에 미치는 影響

        The present study has been undertaken to investigate the effects of garlic added to food on the activities of several enzymes in serum of rats lard and alcohol. Thirty-five males of Sprague-Dawley stains weighed about 130 g were divided into 7groups, earth group receiving a different diet for 10 weeks ; i.e. basal diet plus 15% lard, basal diet plus 5% alcohol, basal diet plus 0.5 % garlic, basal diet plus 15% lard and 0.5% garlic. Determinations were carried out on the net weight gain, food efficiency ratio, weight of organs, and glutamic oxa1acetic transaminase, glutamic pyruvic transaminase, lactate dehydrogenase, alkaline phogphatase activities in serum of rats. The results obtained were as follows : 1. Rats given feed containing lard and alcohol showed significant decrease in net weight gain, but garlic caused an increase in food efficiency ratio. 2. Lard supplementation caused an increase in the weight of liver, kidney, spleen, but another groups did not. 3. AST, ALT, ALP, LDH of serum were significantly increased in lard and alcohol containing group but garlic feeding decreased enzyme activities compared to lard and alcohol containing group.

        V79 배양 세포에 있어 인삼추출물이 DNA복제 및 회복 합성에 미치는 분자생물학적 연구

        본 연구는 인삼추출물이 DNA 복제 및 회복 합성에 미치는 영향을 조사하기 위해 수행되었다. DNA 복제 및 회복 합성도의 측정은 V79 chinese hamster lung cell의 핵 내 DNA에 [3H]-tymidine이 유입되는 양으로 측정하였다. 그 결과 인삼추출물의 농도(0~1ug/㎖)가 높을수록 점점 더 크게 감소하였다. 이와 같이 인삼추출물에 의하여 DNA 복제 합성도가 감소하는 현상은 농도가 더 높을수록 효과적인 것으로 보아 농도에 의존한다는 것을 알 수 있었다. 한편 DNA 회복 합성도에 인삼추출물이 미치는 영향을 알아보기 위해 우선 DNA를 손상시켰다. DNA를 손상시키는 물질은 여러 가지 있으나, 본 실험에서는 자외선과 methylmethane sulfonate(MMS)를 사용하였다. 이렇게 손상된 DNA가 인삼추출물(0~1ug/㎖)에 의하여 얼마나 회복되었는가를 알아보기 위하여 인삼추출물의 농도를 각각 달리하여 V79 cell에 첨가하였다. 그 결과 인삼추출물의 농도가 높을수록 DNA 회복합성도는 증가하였다. 이와 같이 인삼추출물이 DNA 복제 합성은 억제하고, DNA 회복 합성은 증가시키는 상반된 효과를 보여 주었다. 이러한 상반된 효과는 복제합성과 회복합성의 두 과정에서 서로 다른 효소체계가 관여하기 때문인 것으로 보인다. 인삼추출물이 DNA복제 및 회복합성에 미치는 영향에 대하여 보다 더 구체적인 분자생물학적 메커니즘 연구가 이루어져야 할 것이다.

      • Stearic acid, Oleic acid, Elaidic acid에 의한 간세포의 성장 및 지방산 조성의 변화

        This study was carried out in order to investigate the effect of stearic acid (18:0,SA), oleic (18:1cis.0A) and elaidic acid (18:1 trans, EA) on the cell growth and fatty acid composition of Hep-G2 cells. The cells were incubated in serum-free medium 25, 50, 100 and 200uM of fatty acid combined with albumin for 2 days. The fatty acid concentration up to lOOuM showed the normal growth, but the cell growth decreased in the presence of 200uM fatty acid. The treatment of cells with lOOuM of a fatty acid for two days significantly (p<0.05) increased the cellular triglyceride (TG) content in all fatty acid groups compared to control, but TG content was not significantly different among all treatment group, but total cholesterol (TC) was the highest level in EA group. Cellular phospholipid(PL) contents were similar between the control and all faty acid groups.

The contents ratio of triglyceride, total cholesterol in serum of lard-fed group showed significantly increase compared to controls and but the simultaneous Supple-mentation of garlic and lard decreased significantly than those of lard-fed group. 4. The contents of free-cholesterol and ester cholesterol in serum of the simultaneous supplementation of garlic decreased as compared with the lard single fed group. 5. Garlic single-fed group increased plasma HDL-cholesterol compared to controls but in any other groups were not changed. 6. The ratio of contents of VLDL, LDL-cholesterol and HDL-cholesterol in serum was significantly high in the only lard containing group and alcohol group, the garlic supplemented group was showed lower tendency than only lard and alcohol group. The above results suggest that garlic would prevent the metabolic disease by improv-ing hyperlipemia.

        人蔘이 卵巢摘出로 誘發된 흰쥐의 骨多孔症 예방에 미치는 효능

        Objectives : The present study has been undertaken to investigate the effects of Ginseng Radix on prevention of osteoporosis in ovariectomized rats. Methods : In this experiment, the rats were ovariectomized. Rats were administered by 2 kinds of medicine, Estradiol, Ginseng Radix. The levels of bone mineral density, osteocalcin, ALP, phosphorus, calcium in serum, deoxypyridinoline in urine, calcium, phosphorus, ash weight of bone, body weight and uterus weight were measured. Results : The levels of spinal bone mineral density was significantly increased in comparison with OVX group at 8 weeks in Ginseng Radix group. The levels of serum didn t show significant change in comparison with OVX group in Ginseng Radix group. The levels of urine deoxypyridinoline showed significant decrease in comparison with OVX group in Ginseng Radix group. The levels of femoral and fibula-tibial ash weight didn t show significant increase in comparison with OVX group in Ginseng Radix group. The levels of femoral and fibula-tibial calcium showed significant increase in comparison with OVX group in Ginseng Radix group. The levels of femoral and fibula-tibial phosphorus showed significant increase in comparison with OVX group in Ginseng Radix group. The levels of body weight didn t show significant changes in comparison with OVX group in Ginseng Radix group. The levels of uterus weight didn t show significant changes in comparison with OVX group in Ginseng Radix group. Conclusions : Reviewing these experimetal results, it appears that Ginseng Radix have efficacy on prevention of osteoporosis. And further study should be conducted to illustrate in depth the curing and prevention of osteoprosis.

        韓國産 紅花子와 中國産 紅花子가 鎭痛 , 消炎 및 關節炎에 미치는 效果

        This experimental study was done to compare the efficacy of Korean and Chinese Carthami Semen on analgesic effect and remedy of inflammation and arthritis. The results were as follows; 1. In analgesic action on the writhing syndrome in mice induced by 0.7% acetic acid, Korean and Chinese Carthami Semen extract group showed significant decrease in comparison with control group. And in analgesic action on the writhing syndrome in mice induced by Acetylcholine HC1, Korean and chinese Carthami Semen extract group showed significant decrease in comparison with control group. 2. In Anti-inflammatory effects, Chinese Carthami Semen extract group showed significant decrease at 150 and 180 minutes after inducing paw-edema in comparison with control group. 3. In the method of adjuvant arthritis, orally administered for 21 days, Korean Carthami Semen extract group showed significant inhibition of the hind paw edema after 1 day and 21 days. Whereas Chinese Carthami Semen extract group showed some reduction, but the results didn's show significant decrease. 4. In the level of leucocytes count, Korean and Chinese Carthami Semen extract group showed significant decrease in comparison with control group. And in the level of total protein. CRP and glucose, Korean and Chinese Carthami Semen extract group didn't showed significant change in comparison with control group.

        Antiproliferative Effects of Panu ginseng Glycosides on DNA Synthesis in Cultured Mouse Fibroblasts

        Panax ginseng ginsenosides were examined for their affects on the DNA synthesis. The DNA 1 synthesis was measured by the thymidine incorporation into NIH3T3 cells. The ginsenoside, panaxytriol, $Rh_1$ and $Rh_2$ showed reduced [$^{3}H$]-thymidine incorporation. However, other ginsenosides of $Rh_1$, $Rh_2$ and $Rh_3$ did not inhibit DNA synthesis. Among the various ginsenosides, ginsenoside $Rh_2$ was found to be the most inhibitory on DNA synthesis. We suggest $Rh_2$ as one of the potential choice of antiproliferative drugs.

        A Efficacy of Ginseng radix on the Preservation of Spinal Bone Mineral Density and Bone Inorganic Substance of Oophorectomized Rats

        This study was undertaken to investigate the effects of the Ginseng radix in osteoporosis of the oophorectomized rats. In this experiment, the rats were oophorectomized and administered the water extracts of the Ginseng radix. The spinal bone mineral density, calcium, phosphorus and ash weight of the bones were measured. The spinal bone mineral density was significantly increased in the ovariectomized (OVX)-Ginseng radix group at 8 weeks as compared to the OVX-saline group. Furthermore, the calcium and phosphorus contents of the femoral and fibula-tibia were significantly increased in the OVX-Ginseng radix group as compared to the OVX-saline group. The ash weights of the femoral and fibula-tibial bones were increases in the OVX-Ginseng Radix group, although it was not statistically significant. On reviewing these experiments, it appears that the Ginseng radix possess efficacy for the prevention of osteoporosis. Further study would be of value to confirm the efficacy of the Ginseng radix for the treatment and/or prevention of osteoporosis in humans.
